    Psi Performance, Belief in Psi, and Competition in a Game-Show Format

    The current study was designed to explore variables influencing psi performance including gender, belief in psi, and competition between men and women. Features of the experiment involved a methodology using a visual image of the receivers, simple spatial location as targets, and monetary incentive to increase competition in a game-show type of environment. One of the goals of this study was to explore the role of competition between groups of senders and receivers as a facilitator of telepathic communication. Results of a one sample t-test comparing overall hit rate for the sample with expected probability were not significant, t(98)= 1.22,p=. 1 I (one-tailed). However, a trend towards above chance hitting was evident. There was also no significant difference found between hit rates for men and women. In addition, all participants completed a belief questionnaire, on which they indicated their belief in mental telepathy on a 7-point-scale. It was hypothesized that belief ratings would correspond with hit rates—specifically that participants with high belief in psi phenomena would score above chance while those with low belief scores would score below chance. The results of an independent samples t-test show that high belief participants (M= 8.92, SD= 1.75) scored significantly higher on the telepathy task than low belief participants (M= 7.67, SD= 1.53), t(70)= -3.16, p \u3c .001 (one-tailed), with a Cohen\u27s D score of .76 for effect size. Given the strength of these findings, it appears that psi phenomena and mental telepathy, specifically, do occur and may be influenced by belief in psi, group setting, and competition. It is noteworthy when the variable of belief is not taken into consideration, the data appears to be random, yet once belief is considered, findings emerge

    MiR-21 Is Induced by Hypoxia and Down-Regulates RHOB in Prostate Cancer

    Tumour hypoxia is a well-established contributor to prostate cancer progression and is also known to alter the expression of several microRNAs. The over-expression of microRNA-21 (miR-21) has been consistently linked with many cancers, but its role in the hypoxic prostate tumour environment has not been well studied. In this paper, the link between hypoxia and miR-21 in prostate cancer is investigated. A bioinformatic analysis of The Cancer Genome Atlas (TCGA) prostate biopsy datasets shows the up-regulation of miR-21 is significantly associated with prostate cancer and clinical markers of disease progression. This up-regulation of miR-21 expression was shown to be caused by hypoxia in the LNCaP prostate cancer cell line in vitro and in an in vivo prostate tumour xenograft model. A functional enrichment analysis also revealed a significant association of miR-21 and its target genes with processes related to cellular hypoxia. The over-expression of miR-21 increased the migration and colony-forming ability of RWPE-1 normal prostate cells. In vitro and in silico analyses demonstrated that miR-21 down-regulates the tumour suppressor gene Ras Homolog Family Member B (RHOB) in prostate cancer. Further a TCGA analysis illustrated that miR-21 can distinguish between different patient outcomes following therapy. This study presents evidence that hypoxia is a key contributor to the over-expression of miR-21 in prostate tumours, which can subsequently promote prostate cancer progression by suppressing RHOB expression. We propose that miR-21 has good potential as a clinically useful diagnostic and prognostic biomarker of hypoxia and prostate cancer

    Victim evaluations of face-to-face restorative justice conferences: A quasi-experimental analysis

    One major goal of face-to-face restorative justice (RJ) is to help heal the psychological harm suffered by crime victims (Braithwaite, 2002). Substantial evidence from randomized controlled trials (RCTs) has shown that this can be accomplished (Strang, 2002) and more trials are underway (Sherman & Strang, 2004). These outcomes are even more clearly, if less rigorously, demonstrated through retrospective interviews of victims about their feelings before and after RJ took place. We review the responses of victims (N = 210) who participated in trials in Canberra (Australia) and in London, Thames Valley, and Northumbria (UK). Despite substantial variations in offense types, social contexts, nation and race, before-after changes revealed by qualitative and quantitative data are all in the same beneficial direction

    Generation of Functional Human Adipose Tissue in Mice from Primed Progenitor Cells

    Adipose tissue (AT) is used extensively in reconstructive and regenerative therapies, but transplanted fat often undergoes cell death, leading to inflammation, calcification, and requirement for further revision surgery. Previously, we have found that mesenchymal progenitor cells within human AT can proliferate in three-dimensional culture under proangiogenic conditions. These cells (primed ADipose progenitor cells, PADS) robustly differentiate into adipocytes in vitro (ad-PADS). The goal of this study is to determine whether ad-PADS can form structured AT in vivo, with potential for use in surgical applications. Grafts formed from ad-PADS were compared to grafts formed from AT obtained by liposuction after implantation into nude mice. Graft volume was measured by microcomputed tomography scanning, and the functionality of cells within the graft was assessed by quantifying circulating human adiponectin. The degree of graft vascularization by donor or host vessels and the content of human or mouse adipocytes within the graft were measured using species-specific endothelial and adipocyte-specific quantitative real time polymerase chain reaction probes, and histochemistry with mouse and human-specific lectins. Our results show that ad-PADS grafted subcutaneously into nude mice induce robust vascularization from the host, continue to increase in volume over time, express the human adipocyte marker PLIN1 at levels comparable to human AT, and secrete increasing amounts of human adiponectin into the mouse circulation. In contrast, grafts composed of AT fragments obtained by liposuction become less vascularized, develop regions of calcification and decreased content of PLIN1, and secrete lower amounts of adiponectin per unit volume. Enrichment of liposuction tissue with ad-PADS improves vascularization, indicating that ad-PADS may be proangiogenic. Mechanistically, ad-PADS express an extracellular matrix gene signature that includes elements previously associated with small vessel development (COL4A1). Thus, through the formation of a proangiogenic environment, ad-PADS can form functional AT with capacity for long-term survival, and can potentially be used to improve outcomes in reconstructive and regenerative medicine

    Collagen Fiber Regulation in Human Pediatric Aortic Valve Development and Disease

    Congenital aortic valve stenosis (CAVS) affects up to 10% of the world population without medical therapies to treat the disease. New molecular targets are continually being sought that can halt CAVS progression. Collagen deregulation is a hallmark of CAVS yet remains mostly undefined. Here, histological studies were paired with high resolution accurate mass (HRAM) collagen-targeting proteomics to investigate collagen fiber production with collagen regulation associated with human AV development and pediatric end-stage CAVS (pCAVS). Histological studies identified collagen fiber realignment and unique regions of high-density collagen in pCAVS. Proteomic analysis reported specific collagen peptides are modified by hydroxylated prolines (HYP), a post-translational modification critical to stabilizing the collagen triple helix. Quantitative data analysis reported significant regulation of collagen HYP sites across patient categories. Non-collagen type ECM proteins identified (26 of the 44 total proteins) have direct interactions in collagen synthesis, regulation, or modification. Network analysis identified BAMBI (BMP and Activin Membrane Bound Inhibitor) as a potential upstream regulator of the collagen interactome. This is the first study to detail the collagen types and HYP modifications associated with human AV development and pCAVS. We anticipate that this study will inform new therapeutic avenues that inhibit valvular degradation in pCAVS and engineered options for valve replacement
