Effects of α-synuclein levels on cerebral synaptic function: Validation of a novel PET radioligand for the early diagnosis of Parkinson’s disease

Background In Parkinson’s disease, converging evidence supports a pathogenic role for excessive α–synuclein accumulation in synaptic terminals that may propagate back to the soma of vulnerable nerve cells such as neurons in the substantia nigra pars compacta. The resulting loss of dopaminergic terminals in the striatum can be demonstrated in vivo using 18F-Dopa-PET (positron emission tomography). However, there’s currently no validated biomarker of the progressive synaptic dysfunction in other vulnerable areas such as the cerebral cortex. Goal In this longitudinal study, we will test the hypothesis that the loss of synaptic terminals in a mouse model of excessive α–synuclein accumulation can be demonstrated in vivo before the occurrence of behavioural disturbances using 18F-UCB-H, a new PET biomarker developed at CRC. We will also test if this new imaging modality is sensitive enough to study the effect of a disease modifying therapy such as chronic physical exercise. Methods We will use microPET for the in vivo quantification of 18F-UCB-H brain uptake in 16 wild type animals and 16 transgenic (Tg) mice overexpressing human α–syn under the mThy1 promotor every 2 months. Data will be validated against post-mortem analyses after the last PET study. Predictions We predict decreased tracer uptake over time in the basal ganglia and cerebral cortex in Tg mice as compared with WT animals. Also, we predict a relationship between 18F-UCB-H uptake levels in basal ganglia and cerebral cortex and progressive alterations in both motor and cognitive functions, respectively. Further, we also expect that chronic exercise will slow down both motor and cognitive disturbances, as well as the rate of 18F-UCB-H brain uptake decreases. Conclusion If 18F-UCB-H PET proves to be a valid biomarker for the early detection of α–synuclein accumulation in the pre-clinical model of PD, the methods will tested on human clinical populations

REPRESENTAÇÕES DO BRASIL E DO BRASILEIRO NO DISCURSO DO JORNAL ESPANHOL EL PAÍS NO CONTEXTO PRÉ-COPA FIFA DE 2014

A produção de discursos da mídia internacional sobre o cenário político do Brasil esteve em efervescência nos últimos anos devido aos diversos megaeventos esportivos sediados no país, desde a Copa das Confederações, em 2013, e da Copa do Mundo, em 2014, até os Jogos Olímpicos, em 2016, os quais proporcionaram grande visibilidade ao país no exterior. Este trabalho objetiva analisar artigos de opinião do jornal espanhol El País, veiculados entre 2013 e 2014, a fim de observar as estruturas discursivas presentes quanto às representações e estereótipos do Brasil e do brasileiro no contexto das manifestações políticas daquele período. Para tanto, utilizaremos os pressupostos da Análise Crítica do Discurso (ACD), especialmente os trabalhos de Van Dijk (2008; 2012), em diálogo com estudiosos das Ciências Sociais (BOURDIEU, 1989), da Psicologia Social (MOSCOVICI, 1978; 2004), dos Estudos Culturais (HALL, 2000; 2011) e demais autores que contribuíram para a constituição de um panorama sócio-histórico do Brasil (ZWEIG, 2013; AGASSIZ, 1975; RAEDERS, 1988) etc., o que caracteriza um viés multidisciplinar a partir da Linguística. Encontramos na Análise Crítica do Discurso (ACD) nossa abordagem, uma vez que, enquanto perspectiva crítica que tem por finalidade principal o exame das relações entre discurso e poder, esta nos permitirá indagar uma possível polarização intergrupal entre europeus e brasileiros e a consequente (re) produção de representações desfavoráveis do Brasil e do brasileiro e a existência de uma relação hierarquizada entre culturas, a partir do discurso jornalístico espanhol

This study has been started in 1980 under the Agence Française pour la Maîtrise de l’Energie, the Plan Construction and the Secrétariat à la Recherche en Architecture. Relationship between vegetal environment and architecture has been examined by a team of architects, landscape gardeners and geographers taking the prevailing micro climates into consideration. National audienceL'enlèvement de particules par laser impulsionnel (ns) est étudié expérimentalement. Des efficacités supérieures à 90% sont obtenues pour l'enlèvement de particules de polystyrene (PS) de diamètre 385 nm sur des surfaces de Si et Cu. Les fluences seuils d'enlèvement sont déterminées et comparées à une modélisation de l'expansion thermique du substrat.
