    Stroke-related Effects on Maximal Dynamic Hip Flexor Fatigability and Functional Implications

    Introduction: Stroke-related changes in maximal dynamic hip flexor muscle fatigability may be more relevant functionally than isometric hip flexor fatigability. Methods: Ten chronic stroke survivors performed 5 sets of 30 hip flexion maximal dynamic voluntary contractions (MDVC). A maximal isometric voluntary contraction (MIVC) was performed before and after completion of the dynamic contractions. Both the paretic and nonparetic legs were tested. Results: Reduction in hip flexion MDVC torque in the paretic leg (44.7%) was larger than the nonparetic leg (31.7%). The paretic leg had a larger reduction in rectus femoris EMG (28.9%) between the first and last set of MDVCs than the nonparetic leg (7.4%). Reduction in paretic leg MDVC torque was correlated with self-selected walking speed (r2 = 0.43), while reduction in MIVC torque was not (r2 = 0.11). Conclusions: Reductions in maximal dynamic torque of paretic hip flexors may be a better predictor of walking function than reductions in maximal isometric contractions

    Light with a self-torque: extreme-ultraviolet beams with time-varying orbital angular momentum

    Twisted light fields carrying orbital angular momentum (OAM) provide powerful capabilities for applications in optical communications, microscopy, quantum optics and microparticle rotation. Here we introduce and experimentally validate a new class of light beams, whose unique property is associated with a temporal OAM variation along a pulse: the self-torque of light. Self-torque is a phenomenon that can arise from matter-field interactions in electrodynamics and general relativity, but to date, there has been no optical analog. In particular, the self-torque of light is an inherent property, which is distinguished from the mechanical torque exerted by OAM beams when interacting with physical systems. We demonstrate that self-torqued beams in the extreme-ultraviolet (EUV) naturally arise as a necessary consequence of angular momentum conservation in non-perturbative high-order harmonic generation when driven by time-delayed pulses with different OAM. In addition, the time-dependent OAM naturally induces an azimuthal frequency chirp, which provides a signature for monitoring the self-torque of high-harmonic EUV beams. Such self-torqued EUV beams can serve as unique tools for imaging magnetic and topological excitations, for launching selective excitation of quantum matter, and for manipulating molecules and nanostructures on unprecedented time and length scales.Comment: 24 pages, 4 figure

    Biosynthesis of Modular Ascarosides in C. elegans

    The nematode Caenorhabditis elegans uses simple building blocks from primary metabolism and a strategy of modular assembly to build a great diversity of signaling molecules, the ascarosides, which function as a chemical language in this model organism. In the ascarosides, the dideoxysugar ascarylose serves as a scaffold to which diverse moieties from lipid, amino acid, neurotransmitter, and nucleoside metabolism are attached. However, the mechanisms that underlie the highly specific assembly of ascarosides are not understood. We show that the acyl-CoA synthetase ACS-7, which localizes to lysosome-related organelles, is specifically required for the attachment of different building blocks to the 4′-position of ascr#9. We further show that mutants lacking lysosome-related organelles are defective in the production of all 4′-modified ascarosides, thus identifying the waste disposal system of the cell as a hotspot for ascaroside biosynthesis

    The aim of this paper is to provide a close reading of Deleuze's complex account of Freud's Beyond the Pleasure Principle in Difference and Repetition. The first part provides a reading of Beyond the Pleasure Principle itself, showing why Freud feels the need to develop a transcendental account of repetition. In the second, I show the limitations of Freud's account, drawing on the work of Weismann to argue that Freud's transcendental model mischaracterises repetition. In the third part, I show how Freud's account of the death drive is shadowed by Deleuze's own non-representational transcendental account

    Host cell lipid droplets (LD) are essential in the hepatitis C virus (HCV) life cycle and are targeted by the viral capsid core protein. Core-coated LDs accumulate in the perinuclear region and facilitate viral particle assembly, but it is unclear how mobility of these LDs is directed by core. Herein we used two-photon fluorescence, differential interference contrast imaging, and coherent anti-Stokes Raman scattering microscopies, to reveal novel core-mediated changes to LD dynamics. Expression of core protein’s lipid binding domain II (DII-core) induced slower LD speeds, but did not affect directionality of movement on microtubules. Modulating the LD binding strength of DII-core further impacted LD mobility, revealing the temporal effects of LD-bound DII-core. These results for DII-core coated LDs support a model for core-mediated LD localization that involves core slowing down the rate of movement of LDs until localization at the perinuclear region is accomplished where LD movement ceases. The guided localization of LDs by HCV core protein not only is essential to the viral life cycle but also poses an interesting target for the development of antiviral strategies against HCV
