    Fame! I wanna stream forever! Analysis and Critique of Successful Streamers' Advice to the Next Generation

    This paper examines the practice of large-scale and successful streamers in creating ‘how-to’ videos with advice for smaller streamers and content creators. The goal of this study was to investigate how successful streamers encourage newcomers and/or smaller streamers to “grow” on Twitch, examining such factors as the advice they offer, the evidence that such advice works, and their reasons for sharing such information. More critically, how do their materials invoke rhetorics of meritocracy, and how does the creation of such videos further entrench those creators as successful, able to deploy “streaming capital” to buttress their claims? While the initial focus was on Twitch, this research led to a wider ranging exploration as the advice of successful streamers was to employ other platforms in combination, as the field of live streaming has extended far beyond Twitch itself

    Solid-Liquid two-phase partitioning bioreactors for the treatment of gas.-phase VOCs

    [Abstract] Two-Phase Partitioning Bioreactors (TPPBs) consist of a cell-containing aqueous phase and a separate, biocompatible and immiscible phase that partitions toxic substrates to the cells based on their metabolic demand and on maintaining the thermodynamic equilibrium of the system. TPPBs have traditionally used immiscible liquid organic solvents as the substrate delivery phase, however, one of the limitations of organic solvents is their potential bioavailability as substrates, and therefore these TPPB systems have generally been limited to the use of pure strains of organisms incapable of metabolizing the solvent. We have replaced the organic solvent phase in TPPBs with inert polymers (plastic beads). A TPPB employing styrene-butadiene beads as the sequestering phase was used to treat high step change loadings of BTEX in a contaminated air stream. The presence of the polymers allowed the system to effectively capture the incoming VOCs, buffer the cells from high VOC levels and release the VOCs to the cells for biodegradation. The polymer TPPB system demonstrated substantially higher performance than an aqueous phase bioscrubber and comparable performance to a solvent-aqueous TPPB. Also of great interest was the increase in oxygen transfer provided to the system by the addition of polymer beads, which have significant affinity for oxygen. The presence of polymer beads, which are biocompatible and non-bioavailable, provides a simple and effective means of enhancing the bioremediation of toxic organics present in gas streams, and potentially other phases

    Synergistic research synthesis enabling evidence based practice

    Introduction Evidence based practice requires showing upon what we are basing medical opinions and guidelines, or recognising when evidence is absent that guidance is “expert opinion” and research is required to fill evidence gaps. Aerospace is one of the final medical fields to begin organising a critical summary, adapted periodically, of evidence underpinning operations, and the Aerospace Medicine Systematic Review Group is a new initiative to fill this gap. This group facilitates high quality, transparent synthesis of evidence, to inform operational medical guidelines in best practice, while simultaneously guiding future research by identifying research gaps. The group has (A) facilitated a second review with the European Space Agency Medical Office to inform human Lunar and Martian mission medical considerations and (B) developed and published, open access, new review methods to aid others to undertake aerospace medicine systematic reviews. Methods (A) Electronic databases were searched from the start of records to April 2016. Studies were assessed with the Cochrane risk of bias tool. Effect size analysis was used to assess the effect of various g loading on human biomechanical and cardiopulmonary systems. (B) A new rating scale to appraise technical principles of studies to simulate partial gravity was implemented. Additional method guides for developing questions, protocol drafting, data extracting, quantifying effects and scoring a bed rest study quality were also developed. Results (A) The review identified 43 studies that found partial gravity appears unable to protect against cardiovascular and biomechanical changes. (B) The group designed and developed a website (www.aerospacemed.rehab/systematic-review-group) to provide free access to methods developed by the group and provide links to wider resources. Discussion The systematic review informed medical considerations for future human exploration missions and demonstrates how systematic synthesis of the evidence base more strongly and better informs medical operations than expert opinion, basic reviews or disordered individual studies. Limitations in the current conduct and reporting of aerospace medicine research are also highlighted. Continuing development of review methods, published as open access guides on the group website and working with review teams globally, will help bring synergy to, and enable high quality summary, of the aerospace medicine evidence base

    Phage Hunting at the University of Mary Washington: Genome Annotation of Hari and JackRabbit

    Bacillus thuringiensis subspecies Kurstaki (BTK) is often used as a microbial insecticide for pest control and as a simulant for Bacillus anthracis in biowarfare and bioterrorism studies. Students in 2021 Phage Hunters class at University of Mary Washington isolated nine bacteriophages using the host Bacillus thuringiensis subspecies Kurstaki. Two phages, Hari and Jackrabbit, were sent to SEAPHAGES for sequencing are currently being annotated in the lab during the Spring semester. Hari was found in a soil sample obtained from King George, VA while JackRabbit was isolated from Linton, VA. Both samples were isolated from enriched cultures. Hari has a genome length of 161,978 bp, which auto-annotated with 286 features, and a direct terminal repeat of 2,633 bp. Hari is most similar to DIGNKC, SBP8a and PPIsBest by BLAST. JackRabbit has a genome length of 161,552 bp, which auto-annotated with 288 features, and a direct terminal repeat of 2,821 bp

    Global-scale quantification of mineralization pathways in marine sediments: A reaction-transport modeling approach

    [1] The global-scale quantification of organic carbon (Corg) degradation pathways in marine sediments is difficult to achieve experimentally due to the limited availability of field data. In the present study, a numerical modeling approach is used as an alternative to quantify the major metabolic pathways of Corg oxidation (Cox) and associated fluxes of redox-sensitive species fluxes along a global ocean hypsometry, using the seafloor depth (SFD) as the master variable. The SFD dependency of the model parameters and forcing functions is extracted from existing empirical relationships or from the NOAA World Ocean Atlas. Results are in general agreement with estimates from the literature showing that the relative contribution of aerobic respiration to Cox increases from 80% in deep-sea sediments. Sulfate reduction essentially follows an inversed SFD dependency, the other metabolic pathways (denitrification, Mn and Fe reduction) only adding minor contributions to the global-scale mineralization of Corg. The hypsometric analysis allows the establishment of relationships between the individual terminal electron acceptor (TEA) fluxes across the sediment-water interface and their respective contributions to the Corg decomposition process. On a global average, simulation results indicate that sulfate reduction is the dominant metabolic pathway and accounts for approximately 76% of the total Cox, which is higher than reported so far by other authors. The results also demonstrate the importance of bioirrigation for the assessment of global species fluxes. Especially at shallow SFD most of the TEAs enter the sediments via bioirrigation, which complicates the use of concentration profiles for the determination of total TEA fluxes by molecular diffusion. Furthermore, bioirrigation accounts for major losses of reduced species from the sediment to the water column prohibiting their reoxidation inside the sediment. As a result, the total carbon mineralization rate exceeds the total flux of oxygen into the sediment by a factor of 2 globally

    Drug Adverse Event Detection in Health Plan Data Using the Gamma Poisson Shrinker and Comparison to the Tree-based Scan Statistic

    Background: Drug adverse event (AE) signal detection using the Gamma Poisson Shrinker (GPS) is commonly applied in spontaneous reporting. AE signal detection using large observational health plan databases can expand medication safety surveillance. Methods: Using data from nine health plans, we conducted a pilot study to evaluate the implementation and findings of the GPS approach for two antifungal drugs, terbinafine and itraconazole, and two diabetes drugs, pioglitazone and rosiglitazone. We evaluated 1676 diagnosis codes grouped into 183 different clinical concepts and four levels of granularity. Several signaling thresholds were assessed. GPS results were compared to findings from a companion study using the identical analytic dataset but an alternative statistical method—the tree-based scan statistic (TreeScan). Results: We identified 71 statistical signals across two signaling thresholds and two methods, including closely-related signals of overlapping diagnosis definitions. Initial review found that most signals represented known adverse drug reactions or confounding. About 31% of signals met the highest signaling threshold. Conclusions: The GPS method was successfully applied to observational health plan data in a distributed data environment as a drug safety data mining method. There was substantial concordance between the GPS and TreeScan approaches. Key method implementation decisions relate to defining exposures and outcomes and informed choice of signaling thresholds

    Saving endangered whales at no cost

Drastic overexploitation has driven this large, slow-swimming baleen whale to virtual extinction in Europe, while a small remnant population of ~350 individuals remains on the U.S. and Canadian east coast. Although this species has been protected for 70 years, recovery has been slight and extinction is still looming because of accidental mortality from shipstrikes and fishing gear (Figure 1A,B). Seventy five percent of appropriately photographed whales show evidence of entanglement, predominantly with lobster fishing gear, and this percentage has increased from 52% in the 1980s. At the same time, the U.S. lobster fishery is severely overexploited (the inshore fishing mortalities in the two main U.S. regions are 0.69 and 0.84, while 0.2 achieves maximum yield per recruit). We argue here that this endangered whale species can be protected from entanglement mortality, and the fishery can benefit simultaneously, by a large reduction of lobster traps used; a classic win–win situation.This work was supported by the Lenfest Foundation and NSERC

    Receipt of medications for opioid use disorder among youth engaged in primary care: data from 6 health systems

    PURPOSE: Little is known about prevalence and treatment of OUD among youth engaged in primary care (PC). Medications are the recommended treatment of opioid use disorder (OUD) for adolescents and young adults (youth). This study describes the prevalence of OUD, the prevalence of medication treatment for OUD, and patient characteristics associated with OUD treatment among youth engaged in PC. METHODS: This cross-sectional study includes youth aged 16-25 years engaged in PC. Eligible patients had ≥ 1 PC visit during fiscal years (FY) 2014-2016 in one of 6 health systems across 6 states. Data from electronic health records and insurance claims were used to identify OUD diagnoses, office-based OUD medication treatment, and patient demographic and clinical characteristics in the FY of the first PC visit during the study period. Descriptive analyses were conducted in all youth, and stratified by age (16-17, 18-21, 22-25 years). RESULTS: Among 303,262 eligible youth, 2131 (0.7%) had a documented OUD diagnosis. The prevalence of OUD increased by ascending age groups. About half of youth with OUD had documented depression or anxiety and one third had co-occurring substance use disorders. Receipt of medication for OUD was lowest among youth 16-17 years old (14%) and highest among those aged 22-25 (39%). CONCLUSIONS: In this study of youth engaged in 6 health systems across 6 states, there was low receipt of medication treatment, and high prevalence of other substance use disorders and mental health disorders. These findings indicate an urgent need to increase medication treatment for OUD and to integrate treatment for other substance use and mental health disorders
