Survey of aflatoxins in chillies from Pakistan produced in rural, semi-rural and urban environments
Chilli peppers from Pakistan are consumed locally and also exported. Their quality is compromised by aflatoxins (AF) contamination. AF in chillies from rural, semi-rural and urban areas of the Punjab region of Pakistan were determined. Twenty-three (52.3%), 22 (50%) and 29 (65.9%) samples from rural, semi-rural and urban areas respectively, contained levels of aflatoxins which exceeded the European Union limits of >5 µg kg-1 for AFB1 and >10 µg kg-1 for total AF that apply to spices. Mean values for AFB1 in ground samples were 23.8, 14.8 and 14.0 µg kg-1 for rural, semi-rural and urban areas, respectively. Mean total AF in ground samples were 27.7, 17.7 and 16.2 µg kg-1 from equivalent locations. Eleven (50%), 12 (54.5%) and 14 (63.6%) whole samples from rural, semi-rural and urban areas, respectively, contained total levels of AF that exceeded European Union limits. The data indicate that individual localities have particular problems. A limited survey of aflatoxins and zearalenone in feed and feed ingredients from Pakistan
This work presents current information on the presence of aflatoxins (AFs) and zearalenone (ZEN) in feed and feed ingredients from Punjab, Pakistan. The 105 samples tested were concentrated feed, i.e., cotton seed meal (18 samples) and soybean meal (14), and feed ingredients, i.e., crushed corn (17), crushed wheat (15), barley (17). and poultry feed (24). Samples were analyzed using high-performance liquid chromatography equipped with a fluorescence detector. Analysis revealed that 69 of 105 samples were contaminated with AFs, and the highest mean concentrations of AFB1 (6.20 μg/kg) and total AFs (9.30 μg/kg) were found in poultry feed samples. The mean total AF concentrations ranged from the limit of quantification to 165.5 μg/kg. However, 75 of the 105 samples were positive for ZEN. The highest mean concentration (19.45 μg/kg) was found in poultry feed samples. The mean ZEN concentrations were 0.15 to 145.30 μg/kg. The prevalence of AFs and ZEN was high in feed and feed ingredients and needs urgent attention
Aflatoxin B1 in chilies from the Punjab region, Pakistan
The occurrence of aflatoxin B1 (AFB1) in chilies
from Pakistan was determined by using HPLC in work
undertaken in Pakistan.Whole (n=22) and powdered (n=22)
chilies were analyzed. Sixteen (73.0%) and 19 (86.4%)
samples of whole and ground chilies, respectively, were
contaminated. The mean concentration in powdered chilies
(32.20 μg/kg) was higher statistically than in whole chilies
(24.69 μg/kg). Concentrations ranged from 0.00 to 89.56 μg/
kg for powdered chilies, compared with 0.00–96.3 μg/kg for
whole chilies. The limits of detection and quantification were
0.05 μg/kg and 0.53 μg/kg, respectively. The concentrations
were high in general and greater than the statutory limit set
by the European Union. There is considerable scope for

Chronic inflammatory arthritis drives systemic changes in circadian energy metabolism
SignificanceRheumatoid arthritis (RA) is a debilitating chronic inflammatory disease in which symptoms exhibit a strong time-of-day rhythmicity. RA is commonly associated with metabolic disturbance and increased incidence of diabetes and cardiovascular disease, yet the mechanisms underlying this metabolic dysregulation remain unclear. Here, we demonstrate that rhythmic inflammation drives reorganization of metabolic programs in distal liver and muscle tissues. Chronic inflammation leads to mitochondrial dysfunction and dysregulation of fatty acid metabolism, including accumulation of inflammation-associated ceramide species in a time-of-day-dependent manner. These findings reveal multiple points for therapeutic intervention centered on the circadian clock, metabolic dysregulation, and inflammatory signaling
The role of interleukin-1β as a predictive biomarker and potential therapeutic target during clinical ex vivo lung perfusion
BACKGROUND: Extended criteria donor lungs deemed unsuitable for immediate transplantation can be reconditioned using ex vivo lung perfusion (EVLP). Objective identification of which donor lungs can be successfully reconditioned and will function well post-operatively has not been established. This study assessed the predictive value of markers of inflammation and tissue injury in donor lungs undergoing EVLP as part of the DEVELOP-UK study. METHODS: Longitudinal samples of perfusate, bronchoalveolar lavage, and tissue from 42 human donor lungs undergoing clinical EVLP assessments were analyzed for markers of inflammation and tissue injury. Levels were compared according to EVLP success and post-transplant outcomes. Neutrophil adhesion to human pulmonary microvascular endothelial cells (HPMECs) conditioned with perfusates from EVLP assessments was investigated on a microfluidic platform. RESULTS: The most effective markers to differentiate between in-hospital survival and non-survival post-transplant were perfusate interleukin (IL)-1β (area under the curve = 1.00, p = 0.002) and tumor necrosis factor-α (area under the curve = 0.95, p = 0.006) after 30 minutes of EVLP. IL-1β levels in perfusate correlated with upregulation of intracellular adhesion molecule-1 in donor lung vasculature (R(2) = 0.68, p < 0.001) and to a lesser degree upregulation of intracellular adhesion molecule-1 (R(2) = 0.30, p = 0.001) and E-selectin (R(2) = 0.29, p = 0.001) in conditioned HPMECs and neutrophil adhesion to conditioned HPMECs (R(2) = 0.33, p < 0.001). Neutralization of IL-1β in perfusate effectively inhibited neutrophil adhesion to conditioned HPMECs (91% reduction, p = 0.002). CONCLUSIONS: Donor lungs develop a detectable and discriminatory pro-inflammatory signature in perfusate during EVLP. Blocking the IL-1β pathway during EVLP may reduce endothelial activation and subsequent neutrophil adhesion on reperfusion; this requires further investigation in vivo

Carbon storage in Ghanaian cocoa ecosystems
Background: The recent inclusion of the cocoa sector as an option for carbon storage necessitates the need to
quantify the C stocks in cocoa systems of Ghana.
Results: Using farmers’ fields, the carbon (C) stocks in shaded and unshaded cocoa systems selected from the Eastern (ER) and Western (WR) regions of Ghana were measured. Total ecosystem C (biomass C + soil C to 60 cm depth) ranged from 81.8 to 153.9 Mg C/ha. The bulk (~89 %) of the systems’ C stock was stored in the soils. The total C stocks were higher in the WR (137.8 ± 8.6 Mg C/ha) than ER (95.7 ± 8.6 Mg C/ha).
Conclusion: Based on the cocoa cultivation area of 1.45 million hectares, the cocoa sector in Ghana potentially
could store 118.6–223.2 Gg C in cocoa systems with cocoa systems aged within 30 years regardless of shade management. Thus, the decision to include the cocoa sector in the national carbon accounting emissions budget of Ghana is warranted
Combinations of Metarhizium anisopliae with chemical insecticides and their effectiveness in Mahanarva fimbriolata (Hemiptera: Cercopidae) control on sugarcane
Some insecticides can be used jointly with entomopathogenic fungi, and therefore the combi-
nation of chemical and biological control measures can be a safe and effective method to con-
trol insect pests. The aim of this study was to evaluate the costs and efficacy of combinations of
Metarhizium anisopliae (Metschnikoff) Sorokin (Hypocreales: Clavicipitaceae) with thiameth-
oxam and imidacloprid on spittlebug (Mahanarva fimbriolata (Stål); Hemiptera: Cercopidae)
control on sugarcane. The experiment was conducted as a randomized block design (RBD)
with 10 treatments and 4 replications. The treatments included a control (untreated), thia-

methoxam (250 g ha ), imidacloprid (700 g ha ), M. anisopliae (M. a.) (3 × 10 conidia ha ),
A1 (3 × 10 12 M. a. conidia ha −1 + 65 g ha −1 of thiamethoxam), A2 (3 × 10 12 M. a. conidia ha −1 + 125
g ha −1 of thiamethoxam), A3 (3 × 10 12 M. a. conidia ha −1 + 187.5 g ha −1 of thiamethoxam), A4 (3 ×
10 12 M. a. conidia ha −1 + 175 g ha −1 of imidacloprid), A5 (3 × 10 12 M. a. conidia ha −1 + 350 g ha −1 of
imidacloprid), and A6 (3 × 10 12 M. a. conidia ha −1 + 525g ha −1 of imidacloprid). The reductions
in the numbers of M. fimbriolata nymphs per treatment compared to the control were similar
at 15 DAT (days after treatment) in all treatments except combination A5 (M. anisopliae
and thiamethoxam). At 30 DAT, the numbers of nymphs were significantly reduced in all
treatments except A3, and their effectiveness ranged from 14.28% to 92.85%. At 45 DAT the
numbers of M. fimbriolata nymphs per treatment were significantly reduced in the following
treatments: imidacloprid alone at 700g ha -1 , A1, A2, A3, A4 and A6; and the combinations A1
and A2 caused the lowest M. fimbriolata nymph infestations and effectiveness rates of 77.41
and 87.09 %, respectively. At 75 DAT the 2 best control efficacies occurred in treatments A1 (3
× 10 12 M. a. conidia ha -1 of + 65g ha -1 of thiamethoxam) (82.1%) and A5 (78.6%) (3 × 10 12 M. a.
conidia ha −1 + 350 g ha −1 of imidacloprid). At 90 DAT the number of nymphs in the control had
increased 2.8 fold over the number at 75 DAT. Very good control efficacies at 90 DAT occurred
in all treatments with the combination of the fungus with an insecticide. At 105 DAT the
numbers of nymphs had surged in all treatments, and no treatment provided effective control.
The treatments with the highest earnings per hectare were A1 (3 × 10 12 M. a. conidia ha -1 + 65
g thiamethoxam) and M. anisopliae alone at the recommended dose of 3 × 10 12 M. a. conidia
ha -1 . Our findings demonstrate the effectiveness of using either thiamethoxam or imidacloprid
in combination with M. anisopliae to control M. fimbriolata nymphs on sugarcane, but greater
net earnings per hectare occurred with the lowest rate of the thiamethoxam combination than
with any of the imidacloprid combinations.Algunos insecticidas se puede utilizar con hongos entomopatógenos y por lo tanto, la aso-

Detection of early age-related macular degeneration using novel functional parameters of the focal cone electroretinogram
The focal cone electroretinogram is a sensitive marker for macular disease, but have we unlocked its full potential? Typically assessment of waveform parameters is subjective and focuses on a small number of locations (e.g. the a-wave). This study evaluated the discriminatory and diagnostic potential of 4 conventional and 15 novel, objectively determined, parameters in patients with early Age-related Macular Degeneration. Focal cone electroretinograms were recorded in 54 participants with early Age-related Macular Degeneration (72.9±8.2 years) and 54 healthy controls (69±7.7 years). Conventional a and b wave amplitudes and implicit times were measured and compared to novel parameters derived from both the 1st and 2nd derivatives and the frequency-domain power spectrum of the electroretinogram.Statistically significant differences between groups were shown for all conventional parameters, the majority of 1st and 2nd derivative parameters and the power spectrum at 25 and 30 Hz. Receiver operating characteristics showed that both conventional and 1st and 2nd derivative implicit times had provided the best diagnostic potential. A regression model showed a small improvement over any individual parameter investigated. The non-conventional parameters enhanced the objective evaluation of the focal electroretinogram, especially when the amplitude was low. Furthermore, the novel parameters described here allow the implicit time of the electroretinogram to be probed at points other than the peaks of the a and b waves. Consequently these novel analysis techniques could prove valuable in future electrophysiological investigation, detection and monitoring of Age-related Macular Degeneration
Growing networks with local rules: preferential attachment, clustering hierarchy and degree correlations
The linear preferential attachment hypothesis has been shown to be quite
successful to explain the existence of networks with power-law degree
distributions. It is then quite important to determine if this mechanism is the
consequence of a general principle based on local rules. In this work it is
claimed that an effective linear preferential attachment is the natural outcome
of growing network models based on local rules. It is also shown that the local
models offer an explanation to other properties like the clustering hierarchy
and degree correlations recently observed in complex networks. These
conclusions are based on both analytical and numerical results of different
