    Hans-Georg Gadamer: The rediscovery of truth

    When the first publication of Hans-Georg Gadamer's magnum opus Wahrheit und Methode (Truth and Method) came to life in I960, the work was initially received with a slight sense of puzzlement and yet concurrently acknowledged as monumental. The title, in English, Truth and Method, was regarded by the philosophical community both in Germany and abroad as being somewhat obscure, as Gadamer himself would later admit, but the ingeniousness of the book's content could hardly be debated. Since its initial publication, Truth and Method has, respectively helped to expand and light up the horizon of modern hermeneutics by provoking, at once, a reconsideration of the phenomenon of understanding while, at the same time, enlivening the debate over scientific methodology and its exclusive claim to truth. The central aim of the present thesis has been to focus on Part I of Truth and Method, concentrating primarily on the 'guiding humanistic concepts' and the experience of truth in art, to clarify Gadamer's understanding of truth and to shed new light as to how the experience of tmth is to be grasped in relation to the human sciences, i.e. the humanities. The humanistic concepts, I believe, are vital to understanding the experience of truth. One reason, which leads me to this conclusion, is that in Truth and Method Gadamer begins his philosophical undertaking with the elucidation of the humanistic concepts rather than with a direct exposition of truth. By opening with the humanistic concepts, Gadamer seems to demonstrate subtly the phenomenological and ontological nature of knowledge and understanding. The outcome of this manoeuvre is that one comes to realise that truth does not simply belong to method and that it is not something which can be defined solely as 'absolute certainty'. Moreover, in my interview with Professor Gadamer, the humanistic concepts, he explained to me, are the most 'natural' and 'original' concepts. By natural and original he means that these concepts are intrinsic. They evolve from life as well as being a part of life, i.e. a way of living. Thus these concepts, he affirmed, represent 'a way of life' and a way to truth. Consequently, insofar as comprehending the phenomenon of truth, I believe any and every investigation of the concept of truth must begin with the understanding of the humanistic tradition. The following thesis however does not end simply with the humanistic concepts. It also devotes to examining the truth-claim or the 'truth-experience' of art. This part of the inquiry centres on two important questions: (1.) How are we to understand art? (2.) What does it mean to experience art? The challenge here has been to show how Gadamer overcomes Kant's subjectivation of the aesthetic experience and to demonstrate how and why Gadamer considers the experience of art as the 'self-presentation' of being. In surveying the various works of criticism I have tried to draw attention to what seem to me to be the most insightful comments and analyses. If 1 have failed in any way to supply proper acknowledgement to ideas, which might seem close to other critical works, I offer my apologies. As I am sure those in the research business know well, in reflection ideas often interfuse with one's own, making it difficult sometimes to discriminate between one's own ideas from another's.     Do poverty and wealth look the same the world over? A comparative study of 12 cities from five high-income countries using street images

    Urbanization and inequalities are two of the major policy themes of our time, intersecting in large cities where social and economic inequalities are particularly pronounced. Large scale street-level images are a source of city-wide visual information and allow for comparative analyses of multiple cities. Computer vision methods based on deep learning applied to street images have been shown to successfully measure inequalities in socioeconomic and environmental features, yet existing work has been within specific geographies and have not looked at how visual environments compare across different cities and countries. In this study, we aim to apply existing methods to understand whether, and to what extent, poor and wealthy groups live in visually similar neighborhoods across cities and countries. We present novel insights on similarity of neighborhoods using street-level images and deep learning methods. We analyzed 7.2 million images from 12 cities in five high-income countries, home to more than 85 million people: Auckland (New Zealand), Sydney (Australia), Toronto and Vancouver (Canada), Atlanta, Boston, Chicago, Los Angeles, New York, San Francisco, and Washington D.C. (United States of America), and London (United Kingdom). Visual features associated with neighborhood disadvantage are more distinct and unique to each city than those associated with affluence. For example, from what is visible from street images, high density poor neighborhoods located near the city center (e.g., in London) are visually distinct from poor suburban neighborhoods characterized by lower density and lower accessibility (e.g., in Atlanta). This suggests that differences between two cities is also driven by historical factors, policies, and local geography. Our results also have implications for image-based measures of inequality in cities especially when trained on data from cities that are visually distinct from target cities. We showed that these are more prone to errors for disadvantaged areas especially when transferring across cities, suggesting more attention needs to be paid to improving methods for capturing heterogeneity in poor environment across cities around the world

    T cell derived IL-10 is dispensable for tolerance induction in a murine model of allergic airway inflammation

    Regulatory mechanisms initiated by allergen specific immunotherapy are mainly attributed to T cell-derived IL-10. However, it has not been shown that T cell-derived IL-10 is required for successful tolerance induction. Here, we analyze cellular sources and the functional relevance of cell type specific IL-10 during tolerance induction in a murine model of allergic airway inflammation. While tolerance induction was effective in IL-10 competent mice, neutralizing IL-10 prior to tolerogenic treatment completely abrogated the beneficial effects. Cellular sources of IL-10 during tolerance induction were identified by using transcriptional reporter mice as T cells, B cells and to a lesser extent DCs. Interestingly, tolerance induction was still effective in mice with T cell-, B cell-, B and T cell- or DC-specific IL-10 deficiency. In contrast, tolerance induction was not possible in mice lacking IL-10 in all hematopoetic cells, while it was effective in bone marrow chimera that lacked IL-10 only in non-hematopoetic cells. Taken together, allergen specific tolerance depends on IL-10 from hematopoetic sources. The beneficial effects of allergen specific immunotherapy cannot solely be attributed to IL-10 from T cells, B cells or even DCs, suggesting a high degree of cellular redundancy in IL-10 mediated tolerance

    Evaluation of the tuberculin skin test and the interferon-Îł release assay for TB screening in French healthcare workers

    <p>Abstract</p> <p>Introduction</p> <p>Using French cut-offs for the Tuberculin Skin Test (TST), results of the TST were compared with the results of an Interferon-γ Release Assay (IGRA) in Healthcare Workers (HCW) after contact to AFB-positive TB patients.</p> <p>Methods</p> <p>Between May 2006 and May 2007, a total of 148 HCWs of the University Hospital in Nantes, France were tested simultaneously with IGRA und TST. A TST was considered to indicate recent latent TB infection (LTBI) if an increase of >10 mm or if TST ≥ 15 mm for those with no previous TST result was observed. For those with a positive TST, chest X-ray was performed and preventive chemotherapy was offered.</p> <p>Results</p> <p>All HCWs were BCG-vaccinated. The IGRA was positive in 18.9% and TST ≥ 10 mm was observed in 65.5%. A recent LTBI was believed to be highly probable in 30.4% following TST. Agreement between IGRA and TST was low (kappa 0.041). In 10 (16.7%) out of 60 HCWs who needed chest X-ray following TST the IGRA was positive. In 9 (20%) out of 45 HCWs to whom preventive chemotherapy was offered following TST the IGRA was positive. Of those considered TST-negative following the French guidelines, 20.5% were IGRA-positive. In a two-step strategy - positive TST verified by IGRA - 18 out of 28 (64.3%) IGRA-positive HCWs would not have been detected using French guidelines for TST interpretation.</p> <p>Conclusion</p> <p>The introduction of IGRA in contact tracings of BCG-vaccinated HCWs reduces X-rays and preventive chemotherapies. Increasing the cut-off for a positive TST does not seem to be helpful to overcome the effect of BCG vaccination on TST.</p

    Stem cell-associated heterogeneity in Glioblastoma results from intrinsic tumor plasticity shaped by the microenvironment

    The identity and unique capacity of cancer stem cells (CSC) to drive tumor growth and resistance have been challenged in brain tumors. Here we report that cells expressing CSC-associated cell membrane markers in Glioblastoma (GBM) do not represent a clonal entity defined by distinct functional properties and transcriptomic profiles, but rather a plastic state that most cancer cells can adopt. We show that phenotypic heterogeneity arises from non-hierarchical, reversible state transitions, instructed by the microenvironment and is predictable by mathematical modeling. Although functional stem cell properties were similar in vitro, accelerated reconstitution of heterogeneity provides a growth advantage in vivo, suggesting that tumorigenic potential is linked to intrinsic plasticity rather than CSC multipotency. The capacity of any given cancer cell to reconstitute tumor heterogeneity cautions against therapies targeting CSC-associated membrane epitopes. Instead inherent cancer cell plasticity emerges as a novel relevant target for treatment.publishedVersio

    The effect of cartilage and bone density of mushroom-shaped, photooxidized, osteochondral transplants: an experimental study on graft performance in sheep using transplants originating from different species

    BACKGROUND: Differences in overall performance of osteochondral photooxidized grafts were studied in accordance of their species origin and a new, more rigorous cleansing procedure using alcohol during preparation. METHODS: Photooxidized mushroom-shaped grafts of bovine, ovine, human and equine origin were implanted in the femoral condyles of 32 sheep (condyles: n = 64). No viable chondrocytes were present at the time of implantation. Grafts were evaluated at 6 months using plastic embedded sections of non-decalcified bone and cartilage specimens. Graft incorporation, the formation of cyst-like lesions at the base of the cartilage junction as well as cartilage morphology was studied qualitatively, semi-quantitatively using a score system and quantitatively by performing histomorphometrical measurements of percentage of bone and fibrous tissue of the original defects. For statistical analysis a factorial analysis of variance (ANOVA- test) was applied. RESULTS: Differences of graft performance were found according to species origin and cleansing process during graft preparation. According to the score system cartilage surface integrity was best for equine grafts, as well as dislocation or mechanical stability. The equine grafts showed the highest percentage for bone and lowest for fibrous tissue, resp. cystic lesions. The new, more rigorous cleansing process decreased cartilage persistence and overall graft performance. CONCLUSION: Performance of grafts from equine origin was better compared to bovine, ovine and human grafts. The exact reason for this difference was not proven in the current study, but could be related to differences in density of cartilage and subchondral bone between species
