Ultrasound-induced acoustophoretic motion of microparticles in three dimensions
We derive analytical expressions for the three-dimensional (3D)
acoustophoretic motion of spherical microparticles in rectangular
microchannels. The motion is generated by the acoustic radiation force and the
acoustic streaming-induced drag force. In contrast to the classical theory of
Rayleigh streaming in shallow, infinite, parallel-plate channels, our theory
does include the effect of the microchannel side walls. The resulting
predictions agree well with numerics and experimental measurements of the
acoustophoretic motion of polystyrene spheres with nominal diameters of 0.537
um and 5.33 um. The 3D particle motion was recorded using astigmatism particle
tracking velocimetry under controlled thermal and acoustic conditions in a
long, straight, rectangular microchannel actuated in one of its transverse
standing ultrasound-wave resonance modes with one or two half-wavelengths. The
acoustic energy density is calibrated in situ based on measurements of the
radiation dominated motion of large 5-um-diam particles, allowing for
quantitative comparison between theoretical predictions and measurements of the
streaming induced motion of small 0.5-um-diam particles.Comment: 13 pages, 8 figures, Revtex 4.
Focusing of sub-micrometer particles and bacteria enabled by two-dimensional acoustophoresis.
Handling of sub-micrometer bioparticles such as bacteria are becoming increasingly important in the biomedical field and in environmental and food analysis. As a result, there is an increased need for less labor-intensive and time-consuming handling methods. Here, an acoustophoresis-based microfluidic chip that uses ultrasound to focus sub-micrometer particles and bacteria, is presented. The ability to focus sub-micrometer bioparticles in a standing one-dimensional acoustic wave is generally limited by the acoustic-streaming-induced drag force, which becomes increasingly significant the smaller the particles are. By using two-dimensional acoustic focusing, i.e. focusing of the sub-micrometer particles both horizontally and vertically in the cross section of a microchannel, the acoustic streaming velocity field can be altered to allow focusing. Here, the focusability of E. coli and polystyrene particles as small as 0.5 μm in diameter in microchannels of square or rectangular cross sections, is demonstrated. Numerical analysis was used to determine generic transverse particle trajectories in the channels, which revealed spiral-shaped trajectories of the sub-micrometer particles towards the center of the microchannel; this was also confirmed by experimental observations. The ability to focus and enrich bacteria and other sub-micrometer bioparticles using acoustophoresis opens the research field to new microbiological applications
Demonstration of astrocytes in cultured amniotic fluid cells of three cases with neural-tube defect
We have investigated the origin of rapidly adhering (RA) cells in three cases of neural tube defects (two anencephali, one encephalocele). We were able to demonstrate the presence of glial fibrillary acidic (GFA) protein in variable percentages (4–80%) of RA cells cultured for 4–6 days by use of indirect immunofluorescence with GFA antiserum. Cells cultured from amniotic fluids of normal pregnancies and fetal fibroblasts were completely GFA protein negative. GFA protein is well established as a highly specific marker for astrocytes. Demonstration of astrocytes may prove to be a criterion of high diagnostic value for neural tube defects. The percentage of astrocytes decreased with increasing culture time, while the percentage of fibronectin positive cells increased both in amniotic fluid cell cultures from neural tube defects and normal pregnancies
Engineered nonlinear lattices
We show that with the quasi-phase-matching technique it is possible to fabricate stripes of nonlinearity that trap and guide light like waveguides. We investigate an array of such stripes and find that when the stripes are sufficiently narrow, the beam dynamics is governed by a quadratic nonlinear discrete equation. The proposed structure therefore provides an experimental setting for exploring discrete effects in a controlled manner. In particular, we show propagation of breathers that are eventually trapped by discreteness. When the stripes are wide the beams evolve in a structure we term a quasilattice, which interpolates between a lattice system and a continuous system.Peer ReviewedPostprint (published version
Plantando, colhendo, vendendo, mas não comendo: práticas alimentares e de trabalho associadas à obesidade em agricultores familiares do Bonfim, Petrópolis, RJ.
Objetivo: verificar a prevalência de obesidade entre adultos das 86 famílias agricultoras de um bairro de Petrópolis, RJ, e analisar seus determinantes socioculturais. Métodos: estudo quantitativo e qualitativo sobre nutrição, práticas alimentares e de trabalho realizado em 2008. Dados antropométricos foram coletados por inquérito nutricional domiciliar e o material qualitativo por observação participante e entrevistas. Resultados: a prevalência de obesidade foi baixa (9,3%) entre os homens, mas bastante elevada entre as mulheres (29,9%). A prática agrícola local implica em atividade física leve para mulheres e intensa para homens. Essa diferença não é acompanhada na dieta, semelhante para homens e mulheres, com predomínio de alimentos de alto valor calórico. A produção familiar objetiva essencialmente a venda. A agricultura mercantil e a decorrente especialização dos cultivos favorecem comprar alimentos no mercado em vez de produzir para autoconsumo. Conclusão: os aspectos socioculturais e ocupacionais estudados podem ter contribuído para elevar a prevalência de obesidade nas mulheres e podem ser úteis no estudo de outros grupos com características semelhantes. Esta pesquisa ratifica a importância de estudar a obesidade em nível local, integrando abordagens quantitativas e qualitativas para identificar possíveis limitações e portas de entrada para ações de intervenção localmente relevantes
Early signaling, referral, and treatment of adolescent chronic pain: a study protocol
<p>Abstract</p> <p>Background</p> <p>Chronic pain is prevalent among young people and negatively influences their quality of life. Furthermore, chronic pain in adolescence may persist into adulthood. Therefore, it is important early on to promote the self-management skills of adolescents with chronic pain by improving signaling, referral, and treatment of these youngsters. In this study protocol we describe the designs of two complementary studies: a signaling study and an intervention study.</p> <p>Methods and design</p> <p>The signaling study evaluates the Pain Barometer, a self-assessed signaling instrument for chronic pain in adolescents. To evaluate the feasibility of the Pain Barometer, the experiences of youth-health care nurses will be evaluated in semi-structured interviews. Also, we will explore the frequencies of referral per health-care provider. The intervention study evaluates Move It Now, a guided self-help intervention via the Internet for teenagers with chronic pain. This intervention uses cognitive behavioural techniques, including relaxation exercises and positive thinking. The objective of the intervention is to improve the ability of adolescents to cope with pain. The efficacy of Move It Now will be examined in a randomized controlled trial, in which 60 adolescents will be randomly assigned to an experimental condition or a waiting list control condition.</p> <p>Discussion</p> <p>If the Pain Barometer is proven to be feasible and Move It Now appears to be efficacious, a health care pathway can be created to provide the best tailored treatment promptly to adolescents with chronic pain. Move It Now can be easily implemented throughout the Netherlands, as the intervention is Internet based.</p> <p>Trial registration</p> <p>Dutch Trial Register NTR1926</p
THE FREQUENCIES OF HAPTOGLOBIN TYPES IN FIVE POPULATIONS *
Haptoglobin types have been determined by starch gel electrophoresis of blood from five populations. The gene frequencies obtained for allele Hp 1 were as follows: American whites, 043; American Negroes, 0.59; African Negroes, 0.72; Apaches, 0.59; and Asiatic Indians, 0.18. In tribes of the Ivory Coast and Liberia, there was a suggestion of a cline which parallels that for haemoglobin S. Evidence is presented that the condition of ahaptoglobinemia is under genetic control but not by a gene allelic to the Hp 1 -Hp 2 series. The importance of the ahaptoglobinemic individuals for genetic studies and the possibility of selection in the maintenance of the genetic polymorphism are discussed. Correlation of histopathologic characteristics to protein expression and function in malignant melanoma
BACKGROUND: Metastatic melanoma is still one of the most prevalent skin cancers, which upon progression has neither a prognostic marker nor a specific and lasting treatment. Proteomic analysis is a versatile approach with high throughput data and results that can be used for characterizing tissue samples. However, such analysis is hampered by the complexity of the disease, heterogeneity of patients, tumors, and samples themselves. With the long term aim of quest for better diagnostics biomarkers, as well as predictive and prognostic markers, we focused on relating high resolution proteomics data to careful histopathological evaluation of the tumor samples and patient survival information. PATIENTS AND METHODS: Regional lymph node metastases obtained from ten patients with metastatic melanoma (stage III) were analyzed by histopathology and proteomics using mass spectrometry. Out of the ten patients, six had clinical follow-up data. The protein deep mining mass spectrometry data was related to the histopathology tumor tissue sections adjacent to the area used for deep-mining. Clinical follow-up data provided information on disease progression which could be linked to protein expression aiming to identify tissue-based specific protein markers for metastatic melanoma and prognostic factors for prediction of progression of stage III disease. RESULTS: In this feasibility study, several proteins were identified that positively correlated to tumor tissue content including IF6, ARF4, MUC18, UBC12, CSPG4, PCNA, PMEL and MAGD2. The study also identified MYC, HNF4A and TGFB1 as top upstream regulators correlating to tumor tissue content. Other proteins were inversely correlated to tumor tissue content, the most significant being; TENX, EHD2, ZA2G, AOC3, FETUA and THRB. A number of proteins were significantly related to clinical outcome, among these, HEXB, PKM and GPNMB stood out, as hallmarks of processes involved in progression from stage III to stage IV disease and poor survival. CONCLUSION: In this feasibility study, promising results show the feasibility of relating proteomics to histopathology and clinical outcome, and insight thus can be gained into the molecular processes driving the disease. The combined analysis of histological features including the sample cellular composition with protein expression of each metastasis enabled the identification of novel, differentially expressed proteins. Further studies are necessary to determine whether these putative biomarkers can be utilized in diagnostics and prognostic prediction of metastatic melanoma
