    Cathode Assessment for Maximizing Current Generation in Microbial Fuel Cells Utilizing Bioethanol Effluent as Substrate

    Implementation of microbial fuel cells (MFCs) for electricity production requires effective current generation from waste products via robust cathode reduction. Three cathode types using dissolved oxygen cathodes (DOCs), ferricyanide cathodes (FeCs) and air cathodes (AiCs) were therefore assessed using bioethanol effluent, containing 20.5 g/L xylose, 1.8 g/L arabinose and 2.5 g/L propionic acid. In each set-up the anode and cathode had an electrode surface area of 88 cm2, which was used for calculation of the current density. Electricity generation was evaluated by quantifying current responses to substrate loading rates and external resistance. At the lowest external resistance of 27 Ω and highest substrate loading rate of 2 g chemical oxygen demand (COD) per L·day, FeC-MFC generated highest average current density (1630 mA/m2) followed by AiC-MFC (802 mA/m2) and DOC-MFC (184 mA/m2). Electrochemical impedance spectroscopy (EIS) was used to determine the impedance of the cathodes. It was thereby confirmed that the FeC-MFC produced the highest current density with the lowest internal resistance for the cathode. However, in a setup using bioethanol effluent, the AiC-MFC was concluded to be the most sustainable option since it does not require ferricyanide. The data offer a new add-on option to the straw biorefinery by using bioethanol effluent for microbial electricity production

    Certain intracellular bacteria use the host cell cytosol as the replicative niche. Although it has been hypothesized that the successful exploitation of this compartment requires a unique metabolic adaptation, supportive evidence is lacking. For Francisella tularensis, many genes of the Francisella pathogenicity island (FPI) are essential for intracellular growth, and therefore, FPI mutants are useful tools for understanding the prerequisites of intracytosolic replication. We compared the growth of bacteria taken up by phagocytic or nonphagocytic cells with that of bacteria microinjected directly into the host cytosol, using the live vaccine strain (LVS) of F. tularensis; five selected FPI mutants thereof, i.e., Delta iglA, Delta iglC, Delta iglG, Delta iglI, and Delta pdpE strains; and Listeria monocytogenes. After uptake in bone marrow-derived macrophages (BMDM), ASC(-/-) BMDM, MyD88(-/-) BMDM, J774 cells, or HeLa cells, LVS, Delta pdpE and Delta iglG mutants, and L. monocytogenes replicated efficiently in all five cell types, whereas the Delta iglA and Delta iglC mutants showed no replication. After microinjection, all 7 strains showed effective replication in J774 macrophages, ASC(-/-) BMDM, and HeLa cells. In contrast to the rapid replication in other cell types, L. monocytogenes showed no replication in MyD88(-/-) BMDM and LVS showed no replication in either BMDM or MyD88(-/-) BMDM after microinjection. Our data suggest that the mechanisms of bacterial uptake as well as the permissiveness of the cytosolic compartment per se are important factors for the intracytosolic replication.     A method for in-field railhead crack detection using digital image correlation

    Railway infrastructure managers must decide when and how to maintain rails. However, they often have insufficient information about railhead cracks. Therefore, we propose a new method for rail crack detection using a train-mounted digital image correlation (DIC) camera system. The measurement train\u27s weight cause rail bending, allowing the DIC to measure strain concentrations caused by surface-breaking cracks. In this study, we evaluate the method under laboratory conditions. The detected cracks correlate to the actual crack network in the analysed rail field sample. Furthermore, finite element simulations show the method\u27s high sensitivity to crack depths. Existing methods, such as ultra-sonic and eddy-current, produce damage severity indications. The proposed method complements these techniques by providing a discrete description of the surface-breaking cracks and their depth. This information enables infrastructure managers to optimize rail maintenance. Additionally, such detailed measurements can be valuable for research in railhead damage evolution

    Dynamical low-noise microwave source for cold atom experiments

    The generation and manipulation of ultracold atomic ensembles in the quantum regime require the application of dynamically controllable microwave fields with ultra-low noise performance. Here, we present a low-phase-noise microwave source with two independently controllable output paths. Both paths generate frequencies in the range of 6.8356.835\,GHz ±\pm 2525\,MHz for hyperfine transitions in 87^{87}Rb. The presented microwave source combines two commercially available frequency synthesizers: an ultra-low-noise oscillator at 77\,GHz and a direct digital synthesizer for radiofrequencies. We demonstrate a low integrated phase noise of 580μ580\,\murad in the range of 1010\,Hz to 100100\,kHz and fast updates of frequency, amplitude and phase in sub-μ\mus time scales. The highly dynamic control enables the generation of shaped pulse forms and the deployment of composite pulses to suppress the influence of various noise sources.Comment: The following article has been submitted to Review of Scientific Instruments.     Presolar Diamond in Meteorites

    Presolar diamond, the carrier of the isotopically anomalous Xe component Xe-HL, was the first mineral type of presolar dust that was isolated from meteorites. The excesses in the light, p-process only isotopes 124Xe and 126Xe, and in the heavy, r-process only isotopes 134Xe and 136Xe relative to the solar ratios indicate that Xe-HL was produced in supernovae: they are the only stellar source where these two processes are believed to take place. Although these processes occur in supernovae, their physical conditions and timeframes are completely different. Yet the excesses are always correlated in diamond separates from meteorites. Furthermore, the p-process 124Xe/126Xe inferred from Xe-L and the r-process 134Xe/136Xe from Xe-H do not agree with the p-process and r-process ratios derived from the solar system abundance, and the inferred p-process ratio does not agree with those predicted from stellar models. The 'rapid separation scenario', where the separation of Xe and its radiogenic precursors Te and I takes place at the very early stage (7900 sec after the end of the r-process), has been proposed to explain Xe-H. Alternatively, mixing of 20% of material that experienced neutron burst and 80% of solar material can reproduce the pattern of Xe-H, although Xe-L is not accounted for with this scenario

    Effects of forest management practices in clearings on breeding performance of the Red-backed Shrike (Lanius collurio)

    The structure and quality of breeding habitats play an essential role for breeding success of individuals and thus, the viability of populations. Knowledge on habitat requirements can guide management practises to ensure optimal conditions for breeding. In this study, we investigate the effect of different habitat features and forest management strategies on the breeding performance of Red-backed Shrikes (Lanius collurio) breeding in Gribskov forest, Denmark. We found that the availability of available perches for hunting is the main predictor having a positive effect on number of breeding pairs and fledglings. It was also positively associated with the number of years that the clearing has been occupied by breeding pairs. Furthermore, clearings with grazing animals contained breeding pairs for more years than clearings without grazing, suggesting improved foraging conditions in clearings with low vegetation. Our results provide direct applications for forestry management for improving the conservation status of this species
