    Spectral actinic flux in the lower troposphere: measurement and 1-D simulations for cloudless, broken cloud and overcast situations

    In September 2002, the first INSPECTRO campaign to study the influence of clouds on the spectral actinic flux in the lower troposphere was carried out in East Anglia, England. Measurements of the actinic flux, the irradiance and aerosol and cloud properties were made from four ground stations and by aircraft. The radiation measurements were modelled using the uvspec model and ancillary data. For cloudless conditions, the measurements of the actinic flux were reproduced by 1-D radiative transfer modelling within the measurement and model uncertainties of about ±10%. For overcast days, the ground-based and aircraft radiation measurements and the cloud microphysical property measurements are consistent within the framework of 1-D radiative transfer and within experimental uncertainties. Furthermore, the actinic flux is increased by between 60-100% above the cloud when compared to a cloudless sky, with the largest increase for the optically thickest cloud. Correspondingly, the below cloud actinic flux is decreased by about 55-65%. Just below the cloud top, the downwelling actinic flux has a maximum that is seen in both the measurements and the model results. For broken clouds the traditional cloud fraction approximation is not able to simultaneously reproduce the measured above-cloud enhancement and below-cloud reduction in the actinic flux

    Assessment and verification of mean effective diameter of internal channels fabricated by laser powder bed fusion

    Channels with small diameters, used for example in conformal cooling, can nowadays be fabricated by laser powder bed fusion. Measurements of the mean effective diameter and surface topography are important to quantify the flow capabilities through the channel. In this paper, a new method using X-ray computed tomography is developed to obtain the mean effective diameter and mean surface topography height. The developed method is verified by determining the mean effective diameter using incompressible turbulent fluid flow simulations, whereupon the determined mean surface topography height is fed as an input to the simulation. The method is proved to offer a non-destructive and relatively fast approach to measure the mean effective diameter and mean surface topography height in circular channels

    We have evaluated mitochondrial (mt) DNA content in CD4 and CD8 peripheral blood lymphocytes (PBLs) from HIV-infected patients taking highly active antiretroviral therapy (HAART) who display different types of adipose tissue alterations. A cross-sectional study was performed in a total of 23 patients with lipodystrophy (LD): nine patients with fat accumulation, six patients with fat loss, eight patients with combined form, who were compared to 11 individuals infected by HIV without LD (HIV-positive) and 10 seronegative controls (CTRL). PBLs were obtained by standard methods, that is, gradient density centrifugation on Ficoll, and CD4 or CD8 cells were positively isolated by magnetic sorting to eliminate the contamination of platelets. mtDNA content was then measured by an original assay based upon real-time PCR. mtDNA content was significantly increased in CD4 T cells from patients with LD, while no differences were present between CD4 and CD8 cells from HIV-positive and CTRL individuals. Nor were any differences found when comparing LD or HIV-positive patients treated with stavudine or zidovudine, or taking D-drugs or non D-drugs. Patients with fat accumulation had significantly higher mtDNA content compared to HIV-positive and CTRL, this phenomenon regarding both CD4 and CD8 PBLs. Considering all HIV-positive patients (including LD), mtDNA content showed a significant, positive correlation with cholesterolaemia but not with triglyceridaemia and glycaemia. Relatively high mtDNA content in LD patients, as well as the correlation between mtDNA content and cholesterol in all HIV-positive subjects, suggest the involvement of mitochondria in such a pathology. However, further studies are needed to confirm these initial observations and ascertain whether the quantification of mtDNA in PBL is a useful and reliable marker to investigate and monitor HAART-related changes in fat distribution

    Non-Destructive Assessment of the Functional Diameter and Hydrodynamic Roughness of Additively Manufactured Channels

    Metal additive manufacturing, particularly laser powder bed fusion, is increasingly used in the gas turbine industry for the fabrication of channels with small diameters for conformal cooling and flow passage applications. A critical challenge in this context lies in evaluating aspects such as the geometrical and hydraulic diameters, the effective area and the roughness on the internal surface of the channel that affects the flow functionality. This paper proposes a new method to evaluate the geometrical and functional equivalent diameters, i.e., the hydraulic diameter of cylindrical channels and the mean surface topography height on the internal channel surface, using X-ray computed tomography. The developed methods were validated with experimental flow tests, considering the mean surface topography height to be equivalent to the hydrodynamic sand grain roughness, thereby determining the hydraulic diameter and the associated effective area. The method is a much faster approach to determining the available hydraulic diameter compared to flow tests and offers the possibility of evaluating the internal surface characteristics, with discrepancies between the two approaches being less than +/- 3%
