    Contaminantes bacterianos y perfil de susceptibilidad del semen porcino en centros de recogida em Brasil

    Objectivo: Evaluar el perfil microbiológico de siete centros de recogida de semen porcino (CRSP) en el sur de Brasil, así como evaluar la susceptibilidad antimicrobiana a la mayoría de los microorganismos encontrados en los CRSP. Material y métodos: El análisis bacteriológico se realizó en muestras del sistema de purificación de agua, diluyentes de semen, semen crudo y almacenado, bancos de laboratorio y otras superficies de trabajo. Resultados: El crecimiento de una población bacteriana mixta se observó en muestras de agua de todas las CRSP excepto una. Aproximadamente el 85% de la CRSP tenía contaminación significativa en sus superficies de trabajo con al menos un contaminante bacteriano. Un total de 86% de muestras de semen crudo fueron contaminadas con una o más bacterias diferentes, mientras que el 100% de CRSP proporcionaron muestras contaminadas. La susceptibilidad bacteriana a agentes antimicrobianos varió en más del 80% para gentamicina, neomicina y ceftiofur a 40% o menos para penicilina y lincomicina. Conclusiones: La identificación de los puntos críticos proporciona el apoyo necesario para idear mejores estrategias para minimizar la contaminación en CRSP. Además, la evaluación del nivel de resistencia a los antimicrobianos ofrece información precisa para formular protocolos antibacterianos más eficientes que tengan en cuenta el uso racional de los antibióticos

    Using of essential oils in the treatment of mice infected with Trypanosoma evansi

    Objective. This study aimed to test the effectiveness of copaiba, andiroba and aroeira essential oils for controlling trypanosomosis by Trypanosoma evansi with mice as experimental model. Materials and methods. Sixty-six mice were divided into eleven groups (A to L) with six animals each. Group A was the unique composed by healthy and uninfected animals (negative control). Animals in groups B to L were inoculated with 0.1 mL of blood containing 2.7 x 106 trypanosomes. Group B was used as a positive control without treatment. In experiment were tested copaiba (C, D and E), andiroba (F, G and H) and aroeira (I, J and L) oils at doses of 0.6, 0.8 and 1.0 mL kg-1 to infected mice (T. evansi). Results. These protocols did not provide curative efficacy; however, the mice treated with highest dose of copaiba showed a significant increase in the longevity when compared others groups. Conclusions. Previously in our studies, these essential oils have shown trypanocidal activity in vitro, but when they were tested in vivo in mice infected with T. evansi, this trypanocidal activity, or the curative effect was not found, being only able to prolong the lifespan of the animals treated with copaiba oil

    Influence of gastrointestinal parasitism on biochemical variables in blood of laying hens

    ABSTRACT Objective. This study describes the influence of endoparasites and diet on biochemical variables in sera of brown laying hens. Materials and methods. Biochemical parameters (seric levels of cholesterol, triglycerides, total protein, uric acid, albumin, and alanine aminotransferase activity) were assessed in blood samples of poultry. Three populations of laying hens of different properties (n=20 each) represented our experimental design, from which blood (serum) and feces were collected for biochemical and parasitological analyzes, respectively. Results. From 60 feces samples evaluated 49 were positive for the presence of parasites and 11 were negative. Among the positive samples, 23 had infection with Eimeria spp and the other 26 samples had mixed parasitism with Eimeria spp and Heterakis gallinarum, which influenced (increasing) the levels of total protein and globulin. However, when parasites were analyzed separately, it was observed that the infection by Eimeria spp affected parameters such as cholesterol (which increased, p=0.001) and triglycerides, showing lower results than when the hen was parasitized by Eimeria spp. (p=0.01). Significant (p<0.05) difference was observed in all biochemical variables when the three diets were compared. Conclusions. Our results allowed concluding that the parasitic infection and diet in laying hens influenced the results of serum biochemical parameters evaluated. The main conclusion we found that parasitism caused an increase in total protein and globulin (ie immune response), and there was a negative correlation between parasite load and albumin, uric acid and ALT

    Biochemical detection of E-ADA on Neospora caninum tachyzoites and the effects of a specific enzymatic inhibitor

    Objective. This study aimed to investigate the presence and activity of the ecto adenosine deaminase (E-ADA) enzyme in tachyzoites of Neospora caninum (Nc-1 strain), as well as to assess the activity of a well-known E-ADA inhibitor, the deoxycoformycin. Materials and methods. The parasites were grown in cell culture, being subsequently separated in a pellet of tachyzoites, on which the E-ADA activity was tested using the concentrations 0 (control), 0.2, 0.4 and 0.8 mg mL-1. Results. The E-ADA showed high activity, progressively increasing its activity according to the enhancement of the protein concentration. The test was carried out with different concentrations of deoxycoformycin, showing that it was able to inhibit the E-ADA present on the free form of the parasite. Conclusions. Based on these results we conclude that the E-ADA is present on tachyzoites of N. caninum, and deoxycoformycin is able to inhibit this enzyme. In this sense, knowing the negative impact of N. caninum on reproductive issue in cattle (mainly abortion), might it is an alternative in order to deal with this parasitic infection.     Susceptibility of Mice to Trypanosoma evansi Treated with Human Plasma Containing Different Concentrations of Apolipoprotein L-1

    The aim of this study was to test the susceptibility of mice to Trypanosoma evansi treated with human plasma containing different concentrations of apolipoprotein L-1 (APOL1). For this experiment, a strain of T. evansi and human plasma (plasmas 1, 2, and 3) from 3 adult males clinically healthy were used. In vivo test used 50 mice divided in 5 groups (A to E) with 10 animals in each group. Animals of groups B to E were infected, and then treated with 0.2 ml of human plasma in the following outline: negative control (A), positive control (B), treatment with plasma 1 (C), treatment with plasma 2 (D), and treatment with plasma 3 (E). Mice treated with human plasma showed an increase in longevity of 40.9±0.3 (C), 20±9.0 (D) and 35.6±9.3 (E) days compared to the control group (B) which was 4.3±0.5 days. The number of surviving mice and free of the parasite (blood smear and PCR negative) at the end of the experiment was 90%, 0%, and 60% for groups C, D, and E, respectively. The quantification of APOL1 was performed due to the large difference in the treatments that differed in the source plasma. In plasmas 1, 2, and 3 was detected the concentration of 194, 99, and 115 mg/dl of APOL1, respectively. However, we believe that this difference in the treatment efficiency is related to the level of APOL1 in plasmas

    Bacterial Contaminants and Antimicrobial Susceptibility Profile of Boar Semen in Southern Brazil Studs

    Objective: To assess the microbiological profile in seven Boar Studs (BS) in Southern Brazil, as well as evaluate antimicrobial susceptibility response to most commonly found microorganisms in BS. Material and methods: Bacteriologic analysis was carried out in samples from the water purification system, semen extender, raw and stored semen, lab benches, and other working surfaces. Results: Growth of a mixed bacterial population was observed in water samples from all but one BS. Approximately 85% of the BS had significant contamination on their working surfaces with at least one bacterial contaminant. A total of 86% of raw semen samples were contaminated with one or more different bacteria, while 100% of the boar studs provided contaminated samples. Bacterial susceptibility to antimicrobial agents varied from over 80% for gentamycin, neomycin and ceftiofur to 40% or less for penicillin and lincomycin. Conclusions: The identification of the critical points provides necessary support to devise better strategies to minimize contamination in BS. Also, assessing the level of antimicrobial drug resistance offers accurate information to formulate more efficient antibacterial protocols that closely observe the rational use of antibiotics

Los parámetros bioquímicos (niveles séricos de colesterol, triglicéridos, proteínas totales, ácido úrico, albúmina, y la actividad de la alanina aminotransferasa) se evaluaron en muestras de sangre de las aves de corral. Tres poblaciones de gallinas ponedoras de distintas propiedades (n=20 cada uno) representan nuestro diseño experimental, de la que se recogieron la sangre (suero) y las heces de bioquímica y análisis parasitológico, respectivamente. Resultados. De 60 muestras de heces evaluadas, 49 fueron positivos para la presencia de parásitos y 11 fueron negativos. Entre las muestras positivas, 23 tenían infección con Eimeria spp y las otras 26 muestras tenían parasitismo mezclado con Eimeria spp y Heterakis gallinarum, que influyó (aumentando) los niveles de proteína total y globulina. Sin embargo, cuando se analizaron por separado los parásitos, se observó que la infección por Eimeria spp afectó parámetros tales como el colesterol (el cual se incrementó p=0.001) y los triglicéridos, que mostraron resultados más bajos que cuando las gallinas fueron parasitadas por Eimeria spp. (p=0.01). Se observó diferencia (p<0.05) en todas las variables bioquímicas, cuando se compararon las tres dietas. Conclusiones. Nuestros resultados permitieron concluir que la infección parasitaria y la dieta de las gallinas ponedoras influyeron en los resultados de los parámetros bioquímicos séricos evaluados. La principal conclusión que se encontró es que el parasitismo causó un aumento de la proteína total y la globulina (es decir, la respuesta inmune), y hubo una correlación negativa entre la carga de parásitos y albúmina, ácido úrico y ALT
