    Pemetaan Potensi Tanah Putih Menggunakan Sistem Informasi Geografis (SIG)

    Potensi tanah putih pada suatu wilayah dapat diBerdasarkan kondisi geologi, sebagian besar wilayah Kota Kupang memiliki struktur geologi batuan gamping sehingga berpotensi sebagai sumber tanah putih. Penelitian ini bertujuan untuk melakukan pemetaan lokasi-lokasi potensi tanah putih di Kota Kupang dalam sistem informasi geografis (SIG). Hasil dari penelitian ini menunjukkan bahwa terdapat 4 lokasi yang sumber tanah putih di kota Kupang yaitu lokasi milik perusahaan (PTS) di Kecamatan Maulafa, lokasi milik masyarakat (BN) di Kecamatan Kelapa Lima, lokasi milik PT. Anugerah (PTA) di Kecamatan Alak dan lokasi milik PT.Semen Kupang (PTSK) di Kecamatan Alak. Tanah putih yang berasal lokasi Kecamatan Maulafa memiliki kandungan mineral kalsium oksida sebesar (CaO) 55.81%, sedangkan tanah putih milik BN, Kecamatan Kelapa Lima mengandung mineral kalsium oksida sebesar 49.51%. Kandungan CaO yang hampir sama juga dimiliki oleh tanah putih dari PT.Anugerah di Kecamatan Alak (55,39%) dan tanah putih PT.Semen di Kecamatan Alak (53.49%). Berdasarkan RDRTK Kota Kupang Tahun 2011-2031, 3 lokasi yang layak dikembangkan sebagai sumber tanah putih adalah 1 lokasi di kecamatan Maulafa, 2 lokasi di Kecamatan Ala

    Exosomes from metastatic cancer cells transfer amoeboid phenotype to non-metastatic cells and increase endothelial permeability: their emerging role in tumor heterogeneity

    The goal of this study was to understand if exosomes derived from high-metastatic cells may influence the behavior of less aggressive cancer cells and the properties of the endothelium. We found that metastatic colon cancer cells are able to transfer their amoeboid phenotype to isogenic primary cancer cells through exosomes, and that this morphological transition is associated with the acquisition of a more aggressive behavior. Moreover, exosomes from the metastatic line (SW620Exos) exhibited higher ability to cause endothelial hyperpermeability than exosomes from the non metastatic line (SW480Exos). SWATH-based quantitative proteomic analysis highlighted that SW620Exos are significantly enriched in cytoskeletal-associated proteins including proteins activating the RhoA/ROCK pathway, known to induce amoeboid properties and destabilization of endothelial junctions. In particular, thrombin was identified as a key mediator of the effects induced by SW620Exos in target cells, in which we also found a significant increase of RhoA activity. Overall, our results demonstrate that in a heterogeneous context exosomes released by aggressive sub-clones can contribute to accelerate tumor progression by spreading malignant properties that affect both the tumor cell plasticity and the endothelial cell behavior

    Comparison of the Transdermal and Intravenous Administration of Buprenorphine in the Management of Intra- and Postoperative Pain in Dogs Undergoing a Unilateral Mastectomy

    The aim of this prospective clinical study was to evaluate the effectiveness of transdermal patches of buprenorphine as an alternative route for the management of perioperative pain in dogs undergoing a unilateral mastectomy. Our hypothesis was that the transdermal route would allow the obtainment of an analgesic plan comparable to that of the injectable administration. Twelve dogs were divided in two groups. In the BupreP group (six dogs), buprenorphine patches were applied 40 h before the start of the surgery, guaranteeing a dosage of 5-6 μg/kg/h. In the BupreI group (six dogs), 20 μg/kg of buprenorphine was administered intravenously 30 min before the induction of anesthesia, and this was repeated every 6 h for 24 h. The main physiological parameters, sedation scores (0 = no sedation; 11 = deep sedation), and pain scores were monitored from 30 min before the surgery to 24 h after the end of anesthesia. All p values < 0.05 were defined as statistically significant. Thirty minutes before the surgery, the sedation scores were higher in BupreI (score = 10) compared to the BupreP group (score = 1). Moreover, during the mastectomy, the mean arterial pressure significantly increased in both groups even if nobody required additional analgesia. In the postoperative period, the pain scores did not show statistically significant differences between the two groups, maintaining values below the pain threshold at all times of the study. In conclusion, the transdermal administration of buprenorphine could guarantee an analgesic quality equal to that of the injectable route

    Amphiregulin contained in NSCLC-exosomes induces osteoclast differentiation through the activation of EGFR pathway

    Non-small cell lung cancer (NSCLC) remains the leading cause of cancer-related deaths worldwide. The majority of patients are diagnosed in advanced disease stage. Bone metastasis is the most frequent complication in NSCLC resulting in osteolytic lesions. The perfect balance between bone-resorbing osteoclasts and bone-forming osteoblasts activity is lost in bone metastasis, inducing osteoclastogenesis. In NSCLC, the epidermal growth factor receptor (EGFR) pathway is constitutively activated. EGFR binds Amphiregulin (AREG) that is overexpressed in several cancers such as colon, breast and lung. Its levels in plasma of NSCLC patients correlate with poor prognosis and AREG was recently found as a signaling molecule in exosomes derived from cancer cell lines. Exosomes have a key role in the cell-cell communication and they were recently indicated as important actors in metastatic niche preparation. In the present work, we hypothesize a role of AREG carried by exosomes derived from NSCLC in bone metastasis induction. We observed that NSCLC-exosomes, containing AREG, induce EGFR pathway activation in pre-osteoclasts that in turn causes an increased expression of RANKL. RANKL is able to induce the expression of proteolytic enzymes, well-known markers of osteoclastogenesis, triggering a vicious cycle in osteolytic bone metastasis

    Different substrates of non-sustained ventricular tachycardia in post-infarction patients with and without left ventricular dilatation

    Background: We investigated the relationship between nonsustained ventricular tachycardia (NSVT) and left ventricular (LV) dilatation, function, remodeling, and scar tissue extent in patients with previous myocardial infarction (MI). Methods and Results: Eighty-two patients (ages 64610 years) with first previous MI were referred for 24-hour electrocardiogram recording and cine and delayed enhancement (DE) cardiac magnetic resonance (CMR). LVvolumes, ejection fraction, systolic wall thickening, sphericity index, and core and peri-infarctual areas of scar tissue by CMR were evaluated. LV dilatation was observed in 39 patients. Episodes of NSVT were recorded in 32 patients: 23 with LV dilatation and 9 without. In the entire population, NSVTwas related to ejection fraction, LV volumes, LV mass, and sphericity index; end-systolic volume (P5.001) resulted in the only independent predictor at multivariate analysis. In patients without LV dilatation, the occurrence of NSVTwas only positively related with percentage of contracting segments with DE (P5.008). Conversely, in patients with LV dilatation, increase in LV mass (P5.020) and end-systolic volume (P5.038) were independent predictors of NSVT. Conclusions: Necrotic and viable myocardium coexistence within the same wall segments predicted occurrence of NSVT in patients without LV dilatation, whereas LV mass and end-systolic volume were predictors of NSVT in those with LV dilatation.     Quantitative analysis of late gadolinium enhancement in hypertrophic cardiomyopathy

    Background: Cardiovascular Magnetic resonance (CMR) with the late gadolinium enhancement (LGE) technique allows the detection of myocardial fibrosis in Hypertrophic cardiomyopathy (HCM). The aim of this study was to compare different methods of automatic quantification of LGE in HCM patients. Methods: Forty HCM patients (mean age 48 y, 30 males) and 20 normal subjects (mean age 38 y, 16 males) underwent CMR, and we compared 3 methods of quantification of LGE: 1) in the SD2 method a region of interest (ROI) was placed within the normal myocardium and enhanced myocardium was considered as having signal intensity>2 SD above the mean of ROI; 2) in the SD6 method enhanced myocardium was defined with a cut-off of 6 SD above mean of ROI; 3) in the RC method a ROI was placed in the background of image, a Rayleigh curve was created using the SD of that ROI and used as ideal curve of distribution of signal intensity of a perfectly nulled myocardium. The maximal signal intensity found in the Rayleigh curve was used as cut-off for enhanced myocardium. Parametric images depicting non enhanced and enhanced myocardium was created using each method. Three investigators assigned a score to each method by the comparison of the original LGE image to the respective parametric map generated. Results: Patients with HCM had lower concordance between the measured curve of distribution of signal intensity and the Rayleigh curve than controls (63.7 ± 12.3 % vs 92.2 ± 2.3%, p < 0.0001)

    Mitigating heat stress of dairy cows bred in a free-stall barn by sprinkler systems coupled with forced ventilation

    This paper presents the results of a research study which had the objective of investigating the effect of a sprinkler system coupled with forced ventilation on the heat stress of dairy cows bred in a free stall barn without paddock. To this aim, an experiment was carried out inside a free-stall dairy house equipped with two different cooling systems: a fogging system associated with forced ventilation in the resting area and a sprinkler system associated with forced ventilation in the feeding alley. The experiment regarded two adjacent pens of the barn and was constituted by three different trials carried out in the following periods: 27th June – 7th July (P1), 25th July – 4th August (P2), 24th August – 3rd September (P3). The experimental protocol of each trial required that the treatment group was housed in one pen where the two cooling systems were always activated following an established timetable, whereas the control group was housed in the other pen, where the sprinkler system associated with forced ventilation was always deactivated. Climatic parameters were measured inside each pen of the barn and outside. Then, thermal humidity index (THI) was calculated. Rectal temperature and respiration rate of a sample of dairy cows were monitored each day during the three periods considered (P1, P2 and P3). During the three trials the cows of both groups were subjected to climatic conditions that resulted in average daily THI values between 72.8 and 74.7, corresponding to mild or moderate heat stress. However, during daytime, air temperature and relative humidity reached values corresponding to a severe heat stress, as attested by the maximum THI values that were higher than or very close to 80. Furthermore, it was observed that the sprinklers do not influence the microclimatic conditions. However, the physiological parameters values of the treatment group were always significantly lower than the corresponding ones of the control group. Specifically, the system especially influenced the respiration rate that, in the treatment group, was close to 50 breath/min, while in the control group it reached 70 breath/min. The sprinkler system had more limited effects on rectal temperature that, however, in the treatment group was significantly lower than in the control group (38.7°C to 38.8°C vs 39.1°C 39.4°C) in all the three periods of the experiment. These results show that the sprinkler system could be useful to mitigate heat stress in dairy cows

    Quantitative comparison between amyloid deposition detected by (99m)Tc-Diphosphonate imaging and myocardial deformation evaluated by strain echocardiography in transthyretin-related cardiac amyloidosis.

    Methods and results 28 patients with transthyretin mutation and a group of 14 controls underwent echocardiography to quantify left ventricular (LV) dimensions and function, and global (G) longitudinal (L), radial (R) and circumferential (C) strain (S). 99mTc-3, 3-diphosphono-1, 2-propanodicarboxylic-acid-scintigraphy (99mTc-DPD) was used to quantify cardiac amyloidosis (CA). 99mTc-DPD revealed accumulation in 14 of 28 patients (CA-group) and no accumulation (no CA-group) in 14 patients. Cardiac accumulation was mild-moderate in 5 (Mild-Moderate CA-group) and severe in 9 patients (Severe CA-group). Severe CA-group showed higher values of LV septal thickness (LVST), posterior wall thickness and E/E' ratio than the no CA-group and the control group (adj. p<0.05). Ejection fraction was similar among groups (p=0.65). GLS was lower (p=<0.001) in severe CA-group (-12.2 ± 4.5) respect to no CA-group (-19.3 ± 3.0) and to the control group (-20.9 ± 2.5). On the contrary, GCS and GRS were lower (p=<0.05) in mild-moderate CA-group (-10.8 ± 4.1 and 9.5 ± 5.7, respectively) respect to the severe CA-group (-18.9 ± 5.1 and 23.9 ± 6.3 respectively), no CA-group (-19.2 ± 4.1 and 28.4 ± 10.2 respectively) and the control group (-23.9 ± 4.4 and 29.9 ± 8.7 respectively). A correlation was found between the scintigraphic heart retention (HR) index and LVST (r=0.72; p<0.001) and E/E' (r=0.46; p=0.03).
