State Complexity of Reversals of Deterministic Finite Automata with Output
We investigate the worst-case state complexity of reversals of deterministic
finite automata with output (DFAOs). In these automata, each state is assigned
some output value, rather than simply being labelled final or non-final. This
directly generalizes the well-studied problem of determining the worst-case
state complexity of reversals of ordinary deterministic finite automata. If a
DFAO has states and possible output values, there is a known upper
bound of for the state complexity of reversal. We show this bound can be
reached with a ternary input alphabet. We conjecture it cannot be reached with
a binary input alphabet except when , and give a lower bound for the
case . We prove that the state complexity of reversal depends
solely on the transition monoid of the DFAO and the mapping that assigns output

Most Complex Non-Returning Regular Languages
A regular language is non-returning if in the minimal deterministic
finite automaton accepting it there are no transitions into the initial state.
Eom, Han and Jir\'askov\'a derived upper bounds on the state complexity of
boolean operations and Kleene star, and proved that these bounds are tight
using two different binary witnesses. They derived upper bounds for
concatenation and reversal using three different ternary witnesses. These five
witnesses use a total of six different transformations. We show that for each
there exists a ternary witness of state complexity that meets the
bound for reversal and that at least three letters are needed to meet this
bound. Moreover, the restrictions of this witness to binary alphabets meet the
bounds for product, star, and boolean operations. We also derive tight upper
bounds on the state complexity of binary operations that take arguments with
different alphabets. We prove that the maximal syntactic semigroup of a
non-returning language has elements and requires at least
generators. We find the maximal state complexities of atoms of
non-returning languages. Finally, we show that there exists a most complex
non-returning language that meets the bounds for all these complexity measures.Comment: 22 pages, 6 figure
Stability, delivery and functions of human sperm RNAs at fertilization
Increasing attention has focused on the significance of RNA in sperm, in light of its contribution to the birth and long-term health of a child, role in sperm function and diagnostic potential. As the composition of sperm RNA is in flux, assigning specific roles to individual RNAs presents a significant challenge. For the first time RNA-seq was used to characterize the population of coding and non-coding transcripts in human sperm. Examining RNA representation as a function of multiple methods of library preparation revealed unique features indicative of very specific and stage-dependent maturation and regulation of sperm RNA, illuminating their various transitional roles. Correlation of sperm transcript abundance with epigenetic marks suggested roles for these elements in the pre- and post-fertilization genome. Several classes of non-coding RNAs including lncRNAs, CARs, pri-miRNAs, novel elements and mRNAs have been identified which, based on factors including relative abundance, integrity in sperm, available knockout data of embryonic effect and presence or absence in the unfertilized human oocyte, are likely to be essential male factors critical to early post-fertilization development. The diverse and unique attributes of sperm transcripts that were revealed provides the first detailed analysis of the biology and anticipated clinical significance of spermatozoal RNAs
Complexity of Left-Ideal, Suffix-Closed and Suffix-Free Regular Languages
A language over an alphabet is suffix-convex if, for any words

Suffix-convex languages include three special cases: left-ideal, suffix-closed,
and suffix-free languages. We examine complexity properties of these three
special classes of suffix-convex regular languages. In particular, we study the
quotient/state complexity of boolean operations, product (concatenation), star,
and reversal on these languages, as well as the size of their syntactic

Sperm centriole assessment identifies male factor infertility in couples with unexplained infertility - a pilot study
Unexplained infertility affects about one-third of infertile couples and is defined as the failure to identify the cause of infertility despite extensive evaluation of the male and female partners. Therefore, there is a need for a multiparametric approach to study sperm function. Recently, we developed a Fluorescence-Based Ratiometric Analysis of Sperm Centrioles (FRAC) assay to determine sperm centriole quality. Here, we perform a pilot study of sperm from 10 fertile men and 10 men in couples with unexplained infertility, using three centriolar biomarkers measured at three sperm locations from two sperm fractions, representing high and low sperm quality. We found that FRAC can identify men from couples with unexplained infertility as the likely source of infertility. Higher quality fractions from 10 fertile individuals were the reference population. All 180 studied FRAC values in the 10 fertile individuals fell within the reference population range. Eleven of the 180 studied FRAC values in the 10 infertile patients were outliers beyond the 95% confidence intervals (P = 0.0008). Three men with unexplained infertility had outlier FRAC values in their higher quality sperm fraction, while four had outlier FRAC values in their lower quality sperm fraction (3/10 and 4/10, P = 0.060 and P = 0.025, respectively), suggesting that these four individuals are infertile due, in part, to centriolar defects. We propose that a larger scale study should be performed to determine the ability of FRAC to identify male factor infertility and its potential contribution to sperm multiparametric analysis

ABSTRACT. Objective. To determine whether cytokine/chemokine profiles from synovial fluid and sera discriminate mild/moderate osteoarthritis (OA) from normal and severe OA cohorts. Methods. Multiplex technology was used to quantify expression levels for 42 cytokines in the synovial fluid of patients diagnosed with severe OA (n = 20) and mild/moderate OA (n = 12), as well as normal controls (n = 34). The same 42 cytokines were examined in serum samples of patients with severe OA (n = 26) and mild/moderate OA (n = 74) and normal individuals (n = 100)
Gait kinematic analysis in patients with a mild form of central cord syndrome
<p>Abstract</p> <p>Background</p> <p>Central cord syndrome (CCS) is considered the most common incomplete spinal cord injury (SCI). Independent ambulation was achieved in 87-97% in young patients with CCS but no gait analysis studies have been reported before in such pathology. The aim of this study was to analyze the gait characteristics of subjects with CCS and to compare the findings with a healthy age, sex and anthropomorphically matched control group (CG), walking both at a self-selected speed and at the same speed.</p> <p>Methods</p> <p>Twelve CCS patients and a CG of twenty subjects were analyzed. Kinematic data were obtained using a three-dimensional motion analysis system with two scanner units. The CG were asked to walk at two different speeds, at a self-selected speed and at a slower one, similar to the mean gait speed previously registered in the CCS patient group. Temporal, spatial variables and kinematic variables (maximum and minimum lower limb joint angles throughout the gait cycle in each plane, along with the gait cycle instants of occurrence and the joint range of motion - ROM) were compared between the two groups walking at similar speeds.</p> <p>Results</p> <p>The kinematic parameters were compared when both groups walked at a similar speed, given that there was a significant difference in the self-selected speeds (p < 0.05). Hip abduction and knee flexion at initial contact, as well as minimal knee flexion at stance, were larger in the CCS group (p < 0.05). However, the range of knee and ankle motion in the sagittal plane was greater in the CG group (p < 0.05). The maximal ankle plantar-flexion values in stance phase and at toe off were larger in the CG (p < 0.05).</p> <p>Conclusions</p> <p>The gait pattern of CCS patients showed a decrease of knee and ankle sagittal ROM during level walking and an increase in hip abduction to increase base of support. The findings of this study help to improve the understanding how CCS affects gait changes in the lower limbs.</p
Implication of sperm RNAs in transgenerational inheritance of the effects of early trauma in mice.
Small non-coding RNAs (sncRNAs) are potential vectors at the interface between genes and environment. We found that traumatic stress in early life altered mouse microRNA (miRNA) expression, and behavioral and metabolic responses in the progeny. Injection of sperm RNAs from traumatized males into fertilized wild-type oocytes reproduced the behavioral and metabolic alterations in the resulting offspring.We thank M. Rassoulzadegan and V. The sperm factor: paternal impact beyond genes
The fact that sperm carry more than the paternal DNA has only been discovered just over a decade ago. With this discovery, the idea that the paternal condition may have direct implications for the fitness of the offspring had to be revisited. While this idea is still highly debated, empirical evidence for paternal effects is accumulating. Male condition not only affects male fertility but also offspring early development and performance later in life. Several factors have been identified as possible carriers of non-genetic information, but we still know little about their origin and function and even less about their causation. I consider four possible non-mutually exclusive adaptive and non-adaptive explanations for the existence of paternal effects in an evolutionary context. In addition, I provide a brief overview of the main non-genetic components found in sperm including DNA methylation, chromatin modifications, RNAs and proteins. I discuss their putative functions and present currently available examples for their role in transferring non-genetic information from the father to the offspring. Finally, I identify some of the most important open questions and present possible future research avenues
