    Dynamics of the frustrated Ising lattice gas

    The dynamical properties of a three dimensional model glass, the frustrated Ising lattice gas (FILG) are studied by Monte Carlo simulations. We present results of compression experiments, where the chemical potential is either slowly or abruptly changed, as well as simulations at constant density. One time quantities like density and two time ones like correlations, responses and mean square displacements are measured, and the departure from equilibrium clearly characterized. The aging scenario, particularly in the case of density autocorrelations is reminiscent of spin glass phenomenology with violations of the Fluctuation-dissipation theorem, typical of systems with one replica symmetry breaking. The FILG, as a valid on-lattice model of structural glasses can be described with tools developed in spin glass theory and, being a finite dimensional model, can open the way for a systematic study of activated processes in glasses.Comment: to appear in Phys.     Septation of Infectious Hyphae Is Critical for Appressoria Formation and Virulence in the Smut Fungus Ustilago Maydis

    Differentiation of hyphae into specialized infection structures, known as appressoria, is a common feature of plant pathogenic fungi that penetrate the plant cuticle. Appressorium formation in U. maydis is triggered by environmental signals but the molecular mechanism of this hyphal differentiation is largely unknown. Infectious hyphae grow on the leaf surface by inserting regularly spaced retraction septa at the distal end of the tip cell leaving empty sections of collapsed hyphae behind. Here we show that formation of retraction septa is critical for appressorium formation and virulence in U. maydis. We demonstrate that the diaphanous-related formin Drf1 is necessary for actomyosin ring formation during septation of infectious hyphae. Drf1 acts as an effector of a Cdc42 GTPase signaling module, which also consists of the Cdc42-specific guanine nucleotide exchange factor Don1 and the Ste20-like kinase Don3. Deletion of drf1, don1 or don3 abolished formation of retraction septa resulting in reduced virulence. Appressorium formation in these mutants was not completely blocked but infection structures were found only at the tip of short filaments indicating that retraction septa are necessary for appressorium formation in extended infectious hyphae.     Over the past decade, f(R) theories have been extensively studied as one of the simplest modifications to General Relativity. In this article we review various applications of f(R) theories to cosmology and gravity - such as inflation, dark energy, local gravity constraints, cosmological perturbations, and spherically symmetric solutions in weak and strong gravitational backgrounds. We present a number of ways to distinguish those theories from General Relativity observationally and experimentally.     Response of the Mayfly (Cloeon Dipterum) to chronic exposure to Thiamethoxam in outdoor Mesocosms

    Thiamethoxam (TMX) is a widely used neonicotinoid insecticide that has been detected in surface water monitoring programs in North America and Europe. This has led to questions about its toxicity to non-target insects, specifically those with an aquatic life stage. In order to address the uncertainty associated with possible impacts related to environmental exposures, a chronic (35-d) outdoor mesocosm study with a formulated product containing TMX was conducted. The specific focus of the study was the response of mayflies (Ephemeroptera), which have been reported to be particularly sensitive in laboratory studies. A range of concentrations (nominally 0.1, 0.3, 1.0, 3.0, 10.0 µg/L TMX), plus an untreated control were tested and the abundance and emergence of mayflies (Cloeon dipterum) was assessed on a weekly basis for 35 days. Mean measured time-weighted average exposures were within 6% of nominal over the duration of the study with the mean half-life of TMX in each treatment ranging from 7 to 13 days. Statistically significant reductions in both larval abundance and adult emergence were observed at 10.0, 3.0, and 1.0 ug/L following 1, 2, and 3 weeks of exposure, respectively. Exposure to 0.1 and 0.3 µg/L TMX had no statistically significant effect on larval mayfly abundance or adult emergence at any point in the study. These findings support a 35-d NOEC of 0 .3 µg TMX/L f or m ayflies (C. dipterum) under chronic conditions. Further, since the 95th centile of environmental concentrations has been reported at 0.054 µg/L, these results indicate that populations of C. dipterum and similarly sensitive aquatic insects are unlikely to be significantly impacted by TMX exposure in natural systems

    Novel Poxin Stable cGAMP-Derivatives Are Remarkable STING Agonists

    2′,3′-cGAMP is a cyclic A- and G-containing dinucleotide second messenger, which is formed upon cellular recognition of foreign cytosolic DNA as part of the innate immune response. The molecule binds to the adaptor protein STING, which induces an immune response characterized by the production of type I interferons and cytokines. The development of STING-binding molecules with both agonistic as well as antagonistic properties is currently of tremendous interest to induce or enhance antitumor or antiviral immunity on the one hand, or to treat autoimmune diseases on the other hand. To escape the host innate immune recognition, some viruses encode poxin endonucleases that cleave 2′,3′-cGAMP. Here we report that dideoxy-2′,3′-cGAMP (1) and analogs thereof, which lack the secondary ribose-OH groups, form a group of poxin-stable STING agonists.
