    Histone Acetyltransferases as Regulators of Nonhistone Proteins: The Role of Interferon Regulatory Factor Acetylation on Gene Transcription

    When studying transcription factors, it is necessary to investigate posttranslational modifications. Histone acetyltransferases (HATs) are typical of the modification enzymes involved in chromatin regulation. HATs acetylate the transcription factors (nonhistone proteins) as well as histones. Interferon regulatory factors (IRFs) are transcription factors that bind to the interferon regulatory element (IRF-E) and are involved in regulating cell growth, differentiation, and the immune and hematopoietic systems. During the process of binding to a specific DNA element, IRFs also bind to coactivators such as HATs and become modified. This review looks at how IRFs associate with HATs, p300, and PCAF, and thereby contribute to transcriptional activation

    AbstractDendritic cells (DCs) play a major role in in vivo pathogenesis of HIV-1 infection. Therefore, DCs may provide a promising strategy to control and eventually overcome the fatal infection. Especially, immature DCs express all CD1s, the non-MHC lipid antigen -presenting molecules, and HIV-1 Nef down-regulates CD1 expression besides MHC. Moreover, CD1d-restricted CD4+ NKT cells are infected by HIV-1, reducing the number of these cells in HIV-1-infected individuals. To understand the exact role of DCs and CD1-mediated immune response during HIV-1 infection, Nef down-regulation of CD1a-restricted lipid/glycolipid Ag presentation in iDCs was analyzed. We demonstrated the involvement of the association of Nef with hemopoietic cell kinase (Hck) and p21-activated kinase 2 (PAK2), and that Hck, which is expressed strongly in iDCs, augmented this mutual interaction. Hck might be another therapeutic target to preserve the function of HIV-1 infected DCs, which are potential reservoirs of HIV-1 even after antiretroviral therapy

    Training Attention to the Other’s Need Improves Helping Behaviors in Children with Autism Spectrum Disorder: A Behavioral Analysis Using a Two-dimensional Motion Capture System

    While children with autism spectrum disorder (ASD) can acquire helping behaviors through appropriate interventions, changes in behaviors prior to helping (pre-helping behaviors) remain unclear. In the present study, we examined the effects of social skills training (SST) on helping and pre-helping behaviors in two children with ASD by using a two-dimensional motion capture system. During the SST, the children learned one helping behavior that they lent their items to their partners, and we measured their head movements before initiating the helping behavior (i.e., pre-helping behavior). As a result of SST, the participants became able to help others in response to less-explicit social stimuli after the intervention. Regarding pre-helping behaviors, the children with ASD before the intervention looked straight at the helpee (i.e., recipient of the help) more often than did typically developing peers, and such a behavior was shown to increase after SST. These results indicate that although spontaneous attention to social stimuli may be reduced in children with ASD, success in attending to a helpee could lead to the emergence of helping behaviors. Moreover, the changes in pre-helping behavior indicate an increase in children’s attention to the helpee after the intervention, which may have enhanced their sensitivity to persons in need

    Detection of deviance in Japanese kanji compound words

    Reading fluency is based on the automatic visual recognition of words. As a manifestation of the automatic processing of words, an automatic deviance detection of visual word stimuli can be observed in the early stages of visual recognition. To clarify whether this phenomenon occurs with Japanese kanji compounds-since their lexicality is related to semantic association-we investigated the brain response by utilizing three types of deviants: differences in font type, lexically correct or incorrect Japanese kanji compound words and pseudo-kanji characters modified from correct and incorrect compounds. We employed magnetoencephalography (MEG) to evaluate the spatiotemporal profiles of the related brain regions. The study included 22 adult native Japanese speakers (16 females). The abovementioned three kinds of stimuli containing 20% deviants were presented during the MEG measurement. Activity in the occipital pole region of the brain was observed upon the detection of font-type deviance within 250 ms of stimulus onset. Although no significant activity upon detecting lexically correct/incorrect kanji compounds or pseudo-kanji character deviations was observed, the activity in the posterior transverse region of the collateral sulcus (pCoS)-which is a fusiform neighboring area-was larger when detecting lexically correct kanji compounds than when detecting pseudo-kanji characters. Taken together, these results support the notion that the automatic detection of deviance in kanji compounds may be limited to a low-level feature, such as the stimulus stroke thickness.Peer reviewe

    Fructo-oligosaccharides ameliorate steatohepatitis, visceral adiposity, and associated chronic inflammation via increased production of short-chain fatty acids in a mouse model of non-alcoholic steatohepatitis

    Background: Non-alcoholic fatty liver disease (NAFLD) is a hepatic manifestation of metabolic syndrome. Within the spectrum of NAFLD, non-alcoholic steatohepatitis (NASH) in combination with hepatic inflammation and fibrosis can lead to liver cirrhosis and hepatocellular carcinoma. Dysbiosis was reported to contribute to NASH pathogenesis. This study aimed to determine the effects of fructo-oligosaccharides (FOS) on steatohepatitis and visceral adiposity in an obese mouse model of NASH. Methods: Twelve newborn C57BL/6 J male mice were subcutaneously injected with monosodium glutamate (MSG) to induce obesity on a conventional diet. Six mice were also administered 5% FOS via drinking water from 10 weeks of age. At 18 weeks, histological characteristics of the liver and epididymal fat were compared between the groups. Hepatic mRNA expression of lipid metabolism enzymes and SCFA in feces and sera were measured. Results: Hepatic steatosis, inflammatory cell infiltration, and hepatocyte ballooning in the liver and increased hepatic mRNA expression of fatty acid synthase and glycerol-3-phosphate acyltransferase were observed in the MSG-treated mice. FOS treatment improved the liver pathology and blunted the increases in the mRNA expression levels of lipid metabolism enzymes. In addition, FOS inhibited adipocyte enlargement and formation of crown-like structures and reduced the M1 macrophage frequency in the epididymal fat of the MSG mice (39.4% ± 3.0% vs. 22.8% ± 0.7%; P = 0.001). FOS increased not only the fecal concentrations of n-butyric acid (0.04 ± 0.01 vs. 0.38 ± 0.14 mg/g, P = 0.02), propionic acid (0.09 ± 0.03 vs. 0.42 ± 0.16 mg/g, P = 0.02), and acetic acid (0.65 ± 0.16 vs. 1.48 ± 0.29 mg/g, P = 0.03) but also the serum concentration of propionic acid (3.9 ± 0.5 vs. 8.2 ± 0.5 μmol/L, P = 0.001). Conclusions: FOS ameliorates steatohepatitis, visceral adiposity, and chronic inflammation by increasing SCFA production

    Effect of soy protein on the muscle in human

    Background : In recent years, the number of bedridden people is rapidly increasing due to aging or lack of exercise in Japan. This problem is becoming more serious, since there is no countermeasure against it. In the present study, we designed to investigate whether dietary proteins, especially soy, had beneficial effects on skeletal muscle in 59 volunteers with various physical activities. Methods : We subjected 59 volunteers with various physical activities to meal intervention examination. Persons with low and high physical activities were divided into two dietary groups, the casein diet group and the soy diet group. They ate daily meals supplemented with 7.8 g of powdered casein or soy protein isolate every day for 30 days. Bedridden patients in hospitals were further divided into three dietary groups : the no supplementation diet group, the casein diet group and the soy diet group. They were also subjected to a blood test, a urinalysis, magnetic resonance imaging analysis and muscle strength test of the knee before and after the meal intervention study. Results : Thirty-day soy protein supplementation significantly increased skeletal muscle volume in participants with low physical activity, compared with 30-day casein protein supplementation. Both casein and soy protein supplementation increased the volume of quadriceps femoris muscle in bedridden patients. Consistently, soy protein significantly increased their extension power of the knee, compared with casein protein. Although casein protein increased skeletal muscle volume more than soy protein in bedridden patients, their muscle strength changes by soy protein supplementation were bigger than those by casein protein supplementation. Conclusions : The supplementation of soy protein would be one of the effective foods which prevent the skeletal muscle atrophy caused by immobilization or unloading

    平成16年3月に看護学校養成所指定規則が改正し、2年課程(通信制)の教育課程が提示され、翌17年に本学看護学科に通信制課程が開設された。平成19年には全国で20校となったが、本学はその中で唯一短期大学としての教育機関である。そこで、第1期生の卒業時の質問紙調査の結果を踏まえ、あらためて短期大学における2年課程(通信制)の教育の意義と課題について明らかにした。調査結果から、第1期生が2年間の通信教育で、看護学を新しく学び、看護の専門性の理解とともに看護観を大きく変化させ、自信や誇りを持って看護実践に臨んでいることが明らかになった。また、卒業後の進路に活路を見いだし、学びの継続を考えている。この制度の目的である「質の高い看護を提供できる看護実践者として、施設や地域で活躍できる人材の育成」や、短期大学として社会人を対象とする教育の使命としてのリカレント教育や生涯教育としての役割を果たすことに本学の意義があると考える。In March 2004, with the revision of the specific rules and regulation concerning nursing training school, a two-year correspondence course was set up, and in the following year a correspondence course was established in our school\u27s nursing curriculum. As of the year 2007, throughout the country there are twenty schools that have established this course and, among them, our school is the only junior college. On the basis of studying questionnaires filled out at the time of the graduation by those students who took the course for the first time, what we have clarified here are the educational significance and agenda in a two-year nursing correspondence course. It has become clear that, through the two-year correspondence education, these students have studied nursing anew and, with an understanding of the professionalism involved in nursing and great transformation in their sense of nursing, are endeavoring to practice nursing with confidence and pride. They also seek a path for their life in making progress after graduation and are thinking about continuing their education. We believe the purpose of our school to be "the training of human beings who can provide high-quality nursing as nursing practitioners and can flourish institutionally and regionally" and fulfilling the function of recurrent and lifetime education as part of a junior-college mission to target members of our society
