    Breeding for carrot resistance to Alternaria dauci without compromising taste

    Developing carrot varieties highly resistant to Alternaria dauci is a top priority for breeders. Meanwhile, consumers are increasingly demanding as regards final product quality, particularly taste. Bitterness is one of the five common taste sensations, but it is rejected by most carrot consumers. Therefore, providing tools for efficient plant breeding of resistant, low bitter carrots would be helpful. While resistance QTLs (rQTLs) have already been identified for carrot resistance to A. dauci, the genetic control and mapping of the metabolites involved in bitterness perception have not been addressed so far. We identified the metabolites most involved in bitterness by combining chemical and sensory analyses of a set of resistant and susceptible carrot genotypes grown in different environments. We evaluated their genetic control and heritability in a segregating F2:3 population over 2 years of field trials and searched for colocalizations between rQTLs and metabolite QTLs (mQTLs) to evaluate the link between bitterness and resistance traits. Our results suggest that it is possible to increase resistance while favoring low bitter varieties by selecting genomic regions involved in the expression of one or the other trait and counter-selecting others when r- and mQTL colocalization is unfavorable

    Image-Derived Input Function for Human Brain Using High Resolution PET Imaging with [11C](R)-rolipram and [11C]PBR28

    The aim of this study was to test seven previously published image-input methods in state-of-the-art high resolution PET brain images. Images were obtained with a High Resolution Research Tomograph plus a resolution-recovery reconstruction algorithm using two different radioligands with different radiometabolite fractions. Three of the methods required arterial blood samples to scale the image-input, and four were blood-free methods. values was quantified using a scoring system. Using the image input methods that gave the most accurate results with Logan analysis, we also performed kinetic modelling with a two-tissue compartment model.)-rolipram, which has a lower metabolite fraction.     A Temporal -omic Study of Propionibacterium freudenreichii CIRM-BIA1T Adaptation Strategies in Conditions Mimicking Cheese Ripening in the Cold

    Propionibacterium freudenreichii is used as a ripening culture in Swiss cheese manufacture. It grows when cheeses are ripened in a warm room (about 24°C). Cheeses with an acceptable eye formation level are transferred to a cold room (about 4°C), inducing a marked slowdown of propionic fermentation, but P. freudenreichii remains active in the cold. To investigate the P. freudenreichii strategies of adaptation and survival in the cold, we performed the first global gene expression profile for this species. The time-course transcriptomic response of P. freudenreichii CIRM-BIA1T strain was analyzed at five times of incubation, during growth at 30°C then for 9 days at 4°C, under conditions preventing nutrient starvation. Gene expression was also confirmed by RT-qPCR for 28 genes. In addition, proteomic experiments were carried out and the main metabolites were quantified. Microarray analysis revealed that 565 genes (25% of the protein-coding sequences of P. freudenreichii genome) were differentially expressed during transition from 30°C to 4°C (P<0.05 and |fold change|>1). At 4°C, a general slowing down was observed for genes implicated in the cell machinery. On the contrary, P. freudenreichii CIRM-BIA1T strain over-expressed genes involved in lactate, alanine and serine conversion to pyruvate, in gluconeogenesis, and in glycogen synthesis. Interestingly, the expression of different genes involved in the formation of important cheese flavor compounds, remained unchanged at 4°C. This could explain the contribution of P. freudenreichii to cheese ripening even in the cold. In conclusion, P. freudenreichii remains metabolically active at 4°C and induces pathways to maintain its long-term survival

    The controversy of patellar resurfacing in total knee arthroplasty: Ibisne in medio tutissimus?

    Early arthroplasty designs were associated with a high level of anterior knee pain as they failed to cater for the patello-femoral joint. Patellar resurfacing was heralded as the saviour safeguarding patient satisfaction and success but opinion on its necessity has since deeply divided the scientific community and has become synonymous to topics of religion or politics. Opponents of resurfacing contend that the native patella provides better patellar tracking, improved clinical function, and avoids implant-related complications, whilst proponents argue that patients have less pain, are overall more satisfied, and avert the need for secondary resurfacing. The question remains whether complications associated with patellar resurfacing including those arising from future component revision outweigh the somewhat increased incidence of anterior knee pain recorded in unresurfaced patients. The current scientific literature, which is often affected by methodological limitations and observer bias, remains confusing as it provides evidence in support of both sides of the argument, whilst blinded satisfaction studies comparing resurfaced and non-resurfaced knees generally reveal equivalent results. Even national arthroplasty register data show wide variations in the proportion of patellar resurfacing between countries that cannot be explained by cultural differences alone. Advocates who always resurface or never resurface indiscriminately expose the patella to a random choice. Selective resurfacing offers a compromise by providing a decision algorithm based on a propensity for improved clinical success, whilst avoiding potential complications associated with unnecessary resurfacing. Evidence regarding the validity of selection criteria, however, is missing, and the decision when to resurface is often based on intuitive reasoning. Our lack of understanding why, irrespective of pre-operative symptoms and patellar resurfacing, some patients may suffer pain following TKA and others may not have so far stifled our efforts to make the strategy of selective resurfacing succeed. We should hence devote our efforts in defining predictive criteria and indicators that will enable us to reliably identify those individuals who might benefit from a resurfacing procedure.     The subcritical transition to turbulence of Faraday waves in miscible fluids

    International audienceWe study the development and the breaking process of standing waves at the interface between two miscible fluids of small density contrast. In our experiment, a subharmonic wave is generated by a time-periodic vertical acceleration via the Faraday instability. It is shown that its wavelength may be selected not only by the linear process predicted by the Floquet theory and favouring the most unstable modes allowed by the tank geometry, but also by a nonlinear mode competition mechanism giving the preference to subcritical modes. Subsequently, as the standing wave amplitude grows, a secondary destabilization process occurs at smaller scales and produces turbulent mixing at the nodes. We explain this phenomenon as a subcritical parametric resonance instability. Different approaches derived from local and global stability analysis are proposed to predict the critical wave steepness. These theories are then assessed against various numerical and experimental data varying the frequencies and amplitudes of the forcing acceleration
