    On Farm Training and Demonstration of Covered Smut (Sphacelotheca Sorghi Clint) Management Technologies on Sorghum

    Sorghum suffers from a lot of pests among which covered smut is the most important. Covered smut induced by the fungus Sporisorium sorghi and it is extremely seed-borne and seedling infection occurs at the time of germination and emergence of seedlings. The incidence of covered kernel smut varies from place to place, but in Ethiopia, it was estimated to be about 50%. This activity initiated to provide practical training on sorghum covered identification, monitoring, and technical skills to experts and farmers and to demonstrate the effect of seed dressing fungicides and traditional packages against sorghum covered smut. The field demonstration was laid out on a single plot with 4 treatments; Cow urine 1:1 (v/v) mixture, Thiram, Apron plus with 3g/kg, and Control untreated as a check. The plot size of the experiment was 10m x 10m with a plant spacing of 15cm and 75cm row spacing. Data on covered smut incidence and grain yield were recorded yield loss and partial budget analysis was made. Training of trainers on sorghum agronomic practices and diseases was given to 79 male and 37 female development agents for four woreda in central Gondar and development agents also gave training for 7925 male and 502 female farmers. There was no infestation of covered smut on fungicide dressed and cultural cow urine-soaked plots on both locations while an incidence of 2.8% at Gondar Zuria and 22.7% at Mirab Belesa. Relative yield advantage of the fungicides and cow urine is calculated over the control untreated check. Yield advantage of 20-26% on apron plus, 13-30% on Thiram, and 16-35% on cow urine were obtained over the untreated control check. Farmers have to use the nearby available cultural (cow urine) practice method and if not one of the available fungicides for control of covered kernel smut on sorghum production and should also be scaled out

    Pt nanoparticles under oxidizing conditions – implications of particle size, adsorption sites and oxygen coverage on stability

    Platinum nanoparticles are efficient catalysts for different reactions, such as oxidation of carbon and nitrogen monoxides. Adsorption and interaction of oxygen with the nanoparticle surface, taking place under reaction conditions, determine not only the catalytic efficiency but also the stability of the nanoparticles against oxidation. In this study, platinum nanoparticles in oxygen environment are investigated by systematic screening of initial nanoparticle–oxygen configurations and employing density functional theory and a thermodynamics-based approach. The structures formed at low oxygen coverages are described by adsorption of atomic oxygen on the nanoparticles whereas at high coverages oxide-like species are formed. The relative stability of adsorption configurations at different oxygen coverages, including the phase of fully oxidized nanoparticles, is investigated by constructing p–T phase diagrams for the studied systems

    The Statistics of Calcium-Mediated Focal Excitations on a One-Dimensional Cable

    ABSTRACT It is well known that various cardiac arrhythmias are initiated by an ill-timed excitation that originates from a focal region of the heart. However, up to now, it is not known what governs the timing, location, and morphology of these focal excitations. Recent studies have shown that these excitations can be caused by abnormalities in the calcium (Ca) cycling system. However, the cause-and-effect relationships linking subcellular Ca dynamics and focal activity in cardiac tissue is not completely understood. In this article, we present a minimal model of Ca-mediated focal excitations in cardiac tissue. This model accounts for the stochastic nature of spontaneous Ca release on a one-dimensional cable of cardiac cells. Using this model, we show that the timing of focal excitations is equivalent to a first passage time problem in a spatially extended system. In particular, we find that for a short cable the mean first passage time increases exponentially with the number of cells in tissue, and is critically dependent on the ratio of inward to outward currents near the threshold for an action potential. For long cables excitations occurs due to ectopic foci that occur on a length scale determined by the minimum length of tissue that can induce an action potential. Furthermore, we find that for long cables the mean first passage time decreases as a power law in the number cells. These results provide precise criteria for the occurrence of focal excitations in cardiac tissue, and will serve as a guide to determine the propensity of Ca-mediated triggered arrhythmias in the heart

    Microbiological Safety of Street Vended Foods in Jigjiga City, Eastern Ethiopia

    Background: Food safety problems are particularly becoming an increasingly serious threat to public health in developing countries. This study was conducted to assess microbiological safety of street vended foods from May to November, 2014 in Jigjiga City.Methods: A cross-sectional design was used to answer questions concerning the current status of food hygiene and sanitation practire of street food vending sites. Interview and observational assessments were used to collect socio-demographic data about street food venders. One hundred thirty-two samples of street foods were aseptically collected from four ‘kebeles’ of Jigjiga City. Both descriptive and analytical statistical methods were applied.Results: The majority of the street food vendors were women, 120(90.9%), with the average age group of 23-49 years, (42.85%), and 99(66.7%) them were illiterate. The study revealed that 95(72%) of the food samples had pathogenic bacterial contaminations. Three different bacterial species were isolated: E. coli 68(51.5%), S. aureus 85(64.4%) and 26(19.7%) Salmonella species. The highest incidence of S. aureus 23/33(69%) was seen in ‘Sambusa’; the highest incidence of E. coli 24/33(73.5%) was observed in ‘Pasta’, while the highest Salmonella incidence was observed in ‘Ades’.Conclusion: This study revealed that there is a reasonable gap on food safety knowledge among street food venders. The microbial profile was also higher compared to standards set by the World Health Organization. Due attention should be given by the government to improve knowledge about food safety and the quality standard of street foods sold in the City.Keywords: Microbiological safety, Street vended foods, Isolation, Jigjig

    Zingiber Officinale Roscoe and Echinops Kebericho Mesfin Showed Antiplasmodial Activities against Plasmodium Berghei in a Dosedependent Manner in Ethiopia

    BACKGROUND: The emergence and spread of Plasmodium falciparum resistance to antimalarial drugs necessitated the search for new drugs from natural products. Zingiber officinal Roscoe and Echinops Kebericho Mesfin are traditional herbal medicines widely used for the treatment of malaria in Ethiopia. The aim of the study was to assess the toxicity profile and in vivo antiplasmodial activities of 70% methanol crude extracts of both plant materials against Plasmodium berghei.METHODS: Healthy male Swiss Albino mice of age 4-5 weeks and weight 25-36 g were infected by P. berghei. The extracts were administered orally at doses 5000, 2500 and 1250 mg/kg for acute toxicity of E. kebericho Mesfin. Graded doses at 1000, 500 and 250 mg/kg used for four days suppressive studies. Parasitemia, body weight, packed cell volume (PCV) and survival time were determined. SPSS Version 20 was used for the analysis of data of parasitemia, body weight, PCV, and survival times. Statistical significance was determined by one-way ANOVA. Independent ttest was used to compare results. Results were presented as a mean ± standard error of the mean (M ± SEM). All data were analyzed at a 95% confidence interval (α= 0.05).RESULTS: At the dose of 5000 mg/kg, E. kebericho Mesfin showed no toxic effects. The LD50 of extract could go beyond the dose used. In vivo antiplasmodial activity of extracts showed excellent chemo suppression at 500 and 1000 mg/kg in a dose dependent manner compared with the negative control. The chemo suppressions of the 1000 mg/kg of both plant extracts were 49.53 ± 1.90% and 32.83 ± 1.03%, respectively. The survival times of P. berghei infected mice were also a dose dependent manner while failed to prevent weight loss.CONCLUSION: The extracts of both medicinal plants showed antiplasmodial activities against P. berghei. It confirmed the literature findings and their traditional uses.

    Plasma Membrane Localization Is Required for RasA-Mediated Polarized Morphogenesis and Virulence of Aspergillus fumigatus

    ABSTRACT Ras is a highly conserved GTPase protein that is essential for proper polarized morphogenesis of filamentous fungi. Localization of Ras proteins to the plasma membrane and endomembranes through posttranslational addition of farnesyl and palmitoyl residues is an important mechanism through which cells provide specificity to Ras signal output. Although the Aspergillus fumigatus RasA protein is known to be a major regulator of growth and development, the membrane distribution of RasA during polarized morphogenesis and the role of properly localized Ras signaling in virulence of a pathogenic mold remain unknown. Here we demonstrate that Aspergillus fumigatus RasA localizes primarily to the plasma membrane of actively growing hyphae. We show that treatment with the palmitoylation inhibitor 2-bromopalmitate disrupts normal RasA plasma membrane association and decreases hyphal growth. Targeted mutations of the highly conserved RasA palmitoylation motif also mislocalized RasA from the plasma membrane and led to severe hyphal abnormalities, cell wall structural changes, and reduced virulence in murine invasive aspergillosis. Finally, we provide evidence that proper RasA localization is independent of the Ras palmitoyltransferase homolog, encoded by erfB , but requires the palmitoyltransferase complex subunit, encoded by erfD . Our results demonstrate that plasma membrane-associated RasA is critical for polarized morphogenesis, cell wall stability, and virulence in A. fumigatus
