    High anti-SARS-CoV-2 antibody seroconversion rates before the second wave in Manaus, Brazil, and the protective effect of social behaviour measures: results from the prospective DETECTCoV-19 cohort

    Background: The city of Manaus, Brazil, has seen two collapses of the health system due to the COVID-19 pandemic. We report anti-SARS-CoV-2 nucleocapsid IgG antibody seroconversion rates and associated risk factors in Manaus residents before the second wave of the epidemic in Brazil. Methods: A convenience sample of adult (aged ≥18 years) residents of Manaus was recruited through online and university website advertising into the DETECTCoV-19 study cohort. The current analysis of seroconversion included a subgroup of DETECTCoV-19 participants who had at least two serum sample collections separated by at least 4 weeks between Aug 19 and Oct 2, 2020 (visit 1), and Oct 19 and Nov 27, 2020 (visit 2). Those who reported (or had no data on) having a COVID-19 diagnosis before visit 1, and who were positive for anti-SARS-CoV-2 nucleocapsid IgG antibodies at visit 1 were excluded. Using an in-house ELISA, the reactivity index (RI; calculated as the optical density ratio of the sample to the negative control) for serum anti-SARS-CoV-2 nucleocapsid IgG antibodies was measured at both visits. We calculated the incidence of seroconversion (defined as RI values ≤1·5 at visit 1 and ≥1·5 at visit 2, and a ratio >2 between the visit 2 and visit 1 RI values) during the study period, as well as incidence rate ratios (IRRs) through cluster-corrected and adjusted Poisson regression models to analyse associations between seroconversion and variables related to sociodemographic characteristics, health access, comorbidities, COVID-19 exposure, protective behaviours, and symptoms. Findings: 2496 DETECTCoV-19 cohort participants returned for a follow-up visit between Oct 19 and Nov 27, 2020, of whom 204 reported having COVID-19 before the first visit and 24 had no data regarding previous disease status. 559 participants were seropositive for anti-SARS-CoV-2 nucleocapsid IgG antibodies at baseline. Of the remaining 1709 participants who were seronegative at baseline, 71 did not meet the criteria for seroconversion and were excluded from the analyses. Among the remaining 1638 participants who were seronegative at baseline, 214 showed seroconversion at visit 2. The seroconversion incidence was 13·06% (95% CI 11·52–14·79) overall and 6·78% (5·61–8·10) for symptomatic seroconversion, over a median follow-up period of 57 days (IQR 54–61). 48·1% of seroconversion events were estimated to be asymptomatic. The sample had higher proportions of affluent and higher-educated people than those reported for the Manaus city population. In the fully adjusted and corrected model, risk factors for seroconversion before visit 2 were having a COVID-19 case in the household (IRR 1·49 [95% CI 1·21–1·83]), not wearing a mask during contact with a person with COVID-19 (1·25 [1·09–1·45]), relaxation of physical distancing (1·31 [1·05–1·64]), and having flu-like symptoms (1·79 [1·23–2·59]) or a COVID-19 diagnosis (3·57 [2·27–5·63]) between the first and second visits, whereas working remotely was associated with lower incidence (0·74 [0·56–0·97]). Interpretation: An intense infection transmission period preceded the second wave of COVID-19 in Manaus. Several modifiable behaviours increased the risk of seroconversion, including non-compliance with non-pharmaceutical interventions measures such as not wearing a mask during contact, relaxation of protective measures, and non-remote working. Increased testing in high-transmission areas is needed to provide timely information about ongoing transmission and aid appropriate implementation of transmission mitigation measures.     O perfil semiológico do paciente portador de hemorragia digestiva alta

    OBJETIVO: O seguinte estudo objetivou descrever a semiologia do paciente portador de hemorragia digestiva alta, considerando como determinante na avaliação de potencias focos hemorrágicos. METODOLOGIA: Foram realizadas buscas nas plataformas do SciELO, LILACS, PubMed, Scopus e Google Scholar,utilizando os descritores gastrointestinal bleeding, peptic ulcerous disease e varicose hemorrhage, sendo identificados 35 estudos, dos quais foram incluídos 13 artigos completos. Desses estudos, 5 avaliaram as principais etiologias, 2 o surgimento de novos testes diagnósticos, 2 analisaram os aspectos epidemiológicos e 1 a sintomatologia apresentada pelo acometimento da hemorragia digestiva alta. Observou-se inicialmente a abundâncias de informações conceituais sobre o sangramento, como um transtorno clínico comum, acompanhada de inúmeras manifestações, considerando que o foco hemorrágico pode ocorrer em qualquer porção do trato gastrointestinal. Neste estudo, todas as publicações eleitas apresentaram o quadro semiológico composto por algia abdominal, indícios de choque hipovolêmico e taquicardia, alguns exibiram quedas abruptas da pressão arterial, odinofagia, êmese, náuseas e estado ictérico. Os pacientes implicados, cronicamente, já manifestaram ocorrências prévias, devido ao caráter recidivante torna-se essencial investigar a existência de varizes, fístula aorto-entérica, angiodisplasia e doença ulcerosa. CONCLUSÃO: Elucida-se que a hemorragia digestiva alta representa a principal causa de sangramento do trato gastrointestinal, majoritamente manifesta-se como hematêmese ou melena e cursam com o quadro sintomatológico que auxilia na avaliação da gravidade deste e o embasamento de potenciais focos de sangramento e que contribuam para disseminação de informações e intervenções futuras

    O objetivo deste trabalho foi avaliar a atividade fungicida in vitro do óleo essencial das folhas de Piper hispidinervum sobre Bipolaris sorokiniana, Fusarium oxysporum e Colletotrichum gloeosporioides. Para os ensaios biológicos, empregou-se o teste bioanalítico in vitro utilizando as concentrações de 100, 200, 500, 1000, 1500 e 2000 µg.mL-1 do óleo essencial. Estas foram incorporadas no meio de cultura BDA (batata-dextrose-ágar) para avaliação do crescimento ou inibição micelial. O delineamento estatístico utilizado foi o inteiramente casualizado, com quatro repetições. Na concentração de 200 µg.mL-1, observou-se uma inibição total do fitopatógeno Bipolaris sorokiniana enquanto que, para o Fusarium oxysporum e o Colletotrichum gloeosporioides esta ocorreu na concentração de 1000 µg.mL-1.The objective of this study was to evaluate the in vitro antifungal activity of the essential oil of the leaves of Piper hispidinervum against Bipolaris sorokiniana, Fusarium oxysporum and Colletotrichum gloeosporioides. For the biological tests, using the bioanalitic test in vitro the concentrations of 100, 200, 500, 1000, 1500 and 2000 µg.mL-1 the essential oil. This were incorporated into PDA (potato dextrose agar) medium in order to evaluate fungal mycelial growth or inhibition. The statistic design used was completely randomized, with four replicates. In the concentration of 200 µg.mL-1, observed inhibited complete the phytopathogens Bipolaris sorokiniana while the Fusarium oxysporum and Colletotrichum gloeosporioides this is occurred in the concentration of 1000 µg.mL-1

    Em função da interferência do milho em outras plantas, objetivou-se verificar a presença e a concentração de benzoxazolinona no milho por meio de extração aquosa obtida de material vegetal das cultivares AG-1051, C-333 e C-435. A massa fresca das amostras foi colocada em água destilada e deixada em repouso durante quatro horas. O extrato foi filtrado e particionado em solventes com diferentes polaridades. O extrato etéreo desidratado foi filtrado, concentrado em evaporador rotatório e levado à estufa. Este foi purificado por cromatografia de camada delgada. Recristalizou-se em hexano, e os cristais obtidos foram submetidos à análise em espectrometria no Infravermelho. As bandas presentes no produto padrão também foram caracterizadas nas amostras das cultivares de milho testadas. As amostras dos extratos foram qualificadas e quantificadas por espectrofotometria de ultravioleta. Pelos resultados, infere-se que a planta de milho possuiem seu metabolismo a benzoxazolinona, na seguinte ordem decrescente: AG-1051 29 mg g-1, C-333 24 mg g-1 e C-435 21 mg g-1 de BOA.Due to the interference of maize on other plants, the presence and concentration of benzoxazolinone in this particular plant (maize) wasdetermined via the aqueous extract of three varieties of maize (AG-1051, C-333 and C-435). The fresh sample material was allowed to stand in distilled water for four hours. The extract was filtered and partitioned with solvents of varying polarities. The dried etherextract was filtered, concentrated on a rotary evaporator and dried in an oven. The residue was purified by thin layer chromatography and recrystallized from hexane. The crystals were analyzed by infrared spectroscopy. The bands present in the spectrum of the referencesubstance were also observed in the spectra of the samples obtained from the three varieties of maize studied. The extract samples were characterized and quantified by UV spectroscopy. The results indicated that the three varieties of maize possessed benzoxazolinone in their metabolism in the decreasing order AG-1051 29 mg g-1 > C-33324 mg g-1 > C-435 21 mg g-1 of BOA

    Objetivou-se avaliar o efeito inibitório dos óleos essenciais de Cymbopogon citratus (capim-limão), Origanum vulgare (orégano) e Syzygium aromaticum (cravo-da-índia); os experimentos foram realizados com as bactérias Staphylococcus aureus e Escherichia coli, importantes patógenos causadores de contaminações em queijos e outros alimentos. Para quantificação e identificação dos constituintes químicos dos óleos, utilizou-se um cromatógrafo gasoso acoplado a um espectrômetro de massa. Os resultados dos testes in vitro, os óleos essenciais de C. citratus, O. vulgare e S. aromaticum promoveram efeito inibitório sobre as bactérias S. aureus e E. coli, porém o S. aromaticum apresentou melhor formação de halo de inibição nas menores concentrações. Para o efeito sinergístico dos óleos sobre as bactérias não foram observadas diferenças quando comparados com o efeito individual dos mesmos. Foi possível verificar que os óleos possuem efeito inibitório sobre os microrganismos estudados, sendo, portanto uma alternativa no controle microbiológico de alimentos.With the aim of evaluating the inhinibitory effect of the Cymbopogon citratus (lemon grass), Origanun vulgare (oregano) and Syzygium aromaticum (clove), experiments were performed in laboratories, with the Staphylococcus aureus, Escherichia coli bacteria, important pathogenics which contaminate cheese and other foods. For the oil chemical constitutions quantification and identification, the gas chromatography attached mass spectrometry to a was used. In the results in vitro test, the C. citratus, O. vulgare and S. aromaticum essential oils presented antimicrobian activity over S. aureus and E. coli bacteria, however, S. aromaticum presented better formation of inhibition halo in smaller concentrations. For the oil synergetic effect over bacteria and fungi, no significant difference was noticed when compared to their own individual effect. It was possible to verify that the oils had inhibitory effect over the microorganisms studied, being, therefore an alternate option in food microbiological control

    A maioria das plantas são resistentes aos diferentes patógenos, e essa resistência pode estar relacionada à existência de compostos fungistáticos naturalmente produzidos. Com o presente trabalho, avaliou-se a atividade fungitóxica de óleos essenciais de eucaliptos. Os óleos foram obtidos de folhas dos eucaliptos mediante arraste a vapor de água, utilizando o aparelho de Clevenger modificado. Nos ensaios biológicos, foram empregados os fitopatógenos Fusarium oxysporum, Botrytis cinerea e Bipolaris sorokiniana. O crescimento dos microorganismos na presença de diferentes concentrações de óleo (5, 50 e 500 mg/Kg), usando os meios de cultura BDA (Batata-Dextrose-Ágar) e PCA (Batata-Cenoura-Ágar), foi avaliado. Nas concentrações de 500 mg/Kg dos óleos, foram observadas inibições significativas no crescimento micelial das espécies fúngicas, após período de 7 dias. No entanto, o óleo essencial de Eucalyptus urophylla foi o que apresentou maior ação fungitóxica, que foi atribuída à presença do composto denominado globulol, ausente no E.camaldulensis e no E. citriodora.<br>Most plants are resistant to different pathogens and this resistance may be related to the existence of naturally produced fungistatic components. The present work evaluated the fungitoxic activity of essential oils from three eucalyptus species. The essential oils were obtained from eucalyptus leaves by steam distillation using a modified Clevenger apparatus. The phytopatogens Fusarium oxysporum, Botrytis cinerea and Bipolaris sorokiniana were employed in the biological tests. The growth of fungi in the presence of different concentrations of oil (5, 50, and 500 mg/kg), using BDA (Potato-Dextrose-agar) and PCA (Potato-Carrot-agar) culture media were evaluated. Significant inhibition of the micelial growth of the fungal species was observed at the concentration of 500 mg/kg of oil after a period of seven days. The essential oil of Eucalyptus urophylla showed the greatest fungitoxic activity. This fact was attributed to the presence of the compound globulol, which was absent in the oils obtained from E. camaldulensis and E. citriodora
