The adhesive properties of neural cell adhesion molecules (NCAMs) can be modified by alternative splicing of the primary transcript or posttranslational modifications. In the present study, we describe distinct forms of alternative splicing and posttranslational modification of the extracellular domain of NCAM of various endocrine tissues and derived tumor cells of the rat. Using an antiserum detecting the immunoglobulin-like domains of NCAM as well as a monoclonal antibody recognizing the NCAM-specific polysialic acid (PSA), we observed a similar staining pattern in adrenals, pituitary, and neoplastic endocrine cells. In endocrine tumor cells [pheochromocytoma (PC12), insulinoma (RINA2), and pituitary tumor cells (GH3)], NCAM immunoreactivity was most intense at contact sites between the cells. The immunocytochemical data were substantiated by results of in situ hybridization histochemistry. Specifically, higher levels of NCAM mRNA were detected in the adrenal cortex than in the medulla. In the pituitary, NCAM mRNA was more abundant in the anterior and intermediate lobes than in the neural lobe. The sequence of NCAM mRNAs in endocrine cells was analyzed by polymerase chain reaction and S1 nuclease protection assays. We found that major exons 4-13 of the NCAM mRNA in endocrine tissues and related tumor cell lines were homologous to those in the brain. However, PC12, RINA2, and GH3 tumor cells; normal rat pituitaries; and adrenals contained different amounts of NCAM mRNA with an alternative extra exon, termed VASE (also called pi in mouse) between constitutive exons 7 and 8. In addition, in pituitaries, we detected an alternative exon in splice site a between the constitutive exons 12 and 13, termed a15, with or without an AAG triplett. These sites are thought to be important for the adhesive properties of NCAM. Therefore, these results suggest that modifications of NCAM may be important for adhesive interactions in normal and neoplastic endocrine cells

Context: Chemically peculiar A type (Ap) stars are a subgroup of the CP2 stars which exhibit anomalous overabundances of numerous elements, e.g. Fe, Cr, Sr and rare earth elements. The pulsating subgroup of the Ap stars, the roAp stars, present ideal laboratories to observe and model pulsational signatures as well as the interplay of the pulsations with strong magnetic fields and vertical abundance gradients. Aims: Based on high resolution spectroscopic observations and observed stellar energy distributions we construct a self consistent model atmosphere, that accounts for modulations of the temperature-pressure structure caused by vertical abundance gradients, for the roAp star 10 Aquilae (HD 176232). We demonstrate that such an analysis can be used to determine precisely the fundamental atmospheric parameters required for pulsation modelling. Methods: Average abundances were derived for 56 species. For Mg, Si, Ca, Cr, Fe, Co, Sr, Pr, and Nd vertical stratification profiles were empirically derived using the ddafit minimization routine together with the magnetic spectrum synthesis code synthmag. Model atmospheres were computed with the LLModels code which accounts for the individual abundances and stratification of chemical elements. Results: For the final model atmosphere Teff=7550 K and log g=3.8 were adopted. While Mg, Si, Co and Cr exhibit steep abundance gradients Ca, Fe and Sr showed much wider abundance gradients between log tau_5000=-1.5 and 0.5. Elements Mg and Co were found to be the least stratified, while Ca and Sr showed strong depth variations in abundance of up to ~ 6 dex.Comment: 9 pages, 15 figure

Modern business services constitute one of the most dynamic developing branches in the economy. This is especially true in Poland. In the last dozen or so years in several Polish cities offering specific location factors over 500 outsourcing service centres have started up. Together they employ over 150 thousand people (in 2015). Intense economic links within these cities have been leading to the establishment of yet more service centres. This branch has been a driving force in the modern office estate market. The article highlights the issue of location choice for outsourcing centres in Poland. Taking into consideration the high dynamics of development of this based on the knowledge economy sector, this issue may be important for business investors, local government units, as well as local business entities. The authors attempt to identify the factors which influence the decisions of managers choosing the place for this activity. A nationwide empirical study was conducted using standardised questionnaire interview addressed to decision-makers in the processes of choosing a location for such centres. The results of these studies include the list of location factors specifying the gradation of their significance to investors when deciding about location

Entanglement interferometry for precision measurement of atomic scattering properties

We report on a two-particle matter wave interferometer realized with pairs of trapped 87Rb atoms. Each pair of atoms is confined at a single site of an optical lattice potential. The interferometer is realized by first creating a coherent spin-mixture of the two atoms and then tuning the inter-state scattering length via a Feshbach resonance. The selective change of the inter-state scattering length leads to an entanglement dynamics of the two-particle state that can be detected in a Ramsey interference experiment. This entanglement dynamics is employed for a precision measurement of atomic interaction parameters. Furthermore, the interferometer allows to separate lattice sites with one or two atoms in a non-destructive way.Comment: 4 pages, 5 figure

Leydig cells express neural cell adhesion molecules in vivo and in vitro

The neural cell adhesion molecule (NCAM) polypeptides are expressed by numerous tissues during embryonic development, where they are involved in cell-cell interactions. In the adult, NCAM expression is confined to a few cell types, including neurons and peptide-hormone-producing cells. Here we demonstrate that the Leydig cells of the adult rat, mouse, and hamster testes express NCAM as well. Western blotting showed that an NCAM of approximately 120 kDa was present in the adult testes of all three species investigated. This form was also found in freshly isolated mouse Leydig cells and in Leydig cells after 2 days in culture. After 4 days in culture, mouse Leydig cells expressed additional NCAM isoforms of approximately 140 and 180 kDa, indicating changes in alternative splicing of NCAM primary transcripts. Also, NCAM mRNA of all isoforms, as detected by S1-nuclease protection assays, increased with time in culture. The expression of the cell adhesion molecule NCAM by adult Leydig cells may explain the aggregation of Leydig cells in clusters in rodent testes, which could be a prerequisite for functional coordination of groups of Leydig cells. Furthermore, the presence of this neural and endocrine marker may indicate a closer relationship between Leydig cells and neural and peptide-hormone-producing cells than is considered to exist at the present time

Coherent transport of neutral atoms in spin-dependent optical lattice potentials

We demonstrate the controlled coherent transport and splitting of atomic wave packets in spin-dependent optical lattice potentials. Such experiments open intriguing possibilities for quantum state engineering of many body states. After first preparing localized atomic wave functions in an optical lattice through a Mott insulating phase, we place each atom in a superposition of two internal spin states. Then state selective optical potentials are used to split the wave function of a single atom and transport the corresponding wave packets in two opposite directions. Coherence between the wave packets of an atom delocalized over up to 7 lattice sites is demonstrated.Comment: 4 pages, 6 figure

Attainable circulation about airfoils in cascade

From consideration of available information on boundary-layer behavior, a relation among profile thickness, maximum surface velocity, Reynolds number, velocity diagram, and solidity is established for a cascade of airfoils immersed in a two-dimensional incompressible fluid flow. Several cascades are computed to show the effect of various cascade design parameters on minimum required cascade solidity. Comparisons with experimentally determined blade performance show that the derived blade loadings are equal or higher for moderate flow deceleration and somewhat lower for large deceleration. Blades with completely laminar flow appear practical for impulse or reaction blading
