    Briefing: UK Ministry of Defence Force Protection Engineering Programme

    The Defence Science and Technology Laboratory sponsored, QinetiQ-led Force Protection Engineering Research Programme has two main strands, applied and underpinning research. The underpinning strand is led by Blastech Ltd. One focus of this research is into the response of geomaterials to threat loading. The programme on locally won fill is split into four main characterisation strands: high-stress (GPa) static pressureā€“volume; medium-rate pressureā€“volume (split Hopkinson bar); high-rate (flyer plate) pressureā€“volume; and unifying modelling research at the University of Sheffield, which has focused on developing a high-quality dataset for locally won fill in low and medium strain rates. With the test apparatus at Sheffield well-controlled tests can be conducted at both high strain rate and pseudo-static rates up to stress levels of 1 GPa. The University of Cambridge has focused on using one-dimensional shock experiments to examine high-rate pressureā€“volume relationships. Both establishments are examining the effect of moisture content and starting density on emergent rate effects. Blastech Ltd has been undertaking carefully controlled fragment impact experiments, within the dataspace developed by the Universities of Sheffield and Cambridge. The data from experiments are unified by the QinetiQ-led modelling team, to predict material behaviour and to derive a scalable locally won fill model for use in any situation

    Demyelination and axonal preservation in a transgenic mouse model of Pelizaeus-Merzbacher disease

    It is widely thought that demyelination contributes to the degeneration of axons and, in combination with acute inflammatory injury, is responsible for progressive axonal loss and persistent clinical disability in inflammatory demyelinating disease. In this study we sought to characterize the relationship between demyelination, inflammation and axonal transport changes using a Plp1-transgenic mouse model of Pelizaeus-Merzbacher disease. In the optic pathway of this non-immune mediated model of demyelination, myelin loss progresses from the optic nerve head towards the brain, over a period of months. Axonal transport is functionally perturbed at sites associated with local inflammation and 'damaged' myelin. Surprisingly, where demyelination is complete, naked axons appear well preserved despite a significant reduction of axonal transport.     The Aflatoxin QuicktestTMā€”A Practical Tool for Ensuring Safety in Agricultural Produce

    Contamination of corn, peanuts, milk and dairy products with aflatoxins is a worldwide problem, particularly in subtropical regions where the climatic conditions are ideal for the growth of Aspergillus flavus, the fungi that produces these toxins. Developing countries have major difficulties in marketing these products abroad due to the stringent international regulations concerning this carcinogenic toxin. Adding to the problem is the analytical cost involved in monitoring the produce, which require sophisticated instrumentation and qualified personnel, neither of which are available for field testing. The development of a rapid Aflatoxin Quicktestā„¢ provides an effective, reliable and cheaper option for screening levels of aflatoxin above the regulatory thresholds in such produce. The test consists of a lateral flow device (LFD) coated with antibodies specific to aflatoxin B1, although it detects other aflatoxins (i.e. G and M) with high cross-reactivity. Its high sensitivity allows analysis of these toxins in the range 2ā€“40Ā Ī¼g/kg of sample in 15Ā minutes, plus the time for extraction, which varies among different products. Quantification of the test results is done using a Quick Reader, by comparing the readings of individual tests against a standard curve of the analytes in the same manner as it is done with any other analytical equipment. A validation study was carried out using peanuts from Australia and peanuts and corn from Timor-Leste to assess the performance of the Aflatoxin Quicktestā„¢. Results obtained with the LFD showed a good correlation with the standard analytical measurements by HPLC-fluorescence (r2 above 0.90 for all cases), indicating the Aflatoxin Quicktestā„¢ is capable of measuring levels of aflatoxins accurately and reliably. Given their ease of use, low cost and fast processing time, the Aflatoxin Quicktestā„¢ can be used for screening agricultural produce in countries that cannot afford the costly alternative of using specialised personnel and equipment

    Based on an exceptionally long modern ecological dataset (41 years), it has been possible to show that warm weather in England associated with a positive North Atlantic Oscillation (NAO) index causes the spring migration of the green spruce aphid (Elatobium abietinum), a pest species of spruce trees (Picea) to start earlier, continue for longer and contain more aphids. An upward trend in the NAO index during the period 1966-2006 is associated with an increasing population size of E. abietinum. It is important to understand the mechanisms behind the population fluctuations, because this aphid causes considerable damage to Picea plantations. Present day weather associated fluctuations in forest insect pests may be useful analogues in understanding past pest outbreaks in forests

    Why don't they accept Non-Invasive Ventilation? : Insight into the interpersonal perspectives of patients with MND.

    "Objectives. Although non-invasive ventilation (NIV) can benefit survival and quality of life, it is rejected by a substantial proportion of people with motor neurone disease (MND). The aim of this study was to understand why some MND patients decline or withdraw from NIV. Method. Nine patients withMND(male = 7, mean age = 67 years) participated in this study. These patients, from a cohort of 35 patients who were offered NIV treatment to support respiratory muscle weakness, did not participate in NIV treatment when it was clinically appropriate. Semi-structured interviews and interpretative phenomenological analysis (IPA) were employed to explore these patientā€™s experience of MND and their thoughts and understanding of NIV treatment. Results. Using IPA, four themes were identified: preservation of the self, negative perceptions of NIV, negative experience with health care services, and not needing NIV. Further analysis identified the fundamental issue to be the maintenance of perceived self, which was interpreted to consist of the sense of autonomy, dignity, and quality of life. Conclusions. The findings indicate psychological reasons for disengagement with NIV. The threat to the self, the sense of loss of control, and negative views of NIV resulting from anxiety were more important to these patients than prolonging life in its current form. These findings suggest the importance of understanding the psychological dimension involved in decision-making regarding uptake of NIV and a need for sensitive holistic evaluation if NIV is declined.

    Autoantibodies to truncated GAD(96-585) antigen stratify risk of early insulin requirement in adult-onset diabetes

    We investigated whether characterisation of full-length (f-)GADA responses could identify early insulin requirement in adult-onset diabetes. In 179 f-GADA positive participants diagnosed with type 2 diabetes, we assessed associations of truncated (t-)GADA positivity, f-GADA IgG subclasses, and f-GADA affinity with early insulin requirement (<5 years), type 1 diabetes genetic risk score (T1D GRS), and C-peptide. t-GADA positivity was lower in f-GADA positive without early insulin in comparison to f-GADA positive type 2 diabetes requiring insulin within 5 years, and type 1 diabetes (75% vs. 91% and 95% respectively, p<0.0001). t-GADA positivity (in those f-GADA positive) identified a group with a higher type 1 diabetes genetic susceptibility (mean T1D GRS 0.248 vs. 0.225, p=0.003), lower C-peptide (1156 pmol/L vs. 4289 pmol/L, p=1x10-7), and increased IA-2A positivity (23% vs. 6%, p=0.03). In survival analysis, t-GADA positivity was associated with early insulin requirement compared with those only positive for f-GADA, independently from age of diagnosis, f-GADA titre and duration of diabetes [adjusted HR 5.7 (95% CI 1.4, 23.5), p=0.017]. The testing of t-GADA in f-GADA positive individuals with type 2 diabetes identifies those who have genetic and clinical characteristics comparable to type 1 diabetes and stratifies those at higher risk of early insulin requirement
