    Sensitivity of the soil-borne pathogen Phytophthora agathidicida, the causal agent of kauri dieback, to the anti-oomycete fungicides ethaboxam, fluopicolide, mandipropamid, and oxathiapiprolin

    The oomycete Phytophthora agathidicida is the causal agent of kauri dieback, which threatens the survival of endemic kauri (Agathis australis) forests in Aotearoa|New Zealand. Current chemical control of P. agathidicida involves the application of either a mixture of halogenated tertiary amines or phosphite salts with some success, but neither treatment cures the disease. Recently, four anti-oomycete fungicides, all with different modes of action, have become commercially available. Here, we determined the inhibition potential of these fungicides on three P. agathidicida isolates, using agar dilution assays. The average concentration required to inhibit mycelial growth by 50% (EC₅₀) for ethaboxam, fluopicolide, and mandipropamid was 0.0916, 0.372, and 0.0196 µg/mL, respectively. Inhibition of P. agathidicida mycelia by oxathiapiprolin and its commercial formulation, Zorvec® Enicade®, was 0.000152 and 0.000309 µg/mL, respectively. Based on the EC₅₀ values reported in this study, these fungicides are the most effective inhibitors of P. agathidicida mycelia when compared to previously screened fungicides, natural products, and plant extracts. Thus, their performance in this initial screening supports further research into their potential use as a kauri dieback management tool

    Concussion Return to Play Protocols Effect on Athletes\u27 Reporting of Symptoms

    When an athlete suffers a concussion, the standard return to play (RTP) guidelines are that they must sit out a minimum of a week as they go through a slow progression of exertional and neurocognitive tests once they are asymptomatic. This is a great improvement to concussion treatment in the past where athletes were often dangerously returned. However, because of this “blanket” 7-day minimum return policy, we believe the current RTP guidelines are potentially leading athletes to hide their symptoms more so they do not have to sit out. Fifty-three athletes were surveyed from soccer, basketball, lacrosse, and football teams from 3 universities in Southwest Ohio (ages 18-22, 34=male, 19=female). The survey contained questions about their demographics, concussion symptom knowledge, concussion history, potential reporting of concussions, and opinion on their university’s return to play policy. The results showed that 40% of athletes would be willing to hide their symptoms of a mild concussion and 21% believe that their university’s return to play policy affects this decision. While the results do not completely confirm our hypothesis, we still believe that the numbers we received in our study are enough to warrant a consideration of the current “blanket statement” return to play policy in favor of a very similar but more individualized version

    BACKGROUND: Uptake of cervical cancer screening in the United Kingdom (UK) is falling year on year, and a more sophisticated understanding of non-participation may help design interventions to reverse this trend. This study ascertained the prevalence of different non-participant types using the Precaution Adoption Process Model (PAPM). METHODS: Home-based computer-assisted interviews were carried out with 3113 screening-eligible women in Britain. Survey items assessed self-reported screening uptake and intention to attend in future. Responses to these items were used to classify women into one of five different types of non-participants. RESULTS: Of 793 non-participants, 28% were unaware of screening, 15% had decided not to attend and 51% were intending to have screening but were currently overdue. Younger women were more likely to be unaware of screening or to intend to be screened, while older women were more likely to have decided not to be screened. Women from ethnic minority backgrounds were more likely to be unaware of screening than white women. Being in a lower social grade was associated with increased odds of all three types of non-participation. CONCLUSION: The majority of cervical cancer screening non-participants are not making an active decision not to attend but rather are either unaware or unable to act. There are clear sociodemographic differences between non-participant types, which could be used to identify where tailored interventions may be best targeted

    The Alliance for System Safety of UAS through Research Excellence (ASSURE) conducted research focused on minimum pilot procedures and operational practices used by unmanned aircraft systems (UAS) operators today for the purpose of developing recommendations. This research recommends four pilot and 46 operational minimum procedures to operate a civil single-engine, fixed-wing, single-pilot-configured UAS flying in beyond visual line-of-sight (BVLOS) conditions. These recommendations are anticipated to support potential future aircrew procedure requirements for UAS larger than 55 lbs. operating in the National Airspace System (NAS). These procedures were validated using representative Control Stations in simulated environments

    Public understanding of the purpose of cancer screening: A population-based survey

    Objectives In examining informed choice in cancer screening, we investigated public awareness that some screening programmes aim to prevent cancer, while others seek to detect cancer at an early stage. Methods A population-based survey of adults aged 50–70 in England (n = 1433), including data on demographic characteristics and screening experience. Participants were asked to select the main purpose of cervical, breast, and colorectal cancer screening (both faecal occult blood testing and flexible sigmoidoscopy). Results Across all four screening programmes, most people thought the main aim was to catch cancer early (71–78%). Only 18 and 14% knew that cervical screening and flexible sigmoidoscopy, respectively, are primarily preventive. Knowledge of the preventive aspect of these two programmes was low across the board, with few demographic patterns. By contrast, 78 and 73% of the sample were aware that breast screening and the faecal occult blood test, respectively, predominantly aim to detect cancer early. For these programmes, accurate knowledge was socially graded, lower in ethnic minority groups, and positively associated with previous participation in the programmes. Conclusions Our findings suggest that although awareness of the purpose of early detection screening is high, awareness that screening can prevent cancer is low across all demographic groups. Understanding the purpose of screening is a key aspect of informed choice but despite current communication strategies highlighting these differences, people do not seem to have a nuanced understanding of these differing aims. Our findings may be indicative of a broader public scepticism about the preventability of cancer

    Epidermal Renewal after Treatment of Primary Canine Hypothyroidism using Levothyroxine

    Background: Primary canine hypothyroidism, an endocrine disorder that causes imbalances in the hypothalamus-pituitarythyroid axis, is a common cause of endocrine dermatoses, which frequently presents with opaque dry brittle hair. Tissue changes are also visible, such as myxedema, hyperkeratosis, epidermal atrophy, alopecia, and others. This paper describes the skin changes caused by primary hypothyroidism in a female dog before and after treatment with levothyroxine.Case: This case study involved a 7-year-old Dalmatian bitch with a history of weight gain and changes such as rough dry brittle hair. For about a year, the dog also had also presented symmetrical erythematous and alopecia skin lesions in the regions of the hind limbs, lower back and tail, progressive lethargy and fatigue after exercise. A hemogram showed mild normocytic and normochromic regenerative anemia, as well as mild leukopenia and neutropenia. Biochemical tests revealed increased levels of creatinine, total cholesterol, and triglycerides. Skin biopsy revealed the presence of comedones in the epidermis and acanthosis and trichilemmal keratinization. Hormonal tests revealed high TSH and low free T4 and total T4 levels. A cervical ultrasound scan showed changes in the thyroid glands, with hypoechoic parenchyma, in addition to increased size of the right lobe, suggesting thyroiditis. Based on these exams, the patient was diagnosed with primary hypothyroidism, and treatment with levothyroxine was instituted.Discussion: Around 90% of dogs with hypothyroidism show a decrease in total T4 and about 65-75% of them show an increase in TSH levels. In endocrinopathy, there is a decrease in the expression of T3 receptors in the keratinocytes and genes responsible for epidermal renewal. Also, molecules that affect epidermal differentiation bind to intracellular receptors belonging to the steroid/thyroid hormone superfamily. In this study, however, the application of the minimum levothyroxine dose restored the normal epidermal pattern for the species and the remaining parameters returned to normal. The hormone T3 acts in the differentiation of keratinocytes, a fact that, although not yet proven, is believed to be indirect and mediated by the epidermal growth factor or by the expression of the genes responsible for the renewal of the epidermis. Besides, some effects on the epidermis caused by deficiency of thyroid hormones may be due to secondary vitamin A deficiency, which is necessary for epithelial differentiation and binding epidermal growth factor (EGF) to the cell. Still, there is a theory that sex hormones also affect the differentiation of the epidermis, as studies show that deficiency causes effects similar to those caused by hypothyroidism. Some studies indicate the investigation of the relationship between secondary vitamin D deficiency and the increased cellularity of the epidermis of the spayed hypothyroid female rats. Additionally, the mechanism of the formation of hyperkeratosis in hypothyroidism is not yet elucidated. T3 is known to regulate keratingene expression and perhaps also epidermal maturation, epithelial cycle, and normal keratin synthesis. The single treatment with levothyroxine in the female dog was effective in promoting the restoration of gene expression to T3 in keratinocytes.Keywords: dermatopathies, endocrinopathies, hypothyroidism, levothyroxine, synthetic hormone

    Developing a community of practice: report on a survey to determine the scholarly communication landscape in western Canada

    The Council of Prairie and Pacific University Libraries (COPPUL) Scholarly Communications Working Group (SCWG) surveyed COPPUL member libraries with a short questionnaire in November 2012. The stated purpose of the survey was to inform both the educational efforts of COPPUL with regard to scholarly communications, as well as the agenda of a proposed meeting of scholarly communication practitioners in COPPUL libraries. This paper discusses the results in the context of the formation of a Community of Practice (CoP) since conducting the survey. The paper concludes that a CoP has not yet formed; however, it presents the challenges with the formation of the CoP, identifies some of the actions taken so far, and makes recommendations for future direction for continuing to develop the CoP among COPPUL institutions.N

    A shared population of epidemic methicillin-resistant Staphylococcus aureus 15 circulates in humans and companion animals.

    UNLABELLED: Methicillin-resistant Staphylococcus aureus (MRSA) is a global human health problem causing infections in both hospitals and the community. Companion animals, such as cats, dogs, and horses, are also frequently colonized by MRSA and can become infected. We sequenced the genomes of 46 multilocus sequence type (ST) 22 MRSA isolates from cats and dogs in the United Kingdom and compared these to an extensive population framework of human isolates from the same lineage. Phylogenomic analyses showed that all companion animal isolates were interspersed throughout the epidemic MRSA-15 (EMRSA-15) pandemic clade and clustered with human isolates from the United Kingdom, with human isolates basal to those from companion animals, suggesting a human source for isolates infecting companion animals. A number of isolates from the same veterinary hospital clustered together, suggesting that as in human hospitals, EMRSA-15 isolates are readily transmitted in the veterinary hospital setting. Genome-wide association analysis did not identify any host-specific single nucleotide polymorphisms (SNPs) or virulence factors. However, isolates from companion animals were significantly less likely to harbor a plasmid encoding erythromycin resistance. When this plasmid was present in animal-associated isolates, it was more likely to contain mutations mediating resistance to clindamycin. This finding is consistent with the low levels of erythromycin and high levels of clindamycin used in veterinary medicine in the United Kingdom. This study furthers the "one health" view of infectious diseases that the pathogen pool of human and animal populations are intrinsically linked and provides evidence that antibiotic usage in animal medicine is shaping the population of a major human pathogen. IMPORTANCE: Methicillin-resistant Staphylococcus aureus (MRSA) is major problem in human medicine. Companion animals, such as cats, dogs, and horses, can also become colonized and infected by MRSA. Here, we demonstrate that a shared population of an important and globally disseminated lineage of MRSA can infect both humans and companion animals without undergoing host adaptation. This suggests that companion animals might act as a reservoir for human infections. We also show that the isolates from companion animals have differences in the presence of certain antibiotic resistance genes.
