    An updated overview of the geographic and bathymetric distribution of Savalia savaglia

    The distribution of gold coral Savalia savaglia is modified on the basis of bibliographic information and recent occurrence data, collected using a ROV (Remotely Operated Vehicle) and SCUBA divers. The species is long-lived, rare and has been exploited in the past by divers for collection purposes. S. savaglia is listed in Annex II of the SPA/BD Protocol of the Barcelona Convention and has a wider distribution than previously thought, including both the Mediterranean Sea and the Atlantic Ocean. Our results highlighted that specimens mainly live at a depth range of 15-90 m, but may reach as deep as 900 m in the Mediterranean Sea. This species can form monospecific facies of hundreds of colonies, as observed in Montenegro (Adriatic Sea), between 10 and 20 m, and in the Canary Islands, at a depth range of 27-70 m. Recent data highlighted numerous cases of specimens that were endangered by lost fishing gear, which exposed this species to further threats. Considering its longevity and structural role, it is urgent to develop an effective protection measure for S. savaglia, thereby increasing research efforts and implementing protection areas for this species

    The coral assemblage of an off-shore deep Mediterranean rocky bank (NW Sicily, Italy)

    In this study we characterized the deep assemblages dwelling at 200–250 m depth on a large shoal off Capo St. Vito Promontory (Northwestern coast of Sicily, South Tyrrhenian Sea) by means of ROV-imaging. Two assemblages of suspension feeders, dominated by the gorgonian Callogorgia verticillata and by the black coral Leiopathes glaberrima, together with a tanatocoenosis of the colonial yellow scleractinian coral Dendrophyllia cornigera, were examined. The three main species were significatively distributed into two areas corresponding to different habitat preferences: a more elevated hardground hosting black corals and a gently sloping, silted rocky bottom hosting the other coral species. The study area is subjected to a heavy pressure from the professional fishery, resulting in the mechanical damage of numerous colonies, some of which are then overgrown by various epibionts including a parasitic bioluminescent zoanthid, new for the Mediterranean fauna, and tentatively identified as Isozoanthus primoidus. In the Mediterranean Sea, these deep off-shore rocky banks are widely known among recreational and professional fishermen due to their rich fish fauna. However, there has been still little effort into quantifying and characterizing the extent of the impact and its consequences on the benthic communities, which may represent, as in this case, only a partial picture of their original structure and extent

    Brachiopod Fauna from the Deep Mediterranean Sea: Distribution Patterns and Ecological Preferences

    Compared to their fossil counterparts, living brachiopods are investigated far less often, due to their occurrence in remote environments such as dark caves or deep environments. Due to the scarcity of studies targeting in situ brachiopods' populations, large-scale information on their distribution and ecological preferences is still lacking, especially on hardgrounds. The extensive employment of remotely operated vehicles (ROVs), however, has opened up the chance to better explore this taxon's diversity and ecology in the mesophotic and bathyal zones. The analysis of over 600 h of video footage collected from 624 sites, from 40 m to 1825 m, located along the Ligurian and Tyrrhenian coasts of Italy and the Sicily Channel, allowed for a large-scale investigation. The four identified species, Novocrania anomala, Gryphus vitreus, Megerlia truncata and Terebratulina retusa, emerged as common macrofaunal components of the explored habitats, especially between 150 m and 250 m, with high occurrences in the northern areas, especially on offshore seamounts. All species can form dense aggregations of individuals, with M. truncata showing the densest populations on steep rocky terraces (up to 773 individuals m(-2)). Except for G. vitreus, the only species also recorded on soft bottoms, the others were found exclusively on hardgrounds, with N. anomala showing a peculiar ability to exploit anthropogenic substrates such as terracotta amphorae. No stable species-specific associations were noted, even if numerous species were frequently observed together. Although brachiopods do not show the conspicuous tridimensionality of large filter-feeders, their substrate occupancy and their role in pelagic-benthic processes support their importance in deep-sea Mediterranean ecosystems

    Structure and status of the Italian red coral forests: What can a large-scale study tell?

    The precious coral Corallium rubrum (Linnaeus, 1758) is a charismatic Mediterranean species. A recent large-scale investigation along the Italian coast highlighted its widespread occurrence at mesophotic and upper bathyal depths, especially on coralligenous and bathyal vertical hardgrounds. The lack of morphometric data limited the considerations on the structure and health status of the populations, fundamental to identify the most vulnerable sites and the correct management actions. For this reason, a ROV dataset, consisting of 624 dives carried out between 40 m and 1825 m in the Ligurian Sea, Tyrrhenian Sea, and Sicily Channel, was analysed to extrapolate quantitative data to describe the populations. Ten random frames were obtained from each of the 170 sites hosting red coral. Density, height, and entanglement were evaluated for about 15700 colonies counted in the frames. The densest populations were mainly found between 40 m and 80 m, with a clear latitudinal density decrease. The mesophotic populations were characterized by both scattered and densely aggregated colonies, while the bathyal ones were dominated by sparse colonies. This study identified 17 major coral areas based on the geographic proximity of the sites hosting red coral and their topographic and oceanographic affinity. The size-frequency distribution of the heights was skewed towards the smaller classes in almost all populations, with a modal class between 2 cm and 4 cm. This study depicted a stress situation of the populations throughout the entire study area, which could be correlated also to the long-term harvesting pressure carried out in the basins. Two additional sources of direct mortality were pointed out in this study. Entanglement to artisanal and recreational fishing gear interested about 18% of the recorded colonies, mainly at mesophotic depths. Almost all populations suffered from mechanical entanglement, with the highest percentages in the Ligurian Sea and Sicilian areas. This study also highlighted a massive occurrence of recent deep mortality events, mainly along the eastern and southern coast of Sardinia and in the Campanian Archipelago. Thirty sites with extensive patches of dead colonies still in place were reported from 70 m to around 200 m, but their formation remains unclear

    Deep coral oases in the South Tyrrhenian Sea

    A Mediterranean ‘‘roche du large’’ ecosystem, represented by four rocky shoals, located a few miles apart on a muddy bottom at 70–130 m depth in the gulf of St. Eufemia (Calabria, South Tyrrhenian Sea), was studied by means of Remotely Operated Vehicle (ROV) photo imaging. The shoals host highly diversified coral communities, mainly composed of arborescent colonies of gorgonians (Callogorgia verticillata, Paramuricea clavata, Paramuricea macrospina, Bebryce mollis, Villogorgia bebrycoides, Corallium rubrum, and Leptogorgia sarmentosa), and antipatharians (Antipathella subpinnata, Antipathes dichotoma and Parantipathes larix). The coral colonies reach high densities (up to ca. 17 colonies m22) and large sizes, such as the over 1.5 m wide antipatharian colonies. We hypothesized that the abundance and composition of the coral assemblages differed significantly among the rocky shoals and with respect to the surrounding soft bottoms. Various environmental variables were tested as possible explanatory factors of the observed differences. Moreover, due to their off- coast localization, we report here that these unique ecosystems are potentially subjected to a strong pressure from the local fishing activities, which were tentatively characterized. The recorded coral b-diversity among the shoals supports the hypothesis that these habitats behave like small oases of hard substrata interspersed in a muddy bottom. Because of their intrinsic beauty and rarity and their biological and ecological value, we stress the need of specific actions aimed at the urgent protection of these oases of biodiversity

    Circulating levels of IL-1 family cytokines and receptors in Alzheimer's disease: new markers of disease progression?

    BACKGROUND: Although the mechanisms underlying AD neurodegeneration are not fully understood, it is now recognised that inflammation could play a crucial role in the initiation and progression of AD neurodegeneration. A neuro-inflammatory network, based on the anomalous activation of microglial cells, includes the production of a number of inflammatory cytokines both locally and systemically. These may serve as diagnostic markers or therapeutic targets for AD neurodegeneration. METHODS: We have measured the levels of the inflammation-related cytokines and receptors of the IL-1 family in serum of subjects with AD, compared to mild cognitive impairment (MCI), subjective memory complaints (SMC), and normal healthy subjects (NHS). Using a custom-made multiplex ELISA array, we examined ten factors of the IL-1 family, the inflammation-related cytokines IL-1α, IL-1β, IL-18, and IL-33, the natural inhibitors IL-1Ra and IL-18BP, and the soluble receptors sIL-1R1, sIL-1R2, sIL-1R3, and sIL-1R4. RESULTS: The inflammatory cytokines IL-1α and IL-1β, their antagonist IL-1Ra, and their soluble receptor sIL-1R1 were increased in AD. The decoy IL-1 receptor sIL-1R2 was only increased in MCI. IL-33 and its soluble receptor sIL-1R4 were also significantly higher in AD. The soluble form of the accessory receptor for both IL-1 and IL-33 receptor complexes, sIL-1R3, was increased in SMC and even more in AD. Total IL-18 levels were unchanged, whereas the inhibitor IL-18BP was significantly reduced in MCI and SMC, and highly increased in AD. The levels of free IL-18 were significantly higher in MCI. CONCLUSIONS: AD is characterised by a significant alteration in the circulating levels of the cytokines and receptors of the IL-1 family. The elevation of sIL-1R4 in AD is in agreement with findings in other diseases and can be considered a marker of ongoing inflammation. Increased levels of IL-1Ra, sIL-1R1, sIL-1R4, and IL-18BP distinguished AD from MCI and SMC, and from other inflammatory diseases. Importantly, sIL-1R1, sIL-1R3, sIL-1R4, and IL-18BP negatively correlated with cognitive impairment. A significant elevation of circulating sIL-1R2 and free IL-18, not present in SMC, is characteristic of MCI and disappears in AD, making them additional interesting markers for evaluating progression from MCI to AD

    Platelet thrombin receptor antagonism and atherothrombosis

    Clinical manifestations of atherothrombotic disease, such as acute coronary syndromes, cerebrovascular events, and peripheral arterial disease, are major causes of mortality and morbidity worldwide. Platelet activation and aggregation are ultimately responsible for the progression and clinical presentations of atherothrombotic disease. The current standard of care, dual oral antiplatelet therapy with aspirin and the P2Y12 adenosine diphosphate (ADP) receptor inhibitor clopidogrel, has been shown to improve outcomes in patients with atherothrombotic disease. However, aspirin and P2Y12 inhibitors target the thromboxane A2 and the ADP P2Y12 platelet activation pathways and minimally affect other pathways, while agonists such as thrombin, considered to be the most potent platelet activator, continue to stimulate platelet activation and thrombosis. This may help explain why patients continue to experience recurrent ischaemic events despite receiving such therapy. Furthermore, aspirin and P2Y12 receptor antagonists are associated with bleeding risk, as the pathways they inhibit are critical for haemostasis. The challenge remains to develop therapies that more effectively inhibit platelet activation without increasing bleeding complications. The inhibition of the protease-activated receptor-1 (PAR-1) for thrombin has been shown to inhibit thrombin-mediated platelet activation without increasing bleeding in pre-clinical models and small-scale clinical trials. PAR-1 inhibition in fact does not interfere with thrombin-dependent fibrin generation and coagulation, which are essential for haemostasis. Thus PAR-1 antagonism coupled with existing dual oral antiplatelet therapy may potentially offer more comprehensive platelet inhibition without the liability of increased bleeding

    Persistence of pristine deep-sea coral gardens in the Mediterranean Sea (SW Sardinia)

    Leiopathes glaberrima is a tall arborescent black coral species structuring important facies of the deep-sea rocky bottoms of the Mediterranean Sea that are severely stifled by fishing activities. At present, however, no morphological in vivo description, ecological characterization, age dating and evaluation of the possible conservation actions have ever been made for any population of this species in the basin. A dense coral population was reported during two Remotely Operated Vehicle (ROV) surveys conducted on a rocky bank off the SW coasts of Sardinia (Western Mediterranean Sea). L. glaberrima forms up to 2 m-tall colonies with a maximal observed basal diameter of nearly 7 cm. The radiocarbon dating carried out on a colony from this site with a 4 cm basal diameter revealed an approximately age of 2000 years. Considering the size-frequency distribution of the colonies in the area it is possible to hypothesize the existence of other millennial specimens occupying a supposedly very stable ecosystem. The persistence of this ecosystem is likely guaranteed by the heterogeneous rocky substrate hosting the black coral population that represents a physical barrier against the mechanical impacts acted on the surrounding muddy areas, heavily exploited as trawling fishing grounds. This favorable condition, together with the existence of a nursery area for catsharks within the coral ramifications and the occurrence of a meadow of the now rare soft bottom alcyonacean Isidella elongata in small surviving muddy inclaves, indicates that this ecosystem have to be considered a pristine Mediterranean deep-sea coral sanctuary that would deserve special protection
