    Initial diversification of living amphibians predated the breakup of Pangaea

    The origin and divergence of the three living orders of amphibians (Anura, Caudata, Gymnophiona) and their main lineages are one of the most hotly debated topics in vertebrate evolution. Here, we present a robust molecular phylogeny based on the nuclear RAG1 gene as well as results from a variety of alternative independent molecular clock calibrations. Our analyses suggest that the origin and early divergence of the three living amphibian orders dates back to the Palaeozoic or early Mesozoic, before the breakup of Pangaea, and soon after the divergence from lobe-finned fishes. The resulting new biogeographic scenario, age stimate, and the inferred rapid divergence of the three lissamphibian orders may account for the lack of fossils that represent plausible ancestors or immediate sister taxa of all three orders and the heretofore paradoxical distribution of some amphibian fossil taxa. Furthermore, the ancient and rapid radiation of the three lissamphibian orders likely explains why branch lengths connecting their early nodes are particularly short, thus rendering phylogenetic inference of implicated relationships especially difficult

    Cystoid Macular Edema: Causes, Diagnosis and Treatment

    The purpose of this paper is to conduct a review of studies on cystoid macular edema published in the last seven years. Cystoid macular edema is a major cause of loss of visual acuity. It is the final common pathway of many diseases and can be caused by numerous processes including inflammatory, vascular, adverse drug reactions, retinal dystrophy or intraocular tumors. These processes disrupt the blood-retinal barrier, with fluid extravasation to the macular parenchyma. Imaging tests are essential for both detection and monitoring of this pathology. Fluorescein angiography and autofluorescence show the leakage of liquid from perifoveal vessels into the tissue where it forms cystic spaces. Optical coherence tomography is currently the gold standard technique for diagnosis and monitoring. This allows objective measurement of retinal thickness, which correlates with visual acuity and provides more complete morphological information. Based on the underlying etiology, the therapeutic approach can be either surgical or medical with anti-inflammatory drugs. We found that disruption of the blood-retinal barrier for various reasons is the key point in the pathogenesis of cystoid macular edema, therefore we believe that studies on its treatment should proceed on this path

    Effects of regulated deficit irrigation on physiology, yield and fruit quality in apricot trees under Mediterranean conditions

    Scarce water resources mainly in arid and semi-arid areas have caused an increasing interest for applying irrigation protocols aiming to reduce water spends. The effects of regulated deficit irrigation (RDI) on the performance of apricot trees (Prunus armeniaca L. cv. “Búlida”) were assessed in Murcia (SE Spain), during three consecutive growing seasons (2008-2010). The hypothesis was that RDI would not restrict yield but increase fruit quality while saving water. Two irrigation treatments were established: i) control, irrigated to fully satisfy crop water requirements (100% ETc) and ii) RDI, that reduced the amount of applied water to: a) 40% of ETc at flowering and stage I of fruit growth; b) 60% of ETc during the stage II of fruit growth and c) 50% and 25% of ETc during the late postharvest period (from 60 days after harvest). Stem water potential, gas exchanges, trunk cross-sectional area (TCSA), fruit diameter, yield and fruit quality traits were determined. Vegetative growth was decreased by the use of RDI (12% less TCSA on average for the three years), whereas yield was unaffected. In addition, some qualitative characteristics of the fruits, such as the level of soluble solids, sweetness/acidity relation and fruit colour, were improved by the use of RDI. These results and average water savings of approximately 30%, lead us to conclude that RDI strategies are a possible solution for irrigation management in areas with water shortages, such as arid and semi-arid environments

    Maximizing transcription of nucleic acids with efficient T7 promoters

    In vitro transcription using T7 bacteriophage polymerase is widely used in molecular biology. Here, we use 5'RACE-Seq to screen a randomized initially transcribed region of the T7 promoter for cross-talk with transcriptional activity. We reveal that sequences from position +4 to +8 downstream of the transcription start site affect T7 promoter activity over a 5-fold range, and identify promoter variants with significantly enhanced transcriptional output that increase the yield of in vitro transcription reactions across a wide range of template concentrations. We furthermore introduce CEL-Seq(+) , which uses an optimized T7 promoter to amplify cDNA for single-cell RNA-Sequencing. CEL-Seq(+) facilitates scRNA-Seq library preparation, and substantially increases library complexity and the number of expressed genes detected per cell, highlighting a particular value of optimized T7 promoters in bioanalytical applications

    Tratamiento quirúrgico de la patologia urgente del colon izquierdo

    El objetivo de este estudio es evaluar la morbilidad y mortalidad postoperatoria tras la aplicación de un protocolo terapéutico en los pacientes intervenidos de urgencia por patología del colon izquierdo. Como primera opción se practicó el Lavado Anterogrado Peroperatorio en todas las lesiones del colon izquierdo que precisaron intervención urgente. En lesiones irresecables, peritonitis fecaloidea, deterioro del estado general o lesiones asociadas en el resto del colon. se aplicaron otras opciones: la colostomía de descarga, la operación de Hartmann o la colectomía subtotal. Se practicaron 127 resecciones urgentes del colon izquierdo. En 56 casos se optó por la operación de Hartmann. en 38 por la colectomía subtotal y en 33 por el lAP. La complicación más frecuente fue la sepsis abdominal (29%). La mortalidad operatoria global fue del 24%. 390Ai para la operación de Hartmann. 16% para la colectomía subtotal y 6% para el LAP. El LAP es la intervención de elección en aquellos pacientes con buen estado general que presentan patología urgente del colon izquierdo sin peritonitis fecaloidea. ni lesión irreversible del colon derecho

    The Ontogenetic Osteohistology of Tenontosaurus tilletti

    Tenontosaurus tilletti is an ornithopod dinosaur known from the Early Cretaceous (Aptian-Albian) Cloverly and Antlers formations of the Western United States. It is represented by a large number of specimens spanning a number of ontogenetic stages, and these specimens have been collected across a wide geographic range (from central Montana to southern Oklahoma). Here I describe the long bone histology of T. tilletti and discuss histological variation at the individual, ontogenetic and geographic levels. The ontogenetic pattern of bone histology in T. tilletti is similar to that of other dinosaurs, reflecting extremely rapid growth early in life, and sustained rapid growth through sub-adult ontogeny. But unlike other iguanodontians, this dinosaur shows an extended multi-year period of slow growth as skeletal maturity approached. Evidence of termination of growth (e.g., an external fundamental system) is observed in only the largest individuals, although other histological signals in only slightly smaller specimens suggest a substantial slowing of growth later in life. Histological differences in the amount of remodeling and the number of lines of arrested growth varied among elements within individuals, but bone histology was conservative across sampled individuals of the species, despite known paleoenvironmental differences between the Antlers and Cloverly formations. The bone histology of T. tilletti indicates a much slower growth trajectory than observed for other iguanodontians (e.g., hadrosaurids), suggesting that those taxa reached much larger sizes than Tenontosaurus in a shorter time
