There are no existing ultra-sterile lab-on-a-chip systems that can accept solid samples and perform complete chemical analyses without human intervention. The proposed solution is to demonstrate completely automated lab-on-a-chip manipulation of powdered solid samples, followed by on-chip liquid extraction and chemical analysis. This technology utilizes a newly invented glass micro-device for solid manipulation, which mates with existing lab-on-a-chip instrumentation. Devices are fabricated in a Class 10 cleanroom at the JPL MicroDevices Lab, and are plasma-cleaned before and after assembly. Solid samples enter the device through a drilled hole in the top. Existing micro-pumping technology is used to transfer milligrams of powdered sample into an extraction chamber where it is mixed with liquids to extract organic material. Subsequent chemical analysis is performed using portable microchip capillary electrophoresis systems (CE). These instruments have been used for ultra-highly sensitive (parts-per-trillion, pptr) analysis of organic compounds including amines, amino acids, aldehydes, ketones, carboxylic acids, and thiols. Fully autonomous amino acid analyses in liquids were demonstrated; however, to date there have been no reports of completely automated analysis of solid samples on chip. This approach utilizes an existing portable instrument that houses optics, high-voltage power supplies, and solenoids for fully autonomous microfluidic sample processing and CE analysis with laser-induced fluorescence (LIF) detection. Furthermore, the entire system can be sterilized and placed in a cleanroom environment for analyzing samples returned from extraterrestrial targets, if desired. This is an entirely new capability never demonstrated before. The ability to manipulate solid samples, coupled with lab-on-a-chip analysis technology, will enable ultraclean and ultrasensitive end-to-end analysis of samples that is orders of magnitude more sensitive than the ppb goal given in the Science Instruments

Pasta is a staple food with some quality advantages like long shelf life, nutritional value at energy intake in diet and wide acceptance in market. Quinoa is a pseudo-cereal that resurged from ancestral customs, due to its good nutritional quality and its potential given by an easy development of the crop under different environmental conditions with simple technologies. Pasta replaced with quinoa can represent benefits on consumer health and nutrition, as it will have a better nutritional quality compared to the market pasta from 100% wheat semolina. This research includes study of technological products with two levels of replacement of 10% and 30 %, analyzing the sustitution with two varieties Aurora and Blanca Jericó, cultivated in Colombia, as well as the analysis of addition of two substitutes gluten as Carboximetil Cellulose (2%) and pregelatinized corn starch (9%). The quality of prepared pasta was assessed on uncooked pasta (colour, nutritional value proximal, hardness/fracturability and microbiological quality), on cooking (optimum time of cooking and degradation by release of starch) and on cooked pasta (sensory evaluation, mechanical and biological properties). Results showed that pasta had technological standards required in the national legislation and are comparable with commercial pasta 100% wheat semolina, offering a nutritional quality improved product and a good microbiological quality. Differences were unclear among treatments, although pastas prepared with quinoa Blanca Jericó and carboxymethyl cellulose as gluten substitute exhibited better mechanical properties. Even though quality of cooking and mechanical properties are lower in pastas with quinoa flour compared to pastas with wheat semolina, pastas with quinoa flour exhibited promissory characteristics for special diets.Abstract: Pasta is a staple food with some quality advantages like long shelf life, nutritional value at energy intake in diet and wide acceptance in market. Evaluation of a Permutation-Based Evolutionary Framework for Lyndon Factorizations

    Get PDF
String factorization is an important tool for partitioning data for parallel processing and other algorithmic techniques often found in the context of big data applications such as bioinformatics or compression. Duval’s well-known algorithm uniquely factors a string over an ordered alphabet into Lyndon words, i.e., patterned strings which arestrictly smaller than all of their cyclic rotations. While Duval’s algorithm produces a pre-determined factorization, modern applications motivate the demand for factorizations with specific properties, e.g., those that minimize the number of factors or consist of factors with similar lengths. In this paper, we consider the problem of finding an alphabet ordering that yields a Lyndon factorization with such properties. We introduce a flexible evolutionary framework and evaluate it on biological sequence data. For the minimization case, we also propose a new problem-specific heuristic, Flexi-Duval, and a problem-specific mutation operator for Lyndon factorization. Our results show that our framework is competitive with Flexi-Duval for minimization and yields high quality and robust solutions for balancing where no problem-specific algorithm is available

CHANG-ES IV: Radio continuum emission of 35 edge-on galaxies observed with the Karl G. Jansky Very Large Array in D-configuration, Data Release 1

We present the first part of the observations made for the Continuum Halos in Nearby Galaxies, an EVLA Survey (CHANG-ES) project. The aim of the CHANG-ES project is to study and characterize the nature of radio halos, their prevalence as well as their magnetic fields, and the cosmic rays illuminating these fields. This paper reports observations with the compact D configuration of the Karl G. Jansky Very Large Array (VLA) for the sample of 35 nearby edge-on galaxies of CHANG-ES. With the new wide bandwidth capabilities of the VLA, an unprecedented sensitivity was achieved for all polarization products. The beam resolution is an average of 9.6" and 36" with noise levels reaching approximately 6 and 30 microJy per beam for C- and L-bands, respectively (robust weighting). We present intensity maps in these two frequency bands (C and L), with different weightings, as well as spectral index maps, polarization maps, and new measurements of star formation rates (SFRs). The data products described herein are available to the public in the CHANG-ES data release available at www.queensu.ca/changes. We also present evidence of a trend among galaxies with larger halos having higher SFR surface density, and we show, for the first time, a radio continuum image of the median galaxy, taking advantage of the collective signal-to-noise ratio of 30 of our galaxies. This image shows clearly that a typical spiral galaxy is surrounded by a halo of magnetic fields and cosmic rays.Comment: 70 pages, of which 35 pages present the data of each galax

Polyhydroxyalkanoate production from unexplored sugar substrates

Industrial-scale production of biopolymers is restricted by its elevated production costs in comparison with those associated with synthetic (no-biodegradable and no-biocompatible) polymers. In this study we tested for the first time two low-cost carbon substrates (i.e. carob pulp and fique juice) for lab-scale production of polyhydroxyalkanoate (PHA) with Bacillus megaterium. PHA detection and quantification was conducted by gas chromatography/mass spectrometry-selected ion monitoring (GC/MS-SIM). The results suggest that PHA production using carob pulp (from Hymenaea courbaril) may be as high as with sugar cane molasses. Moreover, it could serve for the synthesis of the most commercialized type of PHA (i.e. polyhydroxybutyrate; PHB) and/or other varieties (e.g. polyhydroxy-butyrate-co-valerate; PHBV) with different properties and potential applications
