Preliminary flagellin gene transfection into tumor cells - attempts of generating anti-tumor response in experimental model
The development of new research techniques, especially molecular ones, creates hopes for improved treatment efficacy and a better prognosis in lung cancer. The starting point for very important experiments in the field of immunotherapy is the sensitization of the immune system to the tumor antigen . The aim of the study was to determine whether using flagellin can induce innate antitumor effective, and thus, increase the immunogenicity of the tumor. The test material was the established mouse cell line LLC, derived from Lewis lung cancer. Vectors were constructed by cloning FliC inserts into the pCDH-MSC-T2A Puro plasmid. The cells were transfected directly and indirectly (with pseudoviruses, produced by previously transfected packaging cells HEK 293T). The transfection efficiency was confirmed by RT-PCR. The cells thus prepared were implanted subcutaneously in mice. The control group received non-transfected LLC cells. Then, after 6 weeks, the mice were scarified. The animals were sectioned to isolate the tumor and lungs in which the presence or absence of metastases was assessed. In mice that received immunotherapeutic vaccines containing transfected LLC cells, less tumor mass was found or a complete lack of it, and the lifespan was noticeably prolonged. In addition, no metastases occurred in the group vaccinated with cells transfected with direct technique. It can be concluded that flagellin is effective as an adjuvant in the process of presenting tumor antigens to T cells Thus, in the light of recent studies and reports, it is likely that in this experiment effectively increased immunogenicity of the tumor. Activating anti-tumor cellular responses using flagellin is a promising target for lung cancer immunotherapy

Streszczenie
Wprowadzenie: Proces degradacji białek zależny od retikulum endoplazmatycznego (ang. endoplasmic-reticulum-associated protein degradation - ERAD) odgrywa istotną rolę w utrzymaniu stanu homeostazy wewnątrzkomórkowej i dotyczy przede wszystkim białek, które nie uzyskały posttranslacyjnie prawidłowej konformacji. Mechanizm ten może zostać wykorzystany przez wirusy, zwykle celem uniknięcia detekcji przez układ odpornościowy gospodarza. Zależnie od typu wirusa, rodzaj i sposób degradacji białek bywa odmienny.
Cel pracy: Celem pracy jest przedstawienie mechanizmu ERAD oraz jego znaczenia dla cyklu infekcyjnego wybranych cząsteczek wirusowych. Dokładne poznanie tego procesu może istotnie przyczynić się do wskazania potencjalnie skutecznych, nowych przeciwwirusowych rozwiązań terapeutycznych.
Skrócony opis stanu wiedzy: W pracy omówiono mechanizm ERAD i jego wykorzystanie przez: Poliomawirusy (BK i SV-40), Herpeswirusy (Cytomegalowirus), Retrowirusy (HIV-I) oraz Hepadnawirusy (HBV).
Podsumowanie: Znaczenie ERAD w cyklu infekcyjnym wirusów jest niezwykle istotne w przebiegu zakażenia. Należy dalej badać możliwość wykorzystywania ERAD przez inne rodzaje wirusów. Warto również monitorować skuteczność inhibitorów proteasomu jako potencjalnych leków w walce z chorobami wirusowymi.

Summary
Introduction: The endoplasmic-reticulum-associated protein degradation (ERAD) pathway plays an important role in maintaining intracellular homeostasis and primarily affects proteins that have not achieved correct post-translational conformation. ERAD can be also exploited by viruses, usually in order to avoid detection by the host's immune system. Depending on the type of virus, the character and manner of protein degradation may be different.
Aim of the study: The aim of this paper is to present the ERAD mechanism and its importance for the infection cycle of select viral particles. Gaining knowledge about this process can help to identify potentially effective new antiviral therapies.
Short description of state of knowledge: The ERAD mechanism and its utilization by: Poliomaviruses (BK and SV-40), Herpesviruses (Cytomegalovirus), Retroviruses (HIV-I) and Hepadnaviruses (HBV) are discussed in the paper.
Summation: The ERAD process in the viral infection cycle is extremely important. The possibility of using ERAD by differnet viruses should be further investigated. It could be also useful to exploit proteasome inhibitors as potential drugs in the fight against viral diseases.

EGFR potencjalnym celem dla terapii genowej nowotworów
Receptor dla naskórkowego czynnika wzrostu (ang. epidermal growth factor
receptor; EGFR) to jeden z protoonkogenów, które bardzo często ulegają nadekspresji
w komórkach nowotworowych. Podwyższoną ekspresję tego receptora
zauważono w przypadku raka trzustki, jelita grubego, wątroby, jajników, piersi
i płuc. Zwiększona ilość EGFR w błonie komórkowej komórek nowotworowych
jest szczególnie niepokojąca, ponieważ receptor ten jest zdolny do wiązania wielu
ligandów, nie tylko EGF (ang. epidermal growth factor), ale także innych cząsteczek,
takich jak na przykład TGF-α (ang. transforming growth factor-α), czy
też naskórkowy czynnik wzrostowy wiążący heparynę (HB‑EGF; heparyne-
-binding EGF‑like growth factor). Związanie się liganda ze wspomnianym wyżej
receptorem skutkuje zwiększoną proliferacją komórek nowotworowych, ich
unieśmiertelnieniem i zmniejszeniem wrażliwości na sygnały proapoptotyczne, co
w efekcie przekłada się na większą agresywność nowotworu i gorsze rokowanie.
Z tego powodu możliwość wyciszenia ekspresji EGFR w komórkach nowotworowych
wydaje się niezwykle atrakcyjną strategią w terapii nowotworów. Niniejsza
praca skupia się na zahamowaniu szlaków sygnałowych, w które zaangażowany
jest EGFR, za pomocą plazmidu kodującego wstawki shRNA (ang. short hairpin
RNA) anty-EGFR i anty-HER2. Potencjalne wykorzystanie plazmidu jako nośnika
wstawki wyciszającej wydaje się skuteczną metodą, co potwierdzono późniejszym
rozdziałem elektroforetycznym cDNA uzyskanego w reakcji RT-PCR z wyizolowanego
z komórek linii A549 RNA. Badanie stanu apoptozy w komórkach
przeprowadzono wykorzystując barwienie jodkiem propidyny (ang. propidium
iodide; PI) oraz aneksyną V. Ukazało ono znamienny statystycznie wzrost odsetka
komórek w późnej apoptozie po transfekcji plazmidem ze wstawką shRNA anty-
EGFR. Wyniki badań pozwalają sądzić, że hamowanie genu dla EGFR może
mieć znaczenie w ewentualnych próbach immunoterapii i terapii genowej niedrobnokomórkowego
raka płuca
CD83 – niezidentyfikowany marker ucieczki nowotworu spod nadzoru immunologicznego gospodarza
Komórki dendrytyczne (DC) odgrywają znaczącą rolę w pobudzaniu limfocytów,
zwłaszcza dziewiczych, gdyż stanowią najsilniejszą frakcję komórek prezentujących
antygen (APC, ang. Antigen Presenting Cells). Znaczącą rolę w dojrzewaniu
DC pełni cząsteczka CD83, która jest jednocześnie ich markerem dojrzałości. Jej
ekspresję odkryto, zarówno na innych prawidłowych komórkach APC, jak i na
komórkach nowotworowych, co skłania do refleksji nad pytaniem, czy cząsteczka
CD83 może brać udział w patogenezie tego rodzaju schorzeń.
Celem pracy była ocena częstości występowania cząsteczki CD83, w wybranych
modelowych liniach ustalonych ludzkich nowotworów złośliwych (MDA-231
- rak sutka, TOV-112D - rak jajnika).
Wyniki otrzymano dzięki wykorzystaniu techniki immunofluorescencji bezpośredniej
w połączeniu z odczytem w cytometrze przepływowym.
W wyniku przeprowadzonych badań odnotowano znaczną ekspresję formy CD83
zakotwiczonej w błonie na powierzchni komórek badanych linii ustalonych.
Przeprowadzone doświadczenia wskazują na możliwą rolę cząsteczki CD83 w patogenezie
nowotworów. Jej wysoka ekspresja na powierzchni komórek, w połączeniu
z możliwością jej „złuszczenia” z powierzchni komórki w formie rozpuszczalnej
do mikrośrodowiska nowotworu może nasilać zjawisko immunosupresji
w układzie immunologicznym i tym samym powodować ucieczkę nowotworu
spod nadzoru immunologicznego organizmu gospodarza. W celu określenia dokładnej
roli tytułowej cząsteczki niezbędne są dalsze badania

Streszczenie
Wprowadzenie:
Skóra jako największy i najłatwiej dostępny narząd stanowi atrakcyjny cel dla terapii genowej, która od wielu lat budzi ogromne nadzieje środowiska naukowego. Jednakże, próby terapii przeprowadzone z wykorzystaniem wektorów wirusowych wykazały szereg wad i ograniczeń m.in. obserwowano indukcję odpowiedzi immunologicznej, losową integrację transgenu z genomem gospodarza i/lub niską wydajność jego ekspresji. Dlatego, wciąż poszukuje się alternatywnych, skuteczniejszych i jednocześnie bezpieczniejszych metod transferu genów. Atrakcyjnej alternatywy upatruje się w metodach niewirusowych.
Cel pracy:
Przedstawienie wybranych metod niewirusowego transferu genów wykorzystywanych w terapii genowej chorób skóry.
Skrócony opis stanu wiedzy:
Terapia genowa chorób skóry obejmuje wykorzystanie wektorów plazmidowych jako nośnika genów terapeutycznych, a także metod ich dostarczania do komórek takich jak: elektroporacja, mikroiniekcja, sonikacja, wykorzystanie nośników lipidowych i polimerów kationowych.
Podsumowanie:
Niewirusowe metody transferu genów oferują pewne zalety włączając niską toksyczność, brak infekcyjności oraz łatwość i niskie koszty produkcji w porównaniu z technikami wirusowymi. Niewirusowe metody wydają się być obiecującym narzędziem terapii genowej chorób skóry w szczególności nowotworów tego narządu.

Summary
Introduction:
Skin, the largest and most accessible organ of the human body is considered as an ideal gene therapy target. However, various types of viral vectors used in classical gene therapy have a number of disadvantages, such as possibility of immune response induction, random integration of inserts into the host genome or low expression efficiency. Therefore, there is an urgent need for alternative, non-viral methods of gene transfer.
Aim of the study:
To present methods for non-viral gene transfer used in gene therapy of skin diseases.
Short description of knowledge state:
Gene therapy for skin diseases include the usage of plasmid vectors as a carrier for therapeutic genes and different methods for their delivery into cells such as: electroporation, microinjection, sonication, lipid carriers and cationic polymers.
Summary:
Non-viral gene transfer methods offer some advantages including lower toxicity, non-infectious properties, ease of production and low costs as compared to viral techniques. Non-viral approaches are the promising tool in gene therapy of skin diseases, in particular in skin cancer ceases.
