    From the Beginning: A Start to End Analysis of a Beginning Teacher Program

    This mixed-methods study evaluated the effectiveness of a Beginning Teacher Support Program in the southern region of a school district. The effects of teacher turnover, national teacher shortages, induction programs, mentoring, administrative support, professional development, and other support offered to beginning teachers were analyzed. The researcher created a survey that was administered to high school beginning teachers, mentor teachers, and principals. A focus group and an interview with a regional beginning teacher coordinator were also conducted to further evaluate the effectiveness of the district’s beginning teacher program. The results of the study concluded that beginning teachers benefitted from participating in the district’s beginning teacher support program. Specifically, beginning teachers reported having a veteran mentor, having mentor teachers provide resources, and collaborating with other teachers as an area of effectiveness. Areas that were not effective included co-teaching with mentors, lesson unit planning, data analysis, and outside professional development. The researcher’s recommendations for further study included providing focus groups for beginning teachers to reflect on the beginning teacher support program, future researchers should be cognizant of time management in terms of collecting data, and a more in-depth study of the role of the site-wide beginning teacher coordinator. Recommendations for the district included to continue with the pairing of veteran teachers or mentors with beginning teachers, recruit more mentor teachers to assist beginning teachers, provide more support for lateral entry teachers, and to match lateral entry veteran teachers with lateral entry beginning teachers

    Circumstellar discs in Galactic centre clusters: Disc-bearing B-type stars in the Quintuplet and Arches clusters

    We investigate the circumstellar disc fraction as determined from L-band excess observations of the young, massive Arches and Quintuplet clusters residing in the central molecular zone of the Milky Way. The Quintuplet cluster was searched for L-band excess sources for the first time. We find a total of 26 excess sources in the Quintuplet cluster and 21 in the Arches cluster, of which 13 are new detections. With the aid of proper motion membership samples, the disc fraction of the Quintuplet cluster was derived for the first time to be 4.0 +/- 0.7%. There is no evidence for a radially varying disc fraction in this cluster. In the case of the Arches cluster, a disc fraction of 9.2 +/- 1.2% approximately out to the cluster's predicted tidal radius, r < 1.5 pc, is observed. This excess fraction is consistent with our previously found disc fraction in the cluster in the radial range 0.3 < r < 0.8 pc. In both clusters, the host star mass range covers late A- to early B-type stars, 2 < M < 15 Msun, as derived from J-band photospheric magnitudes. We discuss the unexpected finding of dusty circumstellar discs in these UV intense environments in the context of primordial disc survival and formation scenarios of secondary discs. We consider the possibility that the L-band excess sources in the Arches and Quintuplet clusters could be the high-mass counterparts to T Tauri pre-transitional discs. As such a scenario requires a long pre-transitional disc lifetime in a UV intense environment, we suggest that mass transfer discs in binary systems are a likely formation mechanism for the B-star discs observed in these starburst clusters.Comment: 47 pages, 22 figures, accepted by A&

    BackgroundDesmoplastic small round cell tumor (DSRCT) is a rare sarcoma in adolescents and young adults. The hallmark of this disease is a EWS-WT1 translocation resulting from apposition of the Ewing's sarcoma (EWS) gene with the Wilms' tumor (WT1) gene. We performed morphoproteomic profiling of DSRCT (EWS-WT1), Ewing's sarcoma (EWS-FLI1) and Wilms' tumor (WT1) to better understand the signaling pathways for selecting future targeted therapies.MethodologyThis pilot study assessed patients with DSRCT, Wilms' tumor and Ewing's sarcoma. Morphoproteomics and immunohistochemical probes were applied to detect: p-mTOR (Ser2448); p-Akt (Ser473); p-ERK1/2 (Thr202/Tyr204); p-STAT3 (Tyr 705); and cell cycle-related analytes along with their negative controls.Principal findingsIn DSRCT the PI3K/Akt/mTOR pathway is constitutively activated by p-Akt (Ser 473) expression in the nuclear compartment of the tumor cells and p-mTOR phosphorylated on Ser 2448, suggesting mTORC2 (rictor+mTOR) as the dominant form. Ewing's sarcoma had upregulated p-Akt and p-mTOR, predominantly mTORC2. In Wilm's tumor, the mTOR pathway is also activated with most tumor cells moderately expressing p-mTOR (Ser 2448) in plasmalemmal and cytoplasmic compartments. This coincides with the constitutive activation of one of the downstream effectors of the mTORC1 signaling pathway, namely p-p70S6K (Thr 389). There was constitutive activation of the Ras/Raf/ERK pathway p-ERK 1/2 (Thr202/Tyr204) expression in the Wilms tumor and metastatic Ewing's sarcoma, but not in the DSRCT.ConclusionMORPHOPROTEOMIC TUMOR ANALYSES REVEALED CONSTITUTIVE ACTIVATION OF THE MTOR PATHWAY AS EVIDENCED BY: (a) expression of phosphorylated (p)-mTOR, p-p70S6K; (b) mTORC 2 in EWS and DSRCT; (c) ERK signaling was seen in the advanced setting indicating these as resistance pathways to IGF1R related therapies. This is the first morphoproteomic study of such pathways in these rare malignancies and may have potential therapeutic implications. Further study using morphoproteomic assessments of these tumors are warranted

    The Arches Cluster: Extended Structure and Tidal Radius

    At a projected distance of ~26 pc from Sgr A*, the Arches cluster provides insight to star formation in the extreme Galactic Center (GC) environment. Despite its importance, many key properties such as the cluster's internal structure and orbital history are not well known. We present an astrometric and photometric study of the outer region of the Arches cluster (R > 6.25") using HST WFC3IR. Using proper motions we calculate membership probabilities for stars down to F153M = 20 mag (~2.5 M_sun) over a 120" x 120" field of view, an area 144 times larger than previous astrometric studies of the cluster. We construct the radial profile of the Arches to a radius of 75" (~3 pc at 8 kpc), which can be well described by a single power law. From this profile we place a 3-sigma lower limit of 2.8 pc on the observed tidal radius, which is larger than the predicted tidal radius (1 - 2.5 pc). Evidence of mass segregation is observed throughout the cluster and no tidal tail structures are apparent along the orbital path. The absence of breaks in the profile suggests that the Arches has not likely experienced its closest approach to the GC between ~0.2 - 1 Myr ago. If accurate, this constraint indicates that the cluster is on a prograde orbit and is located front of the sky plane that intersects Sgr A*.     Harnessing NASA Goddard’s Grey Literature: The Power of a Repository Framework

Permission granted through posted policies on copyright owner’s website or through direct contact with copyright owner.The NASA Goddard Library collaborated with several projects on-Center to create a framework for the development of web-accessible repositories of grey literature. Tools and methods for collaboration were developed through a series of prototypes with a variety of Goddard projects based on the Library’s Digital Asset System (DAS), a repository to describe and provide access to project information including images, videos, web sites, and technical reports. Metadata is a key component of this framework. The Goddard Core Metadata Element Set, an extension of the Dublin Core, is used to describe these resources. Additional elements include project name, project code and instrument name. A taxonomy of controlled subject terms has been developed which can also be extended and tailored for each project. This provides consistent searching across the DAS, while at the same time enhancing the search precision for each project collection when it is accessed as a separate collection in the DAS or through its own web site. To-date, the Library has used the framework with a number of different projects. A commemorative CD with key documents, web sites and oral histories by the project managers was developed for the Hitchhiker Project as it was being disbanded. Metadata from the Swift project library was transformed and made more accessible with pointers to documents in the Swift project library. The Landsat Legacy Project, a joint project with the Landsat Program Science Office, the US Geological Survey, and the NASA History Office, is creating an archive of essential technical-, policy-, and science-related documentation. To gather significant documentation from the over 35-year-multi-agency history of Landsat, the Library added components for external submission of documents for potential inclusion in the new repository and for scanning paper documents. In addition, video histories are being captured from Landsat veterans. Through these projects, the NASA Goddard Library has developed a methodology for collaborating with different kinds of projects. It involves procedures for analyzing the needs of a project and determining how the metadata, taxonomy, and interface might need to change, while remaining consistent with the DAS framework for cross-repository access. Ultimately, the Library plans to extend the effort to other projects. (At any given point in time, Goddard has more than 30 projects in various stages of completion.) As part of Goddard’s knowledge management activities, the DAS provides a framework for sharing grey literature that would otherwise be scattered across independent project libraries. Benefits include the ability to more quickly find and reuse information to decrease project costs, enhance safety and promote innovation

    Sustained delivery of nicotinamide limits cortical injury and improves functional recovery following traumatic brain injury

    Previously, we have demonstrated that nicotinamide (NAM), a neuroprotective soluble B-group vitamin, improves recovery of function following traumatic brain injury (TBI). However, no prior studies have examined whether NAM is beneficial following continuous infusions over 7 days post-TBI. The purpose of this study was to investigate the preclinical efficacy of NAM treatment as it might be delivered clinically; over several days by slow infusion. Rats were prepared with either unilateral controlled cortical impact (CCI) injuries or sham procedures and divided into three groups: CCI-NAM, CCI-vehicle and sham. Thirty minutes following CCI, Alzet osmotic mini-pumps were implanted subcutaneously. NAM was delivered at a rate of 50 mg/kg/day for 7 days immediately post-CCI. On day 7 following injury, the pumps were removed and blood draws were collected for serum NAM and nicotinamide adenine dinucleotide (NAD+) analyses. Starting on day 2 post-CCI, animals were tested on a battery of sensorimotor tests (bilateral tactile adhesive removal, locomotor placing and limb-use asymmetry). Continuous infusion of NAM resulted in a significant serum elevation in NAM, but not NAD+. Statistical analyses of the tactile removal and locomotor placing data revealed that continuous administration of NAM significantly reduced the initial magnitude of the injury deficit and improved overall recovery compared to the vehicle-treated animals. NAM treatment also significantly decreased limb-use asymmetries compared to vehicle-treated animals. The overall extent of the cortical damage was also reduced by NAM treatment. No detrimental effects were seen following continuous infusion. The present results suggest that NAM delivered via a clinically relevant therapeutic regimen may truncate behavioral damage following TBI. Thus our results offer strong support for translation into the clinical population

    The Quintuplet Cluster: Extended Structure and Tidal Radius

    The Quintuplet star cluster is one of only three known young (<10<10 Myr) massive (M >104>10^4 M_\odot) clusters within 100\sim100 pc of the Galactic Center. In order to explore star cluster formation and evolution in this extreme environment, we analyze the Quintuplet's dynamical structure. Using the HST WFC3-IR instrument, we take astrometric and photometric observations of the Quintuplet covering a 120×120120''\times120'' field-of-view, which is 1919 times larger than those of previous proper motion studies of the Quintuplet. We generate a catalog of the Quintuplet region with multi-band, near-infrared photometry, proper motions, and cluster membership probabilities for 10,54310,543 stars. We present the radial density profile of 715715 candidate Quintuplet cluster members with M4.7M\gtrsim4.7 M_\odot out to 3.23.2 pc from the cluster center. A 3σ3\sigma lower limit of 33 pc is placed on the tidal radius, indicating the lack of a tidal truncation within this radius range. Only weak evidence for mass segregation is found, in contrast to the strong mass segregation found in the Arches cluster, a second and slightly younger massive cluster near the Galactic Center. It is possible that tidal stripping hampers a mass segregation signature, though we find no evidence of spatial asymmetry. Assuming that the Arches and Quintuplet formed with comparable extent, our measurement of the Quintuplet's comparatively large core radius of 0.620.10+0.100.62^{+0.10}_{-0.10} pc provides strong empirical evidence that young massive clusters in the Galactic Center dissolve on a several Myr timescale.Comment: 25 pages (21-page main text, 4-page appendix), 18 figures, submitted to Ap

    Marginal States in Mean Field Glasses

    We study mean field systems whose free energy landscape is dominated by marginally stable states. We review and develop various techniques to describe such states, elucidating their physical meaning and the interrelation between them. In particular, we give a physical interpretation of the two-group replica symmetry breaking scheme and confirm it by establishing the relation to the cavity method and to the counting of solutions of the Thouless-Anderson-Palmer equations. We show how these methods all incorporate the presence of a soft mode in the free energy landscape and interpret the occurring order parameter functions in terms of correlations between the soft mode and the local magnetizations. The general formalism is applied to the prototypical case of the Sherrington-Kirkpatrick-model where we re-examine the physical properties of marginal states under a new perspective.Comment: 27 pages, 8 figure

    Insulin clearance and the incidence of type 2 diabetes in Hispanics and African Americans: the IRAS Family Study.

    ObjectiveWe aimed to identify factors that are independently associated with the metabolic clearance rate of insulin (MCRI) and to examine the association of MCRI with incident type 2 diabetes in nondiabetic Hispanics and African Americans.Research design and methodsWe investigated 1,116 participants in the Insulin Resistance Atherosclerosis Study (IRAS) Family Study with baseline examinations from 2000 to 2002 and follow-up examinations from 2005 to 2006. Insulin sensitivity (S(I)), acute insulin response (AIR), and MCRI were determined at baseline from frequently sampled intravenous glucose tolerance tests. MCRI was calculated as the ratio of the insulin dose over the incremental area under the curve of insulin. Incident diabetes was defined as fasting glucose ≥126 mg/dL or antidiabetic medication use by self-report.ResultsWe observed that S(I) and HDL cholesterol were independent positive correlates of MCRI, whereas fasting insulin, fasting glucose, subcutaneous adipose tissue, visceral adipose tissue, and AIR were independent negative correlates (all P &lt; 0.05) at baseline. After 5 years of follow-up, 71 (6.4%) participants developed type 2 diabetes. Lower MCRI was associated with a higher risk of incident diabetes after adjusting for demographics, lifestyle factors, HDL cholesterol, indexes of obesity and adiposity, and insulin secretion (odds ratio 2.01 [95% CI 1.30-3.10], P = 0.0064, per one-SD decrease in loge-transformed MCRI).ConclusionsOur data showed that lower MCRI predicts the incidence of type 2 diabetes

    Oxidative Stress Protection by Canary Seed (Phalaris canariensis L.) Peptides in Caco-2 Cells and Caenorhabditis elegans

    During oxidative stress, degenerative diseases such as atherosclerosis, Alzheimer’s, and certain cancers are likely to develop. Recent research on canary seed (Phalaris canariensis) peptides has demonstrated the high in vitro antioxidant potential. Thus, this study aimed to assess the cellular and in vivo antioxidant capacity of a low-molecular-weight (\u3c3 kDa) canary seed peptide fraction (CSPF) using Caco-2 cells and the Caenorhabditis elegans model. The results show that the CSPF had no cytotoxicity effect on Caco-2 cells at any tested concentration (0.3–2.5 mg/mL). Additionally, the cellular antioxidant activity (CAA) of the CSPF was concentration-dependent, and the highest activity achieved was 80% by the CSPF at 2.5 mg/mL. Similarly, incubation with the CSPF significantly mitigated the acute and chronic oxidative damage, extending the lifespan of the nematodes by 88 and 61%, respectively. Furthermore, it was demonstrated that the CSPF reduced the accumulation of reactive oxygen species (ROS) to safe levels after sub-lethal doses of pro-oxidant paraquat. Quantitative real-time PCR revealed that the CSPF increased the expression of oxidative-stress-response-related gene GST-4. Overall, these results show that the CSPFs relied on GST-4 upregulation and scavenging of free radicals to confer oxidative stress protection and suggest that a CSPF can be used as a natural antioxidant in foods for health applications
