    Need of Counselor for the Assessment of Psychological Problems of the Students’ at University Level

    The aim of the present study is to investigate the need of counselor for the assessment of the psychological problems of students at university level. The study was descriptive in nature. The population of the study was all the public-sector universities of Rawalpindi and Islamabad. The study was delimited to undergraduate students and faculty of social science. The total 343 students from Rawalpindi and Islamabad universities were selected through stratified random sampling technique. One self-developed questionnaire were constructed which include four areas, i.e. educational, vocational, social, personal problems of the students. The data was collected through questionnaire they were tested and validated before a well- designed test and interview is conducted on Head of Department(HOD).  The collected data were analyzed with the help of SPSS were interpreted through frequency, mean, standard deviation and chi- square. The finding revealed that students have counseling needs in all area of life. The results of this study show that students face more problem related to vocational and educational process at university stage. So, they need counseling more in vocational and educational areas. Students need a professional counsellor’s guidance to develop their career. Student services should be available on campus to help them develop self-awareness (personal interests, skills, potentials, weaknesses) and assess occupational opportunities such as, employment trends, expected competencies in the field, requirements for employment and job descriptions related to the area.     Role of School Heads’ Leadership Competencies in the Context of School Improvement Initiatives

    The aim of this research was to find out the relationship between school heads leadership competencies and the school results. It further investigated the on ground leadership competency challenges being faced by the head teachers of secondary schools in District Rawalpindi. Mix method approach was used. The study was basically descriptive in nature which required the collection of both qualitative and quantitative data by using a questionnaire and interviews.The information obtained through questionnaire were analyzed through, frequencies, percentage and mean value. Information gathered through interviews were qualitatively described. There were 285 teachers and 93 head teachers from tehsil Rawalpindi, Texila and Kotli Satian using the consecutive sampling technique. A questionnaire was developed for quantitative data collection. The study revealed that there is a significant relationship found between leadership competencies of school heads and school annual results. The study also depicted that the head teachers of the schools confront several challenges to improve their school including: lack of strict accountability in the system, heavy administrative duties, overcrowded classes, students affair management, weak result in Maths and Science, lack of trust of people on the education of government institutions, children coming from low socio economic backgrounds and lack of parents participation in the education of their children, lack of subject competent teachers, assessment practices, students drop out, medias’ role in portraying bad image of the government institution.     Synthesis of Breast Cancer Targeting Conjugate of Temporin-SHa Analog and its Effect on Pro- and Anti-Apoptotic Protein Expression in MCF-7 Cells

    The frog natural product temporin-SHa (FLSGIVGMLGKLFamide) is a potent antimicrobial peptide, as is the analog [S3K]SHa. By solid-phase synthesis, we prepared temporin-SHa and several temporin-SHa analogs with one or more D-alanine residues incorporated. The natural product and the analog [G10a]SHa were found to be cytotoxic in mammalian cell lines and induce cell death. To achieve selectivity, we conjugated the analog [G10a]SHa with a breast cancer targeting peptide (BCTP). The resulting peptide temporin [G10a]SHa-BCTP conjugate was selectively active against the MCF-7 breast cancer cell line with no cytotoxicity in NIH-3T3 fibroblasts. Unlike the natural product or [G10a]SHa, the conjugated peptide induced apoptosis, down regulating the expression of Bcl-2 and survivin and up regulating Bax and caspase-3

    Abstract Background Diabetes Mellitus (DM) poses a serious health problem worldwide and several inflammatory mediators are involved in the pathogenesis of this disease. Honey composed of various constituents which have been proven to have immunomodulatory and anti-inflammatory properties. The aim of this study is to investigate the in vitro and in vivo effects of Ziziphus honey and its isolated crude proteins in modulation of immune system and inflammation involved in the pathogenesis of diabetes. Methodology The proteins from Ziziphus honey were isolated by ammonium sulfate precipitation and estimated by Bradford method. In vitro anti-inflammatory activities were evaluated by inhibition of reactive oxygen species (ROS) from phagocytes via chemiluminescence immunoassay and nitric oxide (NO) by Griess method. Cytotoxicity was evaluated by MTT Assay. The comparative effect of oral and IP routes of honey and isolated proteins was observed in streptozotocin (STZ) induced diabetic male Wistar rats. qRT-PCR technique was utilized for gene expression studies. Results The honey proteins suppressed phagocyte oxidative burst and nitric oxide (NO) at significantly lower concentrations as compared to crude honey. The isolated proteins showed promising anti-inflammatory and hypoglycemic effects along with maintenance of body weight of rodents via both oral and IP routes, with significant down-regulation of inflammatory markers TNF-α, IL-1β, IFN-γ, iNOS, caspase 1, Calgranulin A (S100A8) and NF-κB expression in diabetic rats. Conclusion The isolated honey proteins showed better immunomodulatory and therapeutic potential at significantly lower doses as compared to crude honey

    Sulfamethazine (SMZ) (1) is an antibacterial sulfa drug which suppresses the synthesis of dihydrofolic acid. It is used for the treatment of infections in livestock; such as gastrointestinal, and respiratory tract infections. During the current study, synthesis, characterization, and evaluation of immunomodulatory activities of derivatives of sulfamethazine (SMZ) (3-39) was carried out. These derivatives were synthesized by the reaction of sulfamethazine with a range of acid chlorides. All the compounds were characterized by using modern spectroscopic techniques, such as 1H-, and 13C-NMR, EI-MS, and HRFAB-MS. Compounds 3-10, 14, and 15 were identified as new compounds. Immunomodulatory effect of compounds 3-39 on different parameters of innate immune response was evaluated, including the production of Reactive Oxygen Species (ROS) from human whole blood and isolated polymorphonuclear neutrophils (PMNs), nitric oxide (NO), and pro-inflammatory cytokine TNF-α. All the new compounds, except 14 and 15, showed a significant anti-inflammatory activity. Compounds 3-39 were also evaluated for their anti-bacterial activity and cytotoxicity (3T3 mouse fibroblast cell lines). All the compounds were found to be non-cytotoxic against normal cell lines

    Abstract Ultraviolet-A (UVA) radiation is a major contributor to reactive oxygen species (ROS), reactive nitrite species (RNS), inflammation, and DNA damage, which causes photoaging and photocarcinogenesis. This study aimed to evaluate the UVA protective potential of lipophilic chain conjugated thiourea-substituted aryl group molecules against UVA-induced cellular damages in human dermal fibroblasts (BJ cell line). We tested a series of nineteen (19) molecules for UVA photoprotection, from which 2′,5′-dichlorophenyl-substituted molecule DD-04 showed remarkable UVA protection properties compared to the reference (benzophenone). The results indicate that DD-04 significantly reduced intracellular ROS and nitric oxide (NO) as compared to the UVA-irradiated control (p < 0.001). Moreover, the compound DD-04 showed anti-inflammatory activity as it significantly reduced the levels of tumor necrosis factor-α (TNF-α) and interleukin-1β (IL-1β) pro-inflammatory cytokines produced by THP-1 (human monocytic) cells (p < 0.05). DNA damage was also prevented by DD-04 treatment in the presence of UVA. It was observed that DD-04 significantly reduced the number of cyclobutane pyrimidine dimers (CPDs) when compared to the UVA-irradiated control (p < 0.001). Finally, the DNA strand breaks were checked and a single intact DNA band was seen upon treatment with DD-04 in the presence of UVA. In conclusion, DD-04 can be considered a potential candidate UVA filter due to its photoprotective potential

    Objective: To explore anti-hyperalgesic properties of methanol extract of Piptadeniastrum africanum stem bark (PAME) and it possible action mechanism. Methods: PAME was tested on carrageenan induced hyperalgesia using plantar test (thermal) and analgesymeter (mechanical) in rats, on prostaglandin E2 (PGE2) induced mechanical hyperalgesia and vincristine induced neuropathic pain in rat, both with analgesymeter. Modulators of NO/ cGMP/K+ channel pathway and endogenous opioids receptor antagonists and/or agonists were used to determine the possible action mechanism of PAME. Results: PAME significantly decreased carrageenan induced thermal and mechanical hyperalgesia, as well as PGE2 induced mechanical hyperalgesia. PAME significantly protected the animals against the installation of neuropathic pain. Anti-nociception activity produced by PAME was significantly blocked in animals pre treated with all the antagonists (naloxone, NW-nitro-L-arginine methyl ester (L-NAME), methylene blue and glibenclamide). Conclusions: Results of this study reveal that, PAME administrate orally, can induce anti-hyperalgesic action against installation of inflammatory pain as well as neuropathic pain. The mechanism underlying PAME anti-hyperalgesic effect could probably be associated with an activation of opioid receptors and NO/cGMP/K+ channel pathway

    A New Model for Brain Tumor Detection Using Ensemble Transfer Learning and Quantum Variational Classifier

    A brain tumor is an abnormal enlargement of cells if not properly diagnosed. Early detection of a brain tumor is critical for clinical practice and survival rates. Brain tumors arise in a variety of shapes, sizes, and features, with variable treatment options. Manual detection of tumors is difficult, time-consuming, and error-prone. Therefore, a significant requirement for computerized diagnostics systems for accurate brain tumor detection is present. In this research, deep features are extracted from the inceptionv3 model, in which score vector is acquired from softmax and supplied to the quantum variational classifier (QVR) for discrimination between glioma, meningioma, no tumor, and pituitary tumor. The classified tumor images have been passed to the proposed Seg-network where the actual infected region is segmented to analyze the tumor severity level. The outcomes of the reported research have been evaluated on three benchmark datasets such as Kaggle, 2020-BRATS, and local collected images. The model achieved greater than 90% detection scores to prove the proposed model's effectiveness
