    Structural characterization of the carbohydrate-binding module of NanA sialidase, a pneumococcal virulence factor

    This research was supported by the University of St Andrews and grants provided by the Medical Research Council.Background: Streptococcus pneumoniae Neuraminidase A (NanA) is a multi-domain protein anchored to the bacterial surface. Upstream of the catalytic domain of NanA is a domain that conforms to the sialic acid-recognising CBM40 family of the CAZY (carbohydrate-active enzymes) database. This domain has been identified to play a critical role in allowing the bacterium to promote adhesion and invasion of human brain microvascular endothelial cells, and hence may play a key role in promoting bacterial meningitis. In addition, the CBM40 domain has also been reported to activate host chemokines and neutrophil recruitment during infection. Results: Crystal structures of both apo- and holo- forms of the NanA CBM40 domain (residues 121 to 305), have been determined to 1.8 angstrom resolution. The domain shares the fold of other CBM40 domains that are associated with sialidases. When in complex with alpha 2,3- or alpha 2,6-sialyllactose, the domain is shown to interact only with the terminal sialic acid. Significantly, a deep acidic pocket adjacent to the sialic acid-binding site is identified, which is occupied by a lysine from a symmetry-related molecule in the crystal. This pocket is adjacent to a region that is predicted to be involved in protein-protein interactions. Conclusions: The structural data provide the details of linkage-independent sialyllactose binding by NanA CBM40 and reveal striking surface features that may hold the key to recognition of binding partners on the host cell surface. The structure also suggests that small molecules or sialic acid analogues could be developed to fill the acidic pocket and hence provide a new therapeutic avenue against meningitis caused by S. pneumoniae.Publisher PDFPeer reviewe

    Exploring The Neural Correlates of Reading Comprehension and Social Cognition Deficits in College Students with ADHD

    Attention-Deficit/Hyperactivity Disorder (ADHD) is a neurodevelopmental disorder characterized by inattention, impulsivity, and hyperactivity. Symptoms of this disorder have been shown to adversely impact academic and social functioning of those with ADHD. College students with ADHD, compared to their non-ADHD peers, are at increased risk for academic and social difficulties. Given the reading-intensive and socially-driven environment of the college campus, empirical literature examining the reading comprehension and social cognition of college students are wanting. The current investigation utilized the Nelson-Denny Reading Test (NDRT) and Faux Pas Recognition test (FPRT) to assess reading comprehension and social cognition, respectively, in college students with (n = 3) and without ADHD (n = 9). The Short Story Task (SST) was administered during functional magnetic resonance imaging (fMRI) to examine neural correlates of narrative comprehension and theory of mind (ToM) while reading short fictional stories of varying prose complexity. The ADHD and control groups did not differ in IQ, GPA, or scores of NDRT, FPRT, or SST, suggesting that they had comparable academic performance, narrative comprehension, and social cognition. The fMRI analysis of SST showed that the ADHD group demonstrated increased activation in the left anterior cingulate (ACC) and parahippocampal gyrus (PHG) while reading the complex story compared to the simple story. This differential activation was not observed in the CTRL group, suggesting that the ADHD group required more neural resources to process the emotional components of the complex story to achieve the comparable performance on the SST. The ADHD group additionally exhibited lower activation in the narrative comprehension and ToM networks (medial prefrontal cortex, Broca’s area, angular gyri). Collectively, these results indicate that while ADHD and CTRL groups did not differ behaviorally, they exhibit differential neural activation patterns in tasks related to narrative comprehension and social cognition. Further investigations may inform the development of educational and psychosocial interventions to improve academic and social functioning in young adults with ADHD

    Positive affect in young children's multiliteracies learning endeavors

    This chapter focuses on the processes and conditions for the emergence of positive affect during children's multiliteracies learning endeavors in a Finnish pre-primary school that culminated in an exhibition at a local library. It identifies three thematic categories of activity that correlated strongly with positive, affective outcomes, namely, making and producing, sharing experiences, and sustaining interest. The chapter demonstrates how positive affect is evoked when children are afforded opportunities to create, make, and share different texts and interests in the course of their multimodal, multisensory, and playful activities, developing skills and dispositions of competence, relatedness, and agency.Peer reviewe

    Cognitive Flexibility and Academic Performance in College Students with ADHD: An fMRI Study

    Cognitive flexibility, or the ability to change behavior or cognitive action appropriately in response to context shifts, is crucial to college-level learning, as it is needed for solving problems that require a transfer of familiar knowledge to novel concepts. Cognitive flexibility is known to involve neural areas concentrated in the frontal lobes, such as the inferior and superior frontal gyri and the anterior cingulate cortex, but the activation of this network is typically weaker in the ADHD population. In academic settings, individuals with ADHD tend to perform below healthy peers, have lower GPAs, are more likely to be on academic probation, and have decreased motivation. While most of these downfalls are attributed to attentional deficits, no investigations have been done to assess any effects of cognitive flexibility on academic performance. Therefore, this study used the Wisconsin Card Sorting Task (WCST) in the fMRI to assess cognitive flexibility in college students with and without ADHD and compared task performance to academic performance to determine if there is any relationship. We found that the ADHD group presented more perseverative errors on the WCST, and this performance was negatively correlated to GPA, suggesting that cognitive flexibility deficits impair academic performance. We also observed decreased activation of the anterior cingulate cortex and inferior and superior frontal gyri in subjects with ADHD as compared to controls, suggesting a possible neural network involved in these behavioral deficits

    Design and Validation of a Portable Handheld Device to Produce Fine Fibers Using Centrifugal Forces

    In the present research project, a novel portable battery-powered handheld device able to produce micron and submicron fibers using centrifugal forces is proposed. The design includes spinnerets with a clamshell configuration with multiple chambers or reservoirs (2, 4, and 8) and different exit orifice diameters (400, 500, 600, and 800 µm). The rotational speed is controlled via an Arduino microcontroller. To validate the design, a series of experiments were conducted and the effect of the orifice diameter, number of chambers, and velocity on the resulting fibers’ diameter and yield was studied. For the experiments, a polymeric solution of Polyvinyl Alcohol (PVA) was prepared. The fiber yield was gravimetrically quantified, and the fiber morphology and diameter were analyzed by means of scanning electron microscopy (SEM). The experimental results showed that spinnerets with an orifice diameter of 500 microns yielded the greatest amount of fibers (0.0777 g). In addition, the number of chambers also affected the amount of fibers produced, and it was determined that the fiber diameter size is dependent on the spinneret speed. Fibers 80 nm in diameter were observed at 6500 rpm

    Expression capable library for studies of Neisseria gonorrhoeae, version 1.0

    Background The sexually transmitted disease, gonorrhea, is a serious health problem in developed as well as in developing countries, for which treatment continues to be a challenge. The recent completion of the genome sequence of the causative agent, Neisseria gonorrhoeae, opens up an entirely new set of approaches for studying this organism and the diseases it causes. Here, we describe the initial phases of the construction of an expression-capable clone set representing the protein-coding ORFs of the gonococcal genome using a recombination-based cloning system. Results The clone set thus far includes 1672 of the 2250 predicted ORFs of the N. gonorrhoeae genome, of which 1393 (83%) are sequence-validated. Included in this set are 48 of the 61 ORFs of the gonococcal genetic island of strain MS11, not present in the sequenced genome of strain FA1090. L-arabinose-inducible glutathione-S-transferase (GST)-fusions were constructed from random clones and each was shown to express a fusion protein of the predicted size following induction, demonstrating the use of the recombination cloning system. PCR amplicons of each ORF used in the cloning reactions were spotted onto glass slides to produce DNA microarrays representing 2035 genes of the gonococcal genome. Pilot experiments indicate that these arrays are suitable for the analysis of global gene expression in gonococci. Conclusion This archived set of Gateway® entry clones will facilitate high-throughput genomic and proteomic studies of gonococcal genes using a variety of expression and analysis systems. In addition, the DNA arrays produced will allow us to generate gene expression profiles of gonococci grown in a wide variety of conditions. Together, the resources produced in this work will facilitate experiments to dissect the molecular mechanisms of gonococcal pathogenesis on a global scale, and ultimately lead to the determination of the functions of unknown genes in the genome

    CD147 mediates intrahepatic leukocyte aggregation and determines the extent of liver injury

    Background: Chronic inflammation is the driver of liver injury and results in progressive fibrosis and eventual cirrhosis with consequences including both liver failure and liver cancer. We have previously described increased expression of the highly multifunctional glycoprotein CD147 in liver injury. This work describes a novel role of CD147 in liver inflammation and the importance of leukocyte aggregates in determining the extent of liver injury. Methods: Non-diseased, progressive injury, and cirrhotic liver from humans and mice were examined using a mAb targeting CD147. Inflammatory cell subsets were assessed by multiparameter flow cytometry. Results: In liver injury, we observe abundant, intrahepatic leukocyte clusters defined as ≥5 adjacent CD45+ cells which we have termed “leukocyte aggregates”. We have shown that these leukocyte aggregates have a significant effect in determining the extent of liver injury. If CD147 is blocked in vivo, these leukocyte aggregates diminish in size and number, together with a marked significant reduction in liver injury including fibrosis. This is accompanied by no change in overall intrahepatic leukocyte numbers. Further, blocking of aggregation formation occurs prior to an appreciable increase in inflammatory markers or fibrosis. Additionally, there were no observed, “off-target” or unpredicted effects in targeting CD147. Conclusion: CD147 mediates leukocyte aggregation which is associated with the development of liver injury. This is not a secondary effect, but a cause of injury as aggregate formation proceeds other markers of injury. Leukocyte aggregation has been previously described in inflammation dating back over many decades. Here we demonstrate that leukocyte aggregates determine the extent of liver injury
