Physical Treatments and Propolis Extract to Enhance Quality Attributes of Fresh-Cut Mixed Vegetables
The impact of individual and combined application of propolis extract (PE) plus ultrasound (ULT) or thermal treatment (TT) on the microbiological, nutritional, physicochemical and sensory quality of fresh-cut mixed vegetables for soup (celery, leek and butternut squash) stored at 5C was studied. The use of PE slightly reduced microbial growth, inhibited the activity of browning related enzymes and improved quality attributes during refrigerated storage. Moreover, PE was effective to reduce ascorbic acid losses in the product maintaining its nutritional quality. TT greatly lowered the initial microbial load (1.7–2.2 log units) and also reduced microbial growth on mixed vegetables extending its microbiological shelf-life to 5 days. Furthermore, TT inactivated browning enzymes improving visual quality; however, ascorbic acid degradation adversely affected nutritional quality. Meanwhile, ULT caused a significant microbial inactivation, reduced browning enzyme activity and also the inhibitory effect on polyphenoloxidase enzyme was enhanced by combining ULT with PE. Practical Applications: The use of natural agents, as propolis extract, along with physical treatments to preserve the quality of fresh-cut mixed vegetables for soup might be an interesting option to address the concerns of the consumer about the use of synthetic chemical antimicrobials potentially harmful for health.Fil: Alvarez, María Victoria. Modulation of the immuno-coagulative response in a pneumococcal infection in malnourished mice nasally treated with Lactobacillus casei
We studied the systemic effects of the intranasal administration of Lactobacillus casei on the immuno-coagulative response in pneumoccocal infection in immunocompromised mice. Weaned mice consumed a protein-free diet (PFD) for 21 days and were therefore malnourished. Malnourished mice were fed a balanced conventional diet (BCD) for 7 days (BCD group) or a BCD for 7 days with nasal administration of viable L. casei on days 6 and 7 (BCD+LcN group). The malnourished control mice (MNC) received a PFD, whereas the well-nourished control mice (WNC) continually consumed a BCD. At the end of the treatment period, the mice were infected with Streptococcus pneumoniae. At different times after infection, we analysed the following parameters: global coagulation system, activation of coagulation, coagulation inhibitors, platelet count, leukocyte count and myeloperoxidase (MPO) activity, total proteins, albumin and acute phase proteins (APPs). The MNC group showed greater impairment in the coagulation tests and an increase in the positive APPs. These parameters were normalized by the L. casei treatment. However, the number of leukocytes, decreased by malnutrition, was improved only by the administration of L. casei. After infection, the BCD+LcN group showed similar results to those of the WNC group for most of the haemostatic parameters. The BCD+LcN group did not show significant variations in the prothrombin time or in the level of anticoagulant protein C, but showed higher levels of fibrinogen, platelets, albumin, leukocytes and MPO activity compared with the different experimental groups. The intranasal administration of L. casei was effective in modulating the pro-inflammatory aspects of coagulation without affecting coagulation itself.Fil: Zelaya, María Hortensia del Rosario. Oregano essential oil-pectin edible films as anti-quorum sensing and food antimicrobial agents
Edible films can be used as carriers for antimicrobial compounds to assure food safety andquality; in addition, pathogenesis of food bacteria is related to a cell to cell communicationmechanism calledquorum sensing(QS). Oregano essential oil (OEO) has proved to beuseful as food antimicrobial; however, its foodapplications can be compromised by thevolatile character of its active constituents.Therefore, formulation of edible films containingOEO can be an alternative to improve its food usages. QS inhibitory activity of OEO andpectin-OEO films was evaluated usingChromobacterium violaceumas bacterial model.Additionally, antibacterial activity was tested againstEscherichia coliO157:H7,SalmonellaCholeraesuis,Staphylococcus aureus,andListeria monocytogenes. OEO was effective toinhibit bacterial growth at MIC of 0.24 mg/mL for all tested bacteria and MBC of 0.24,0.24, 0.48, and 0.24 mg/mL againstE. coliO157:H7,S.Choleraesuis,S. aureus, andL.monocytogenes, respectively. Pectin-films incorporated with 36.1 and 25.9 mg/mL of OEOshowed inhibition diameters of 16.3 and 15.2 mm forE. coliO157:H7; 18.1 and 24.2 mm forS.Choleraesuis; 20.8 and 20.3 mm forS. aureus; 21.3 and 19.3 mm forL. monocytogenes,respectively. Pectin-OEO film (15.7 mg/mL) was effective againstE. coliO157:H7 (9.3 mm),S. aureus(9.7 mm), andL. monocytogenes(9.2 mm), but not forS.Choleraesuis. Allconcentrations of OEO (0.0156, 0.0312, 0.0625 and 0.125 mg/mL) and pectin-OEO films(15.7, 25.9 and 36.1 mg/mL) showed a significant anti-QS activity expressed as inhibition ofviolacein production byC. violaceum. Buenos Aires, Argentina.8Research networks provide a framework for review, synthesis and systematic testing of theories by multiple scientists across international borders critical for addressing global-scale issues. In 2012, a GHG research network referred to as MAGGnet (Managing Agricultural Greenhouse Gases Network) was established within the Croplands Research Group of the Global Research Alliance on Agricultural Greenhouse Gases (GRA). With involvement from 46 alliance member countries, MAGGnet seeks to provide a platform for the inventory and analysis of agricultural GHG mitigation research throughout the world. To date, metadata from 315 experimental studies in 20 countries have been compiled using a standardized spreadsheet. Most studies were completed (74%) and conducted within a 1-3-year duration (68%). Soil carbon and nitrous oxide emissions were measured in over 80% of the studies. Among plant variables, grain yield was assessed across studies most frequently (56%), followed by stover (35%) and root (9%) biomass. MAGGnet has contributed to modeling efforts and has spurred other research groups in the GRA to collect experimental site metadata using an adapted spreadsheet. With continued growth and investment, MAGGnet will leverage limited-resource investments by any one country to produce an inclusive, globally shared meta-database focused on the science of GHG mitigation
Foliar morpho-anatomy of Araujia odorata (Apocynaceae - Asclepiadoideae) in an altitudinal gradient: Histochemistry of laticifers
Araujia odorata, es un subarbusto nativo de Argentina, Brasil, Paraguay y Uruguay, cuyo látex, raíces y hojas son utilizados en medicina popular. Se plantea como objetivo realizar un estudio morfo-anatómico foliar de seis poblaciones del Noroeste Argentino en un gradiente altitudinal (359-2155 m.s.n.m) y determinar la naturaleza de los compuestos presentes en laticíferos de tallos y frutos mediante técnicas convencionales de anatomía vegetal. Las poblaciones estudiadas no evidenciaron diferencias morfo-anatómicas significativas. Presentan hojas simples, venación pinnada broquidódroma, lámina isolateral anfiestomática, estomas braqui, anomo y anficiclocíticos, tricomas eglandulares, nervio medio con haz bicolateral y laticíferos no-articulados continuos en pecíolo, tallo y frutos. Se observan diferencias en los parámetros foliares cuantificados, sin embargo, solo la de densidad de tricomas, estomas y el espesor de cutícula se correlacionan positivamente con el gradiente altitudinal indicando plasticidad fenotípica. Se realiza por primera vez un análisis histoquímico de los laticíferos y otros idioblastos del tallo A. odorata.Araujia odorata is a sub-shrub native from Argentina, Brazil, Paraguay and Uruguay, whose latex, roots and leaves are used in traditional medicine. The objective of this work is to study the foliar morpho-anatomy of six populations in an altitudinal gradient (359-2155 m.a.s.l.) of Northwestern Argentine and to determine the nature of the compounds present in the laticiferous of the stems and fruits using conventional techniques for plant anatomy. The populations under study did not show significant morpho-anatomical differences. They presented simple leaves, pinnated brochydodromous venation, amphiestomatic isolateral lamina, brachy, anomo and amphicyclocytic stomata, eglandular trichomes, midvein with bicolateral vascular bundle and non-articulated laticifers continuous in the petiole, stem and fruits. Differences in the quantified foliar parameters are observed, however, only the density of trichomes, stomata and the thickness of the cuticle are positively correlated with the altitudinal gradient, indicating phenotypic plasticity. Lacticaseibacillus rhamnosus CRL1505 Peptidoglycan Modulates the Inflammation-Coagulation Response Triggered by Poly(I:C) in the Respiratory Tract
Lacticaseibacillus rhamnosus CRL1505 beneficially modulates the inflammation-coagulation response during respiratory viral infections. This study evaluated the capacity of the peptidoglycan obtained from the CRL1505 strain (PG-Lr1505) to modulate the immuno-coagulative response triggered by the viral pathogen-associated molecular pattern poly(I:C) in the respiratory tract. Adult BALB/c mice were nasally treated with PG-Lr1505 for two days. Treated and untreated control mice were then nasally challenged with poly(I:C). Mice received three doses of poly(I:C) with a 24 h rest period between each administration. The immuno-coagulative response was studied after the last administration of poly(I:C). The challenge with poly(I:C) significantly increased blood and respiratory pro-inflammatory mediators, decreased prothrombin activity (PT), and increased von Willebrand factor (vWF) levels in plasma. Furthermore, tissue factor (TF), tissue factor pathway inhibitor (TFPI), and thrombomodulin (TM) expressions were increased in the lungs. PG-Lr1505-treated mice showed significant modulation of hemostatic parameters in plasma (PT in %, Control = 71.3 ± 3.8, PG-Lr1505 = 94.0 ± 4.0, p < 0.01) and lungs. Moreover, PG-Lr1505-treated mice demonstrated reduced TF in F4/80 cells from lungs, higher pro-inflammatory mediators, and increased IL-10 compared to poly(I:C) control mice (IL-10 in pg/mL, Control = 379.1 ± 12.1, PG-Lr1505 = 483.9 ± 11.3, p < 0.0001). These changes induced by PG-Lr1505 correlated with a significant reduction in lung tissue damage. Complementary in vitro studies using Raw 264.7 cells confirmed the beneficial effect of PG-Lr1505 on poly(I:C)-induced inflammation, since increased IL-10 expression, as well as reduced damage, production of inflammatory mediators, and hemostatic parameter expressions were observed. In addition, protease-activated receptor-1 (PAR1) activation in lungs and Raw 264.7 cells was observed after TLR3 stimulation, which was differentially modulated by PG-Lr1505. The peptidoglycan from L. rhamnosus CRL1505 is able to regulate inflammation, the procoagulant state, and PAR1 activation in mice and macrophages in the context of the activation of TLR3 signaling pathways, contributing to a beneficial modulation of inflammation-hemostasis crosstalk.Fil: Zelaya, María Hortensia del Rosario. Morpho-chemical characterization of new confectionery sunflower (Helianthus annuus L.) genotypes from Argentina
Argentina is the world’s leading exporter of confectionery sunflower. This work characterized the morphometric and nutritional composition of sunflower seeds from new confectionery genotypes. Average morphometric variables per seed were: weight 0.11 and 0.06 g with and without achene, respectively; length 15.2 mm; and width 8.50 mm, with most calibers greater than 9.51 mm. Proximate analysis indicated that seeds without shell were an important source of lipids (49 %), as well as proteins (28 %), carbohydrates (11 %), and minerals (4.8 % ashes). Fatty acid composition showed that polyunsaturated fatty acids were the major components (56 %), followed by monounsaturated ones (34 %), and saturated ones (11 %). The mid-oleic genotype developed by INTA showed significantly greater oleic acid (47 %) and lower linoleic acid (42 %) than the commercial hybrid. Proteic composition showed 33 % of essential amino acids (EAA) while fiber and sugars represented 3.9 % and 7.3 % respectively; macroelements such as K, Mg and Ca, and microelements such as Mn, Fe, Cu, Zn, Se, Co, Mo and Na. New confectionery sunflower hybrids developed by INTA stand out for their high quality in edible oil, proteins and essential components, all very valuable for the food industry.Argentina es el principal exportador mundial de girasol confitero. Este trabajo caracterizó la composición morfométrica y nutricional de nuevos genotipos de girasol confitero. Las variables morfométricas promedio por semilla fueron: peso 0,11 g y 0,06 g, con y sin aquenio respectivamente; longitud 15,2 mm; ancho 8,50 mm; la mayoría de calibres superiores a 9,51 mm. El análisis composicional indicó que las semillas sin cáscara aportan 49 % de lípidos, 28 % de proteínas, 11 % de carbohidratos y 4,8 % de minerales. La composición de ácidos grasos mostró que los poliinsaturados fueron los componentes principales (56 %), seguidos de los monoinsaturados (34 %) y los saturados (11 %). El genotipo medio oleico presentó porcentaje de ácido oleico significativamente mayor (47 %) y porcentaje de ácido linoleico menor (42 %) que el híbrido comercial. La composición de proteínas fue de 33 % de aminoácidos esenciales (AAE); fibra y azúcares de 3,9 % y 7,3 %, respectivamente; macroelementos K, Mg y Ca, y microelementos Mn, Fe, Cu, Zn, Se, Co, Mo y Na. Zuccagnia-type Propolis from Argentina: A potential functional ingredient in food to pathologies associated to metabolic syndrome and oxidative stress
The effect of Argentine propolis extracts against enzymes related to metabolic syndrome and oxidative stress, as well as the botanical origin of raw propolis, were studied. Histological and chemical analyses of propolis samples revealed that the botanical origin is Zuccagnia punctata, an Argentine medicinal plant. The melissopalynological analysis showed both pollen grains of Z. punctata and the other plant species. This result indicates that the differences found in the botanical remains compared to the palynological studies may have been caused by the bees selecting resinous shrubs mainly of Z. punctata for the production of propolis and other plants with flowers for the production of honey. The richness of propolis was remarkable in two flavonoid precursors (2´,4´-dihydroxy-3´-methoxychalcone, 2´,4´-dihydroxychalcone), the major chemical components of Z. punctata. The hydroalcoholic extracts of Argentine propolis and Z. punctata were able to inhibit enzymes associated with the metabolic syndrome, including α-glucosidase, α-amylase and lipase, with IC50 values between 7 to 14, 37 to 48, and 13 to 28 µg/mL, respectively. Biological activity was mainly attributed to chalcones. Oxygen and nitrogen reactive species scavenging activity was determined by the assays of superoxide radical (O2•−), hydroxyl radical (HO•), hydrogen peroxide (H2O2), nitric oxide (NO•), and cation radical (ABTS•+). Results showed SC50 values between 115 to 278, 12.50 to 46; 39 to 92; 50 to 104.50 and 23 to 33.75 µg/mL, respectively. This study suggests for the first time that propolis from Argentina is highly effective in inhibiting enzymes related to the metabolic syndrome and in free-radical scavenging that would justify its use as a dietary supplement or as a functional ingredient in special food. Practical Application: Propolis from Catamarca, Argentina, is traditionally used as medicine and food. Its botanical origin is Zuccagnia punctata, an endemic plant species popularly used as a medicine in Argentina. Propolis has the ability to regulate the activity of enzymes involved in the carbohydrate and lipid metabolism, and consequently in metabolic syndrome. Besides, its antioxidant capacity makes it a natural product that can be used as a dietary supplement or as a functional ingredient in special foods. It is important to highlight that in the Argentine Food Code, propolis was incorporated in 2008 as a dietary supplement and the present results give major added value to this product.Fil: Salas, Ana Lilia. Impact of aetiological treatment on conventional and multiplex serology in chronic Chagas disease
The main criterion for treatment effectiveness in Chagas Disease has been the seronegative conversion of previously reactive serology, generally achieved many years post-treatment. The lack of reliable tests to ensure parasite clearance and to examine the effect of treatment is the main difficulty in evaluating treatment for chronic Chagas disease. Decreases of conventional and non-conventional serological titers can be useful tools to monitor the early impact of treatment. We serially measured changes in antibody levels, including seronegative conversion as well as declines in titers in 53 benznidazole-treated and 89 untreated chronically T. cruzi-infected subjects. Seronegative conversion as well as decreases of titers was significantly higher in treated compared with untreated patients. A strong concordance was found between decreases of titers of conventional and non-conventional serologic tests post-treatment, reaffirming the findings. When seronegative conversion plus decreases of titers were considered altogether, the impact of treatment was higher, in a shorter follow-up period than previously considered. New tools for monitoring the effectiveness of treatment of chronic Chagas disease are necessary, and the results showed in this study is a contribution to researchers and physicians who assist patients suffering from this disease
