    Nucleic acid i-motif structures in analytical chemistry

    Under the appropriate experimental conditions of pH and temperature, cytosine-rich segments in DNA or RNA sequences may produce a characteristic folded structure known as an i-motif. Besides its potential role in vivo, which is still under investigation, this structure has attracted increasing interest in other fields due to its sharp, fast and reversible pH-driven conformational changes. This 'on/off' switch at molecular level is being used in nanotechnology and analytical chemistry to develop nanomachines and sensors, respectively. This paper presents a review of the latest applications of this structure in the field of chemical analysis

    Optimization and performance criteria of a Stokes polarimeter based on two variable retarders

    In this paper we present the analysis, optimization and implementation of several Stokes polarimeter configurations based on a setup including two variable retarders. The polarimeter analysis is based on the Mueller-Stokes formalism, and as a consequence, it is suitable to deal with depolarized light. Complete Stokes polarimeters are optimized by minimizing the amplification of simulated errors into the final solution. Different indicators useful to achieve this aim, as the condition number or the equally weighted variance, are compared in this paper. Moreover, some of the optimized polarimeters are experimentally implemented and it is studied the influence of small deviations from the theoretical ones on the amplification of the Stokes component error.     Meditation practice is associated with a values-oriented life: the mediating role of decentering and mindfulness

    Clarification of personal values and meditation practice has been associated in most meditation traditions and in academic texts. Both values-related behavior and meditation practice increases well-being, but their relationship has not been well studied. It has been suggested that values, together with self-regulation, psychological flexibility, and exposure, are key mechanisms of action in mindfulness. The aims of this study were to explore the association between meditation and values and to examine the processes involved in this relation. A sample of 235 meditation-naĂŻve participants and 274 subjects with varying levels of experience in meditation practice completed an online assessment protocol composed of Five Facets of Mindfulness Questionnaire (FFMQ), Decentering Questionnaire (EQ), Valued Living Questionnaire (VLQ), and Engagement with Life Scale (ELS). Results revealed that daily meditators were more consistent, aware, and life-fulfilled about their values; moreover, these measures correlated with the mindfulness process and decentering. The relation between meditation practice and values-related behavior (assessed by the VLQ) was mediated by decentering. The association between meditation practice and Valued Living and Life Fulfillment (measured by the ELS) was mediated by the decentering, describing, and nonjudging dimensions of mindfulness. The findings in this study support the relation between meditation and personal values, mediated by the decentering, describing, and non-judging facets of mindfulness

    Changes of liver hemodynamic and elastography parameters in patients with colorectal liver metastases receiving preoperative chemotherapy: 'a note of caution'

    BACKGROUND: New systemic chemotherapy agents have improved prognosis in patients with colorectal liver metastases (CLM), but some of them damage the liver parenchyma and ultimately increase postoperative morbidity and mortality after liver resection. The aims of our study were to determine the degree of hemodynamic and pathological liver injury in CLM patients receiving preoperative chemotherapy and to identify an association between these injuries and postoperative complications after liver resection. METHODS: This is a prospective descriptive study of patients with CLM receiving preoperative chemotherapy before curative liver resection from November 2013 to June 2014. All patients had preoperative elastography and hepatic hemodynamic evaluation. We analyzed clinical preoperative data and postoperative outcomes after grouping the patients by chemotherapy type, development of sinusoidal obstructive syndrome (SOS), and development of major complications. RESULTS: Eleven from the 20 patients included in the study received preoperative oxaliplatin-based chemotherapy (OBC). Nine patients had SOS at pathological analysis and five patients developed major complications. Patients receiving preoperative OBC had higher values of hepatic venous pressure gradient (HVPG) and developed more SOS and major complications. Patients developing SOS had higher values of HVPG and developed more major complications. Patients with major complications had higher values of HVPG, and patients with a HVPG of 5 mmHg or greater had more major complications than those under 5 mmHg (20 vs 80%, p = 0.005). CONCLUSIONS: OBC and SOS impair liver hemodynamics in CLM patients. An increase in major complications after liver resection in these patients develops at subclinical HVPG levels

    MAGIC VHE Gamma-Ray Observations Of Binary Systems

    There are several types of Galactic sources that can potentially accelerate charged particles up to GeV and TeV energies. We present here the results of our observations of the source class of gamma-ray binaries and the subclass of binary systems known as novae with the MAGIC telescopes. Up to now novae were only detected in the GeV range. This emission can be interpreted in terms of an inverse Compton process of electrons accelerated in a shock. In this case it is expected that protons in the same conditions can be accelerated to much higher energies. Consequently they may produce a second component in the gamma-ray spectrum at TeV energies. The focus here lies on the four sources: nova V339 Del, SS433, LS I +61 303 and V404 Cygni. The binary system LS I +61 303 was observed in a long-term monitoring campaign for 8 years. We show the newest results on our search for superorbital variability, also in context with contemporaneous optical observations. Furthermore, we present the observations of the only super-critical accretion system known in our galaxy: SS433. Finally, the results of the follow-up observations of the microquasar V404 Cygni during a series of outbursts in the X-ray band and the ones of the nova V339 Del will be discussed in these proceedings.Comment: Proceedings of the 35th International Cosmic Ray Conference (ICRC 2017), Bexco, Busan, Korea (arXiv:1708.05153

    Specialist palliative and end-of-life care for patients with cancer and SARS-CoV-2 infection: a European perspective

Here, we report SPCT involvement during the first wave of the pandemic and compare outcomes for patients with cancer who received and did not receive SPCT input from multiple European cancer centres. Methods: From the OnCovid repository (N = 1318), we analysed cancer patients aged â©Ÿ18 diagnosed with COVID-19 between 26 February and 22 June 2020 who had complete specialist palliative care team data (SPCT+ referred; SPCT− not referred). Results: Of 555 eligible patients, 317 were male (57.1%), with a median age of 70 years (IQR 20). At COVID-19 diagnosis, 44.7% were on anti-cancer therapy and 53.3% had â©Ÿ1 co-morbidity. Two hundred and six patients received SPCT input for symptom control (80.1%), psychological support (54.4%) and/or advance care planning (51%). SPCT+ patients had more ‘Do not attempt cardio-pulmonary resuscitation’ orders completed prior to (12.6% versus 3.7%) and during admission (50% versus 22.1%, p < 0.001), with more SPCT+ patients deemed suitable for treatment escalation (50% versus 22.1%, p < 0.001). SPCT involvement was associated with higher discharge rates from hospital for end-of-life care (9.7% versus 0%, p < 0.001). End-of-life anticipatory prescribing was higher in SPCT+ patients, with opioids (96.3% versus 47.1%) and benzodiazepines (82.9% versus 41.2%) being used frequently for symptom control. Conclusion: SPCT referral facilitated symptom control, emergency care and discharge planning, as well as high rates of referral for psychological support than previously reported.     Resistive-based micro-kelvin temperature resolution for ultra-stable space experiments

    High precision temperature measurements are a transversal need in a wide area of physical experiments. Space-borne gravitational wave detectors are a particularly challenging case, requiring both high precision and high stability in temperature measurement. In this contribution, we present a design able to reach 1 ”K/Hz---v in most of the measuring band down to 1 mHz, and reaching 20 ”K/Hz---v at 0.1 mHz. The scheme is based on resistive sensors in a Wheatstone bridge configuration which is AC modulated to minimize the 1/f noise. As a part of our study, we include the design of a test bench able to guarantee the high stability environment required for measurements. We show experimental results characterising both the test bench and the read-out, and discuss potential noise sources that may limit our measurement.Peer ReviewedPostprint (published version

    Positron emission tomography assessment of large vessel inflammation in patients with newly diagnosed, biopsy-proven giant cell arteritis: a prospective, case-control study

    BACKGROUND: Positron emission tomography (PET) scan is emerging as a promising imaging technique to detect large-vessel inflammation in giant cell arteritis (GCA). However, the lack of a standardised definition of arteritis based on (18)fluorodeoxyglucose (FDG) uptake is an important limitation to the use of PET scan for diagnostic purposes. OBJECTIVE: To prospectively assess the intensity and distribution of FDG uptake at different vascular territories in patients with newly diagnosed GCA compared with controls. METHODS: 32 consecutive, biopsy-proven, GCA patients treated with glucocorticoids for ≀3 days were included. The control group consisted of 20 individuals, who underwent PET/CT for cancer staging. Maximal standardised uptake value (SUVm) was calculated at four aortic segments, supraaortic branches and iliac-femoral territory. Sensitivity and specificity was calculated by receiver-operator characteristic curves (ROC) analysis. RESULTS: Mean SUVm was significantly higher in patients than in controls in all vessels explored and correlated with acute-phase reactants and serum IL-6. Mean of the SUVm at all the vascular territories had an area under the curve (AUC) of 0.830, and a cut-off of 1.89 yielded a sensitivity of 80% and a specificity of 79% for GCA diagnosis. There were no significant differences in AUC among the vascular beds examined. CONCLUSIONS: FDG uptake by large vessels has a substantial sensitivity and specificity for GCA diagnosis

    Impact of fertilization with pig slurry on the isotopic composition of nitrate retained in soil and leached to groundwater in agricultural areas

    The isotopic composition of N and O of nitrate (NO3−) is usually employed to trace its sources of pollution in groundwater. In agricultural areas, the amount of NO3− that reaches the aquifers after fertilization is controlled by different transformation processes that can affect the nitrogen species isotopic composition. Aiming to address the reliability of using isotope tools to trace sources of groundwater NO3−, the goal of this study was to check the effect of fertilization on the isotopic composition of N compounds retained and leached from soils. The concentration and isotopic composition (ÎŽ15N and ÎŽ18O) of ammonium (NH4+), NO3− and nitrite (NO2−) was characterized after the application of pig slurry in lysimeters containing either soil under fallow (LF) or the same soil continuously cropped and fertilized (LC) during the previous six years. Results showed that the leached NO3− isotopic signature did not directly reflect the isotopic composition of the applied pig slurry. Just after fertilization, nitrification led to lower ÎŽ15NNO3 values in soil extracts and leachates (e.g. from +5.9 ± 0.9Âż to +3.8 ± 3.1Âż in soil extracts of LF lysimeters). These values increased after complete nitrification (+11.5 ± 1.3Âż) towards the ÎŽ15Nbulk of pig slurry (+19.6 ± 0.5Âż). Later on, due to soil organic matter and plant debris mineralization and subsequent nitrification, values decreased towards the initial ÎŽ15NNO3 of soil but remained above them (+8.6 ± 1.0Âż). Both LF and LC experiments showed a similar trend and the latter ones allowed to reinforce that long-term fertilization with pig slurry can increase the soil ÎŽ15NNO3. Concerning the ÎŽ18O of NO3− from soil extracts and leachates, it mainly depended on the ÎŽ18O of irrigation water and oxygen, after nitrification of NH4+ from pig slurry. Therefore, studies aiming to trace groundwater NO3− pollution sources in rural areas by using an isotopic approach should consider the fertilization history of each setting. Also, analyzing the ÎŽ15Nbulk of soil is recommended, since it could mask the isotopic signature of the N applied through fertilization
