    Comparison between the estimated of nonparametric methods by using the methodology of quantile regression models

    This paper study two stratified quantile regression models of the marginal and the conditional varieties. We estimate the quantile functions of these models by using two nonparametric methods of smoothing spline (B-spline) and kernel regression (Nadaraya-Watson). The estimates can be obtained by solve nonparametric quantile regression problem which means minimizing the quantile regression objective functions and using the approach of varying coefficient models. The main goal is discussing the comparison between the estimators of the two nonparametric methods and adopting the best one between them

    A study of Immunoglobulins,Complements and Some Hematological Parameters Levels in Thalassemic Patients in Related Bacterial Infections

    Background: Thalassemia and abnormal hemoglobin are the most common genetic disorders and are considered health problems in many developing countries. Beta-Thalassemia major is the most familiar type, in which the beta-globin chain synthesis is impaired. Objective: To evaluation the concentrations of immunoglobulins  IgM, IgG and  IgA also the complements  C3 and C4 between β-Thalassimic children patients and control group, and find the differences of some hematological parameters between the two  groups, finally, isolation of some bacterial species causes the bacterial infections. Patients and Methods: The present study was performed on 40 β-thalassemia major children who were registered in the thalassemia center at Al-Karama teaching hospital in Baghdad and 20 healthy children as a control group from the period of September 2015-Augest 2016, the serum immunoglobulins (IgG, IgM, IgA) and complement C3, C4 levels were measured also blood culture was done to identify the causative infection. Results: The mean serum concentrations of IgM, IgG and IgA in β-thalassemia major children were 1834.1 ± 312.2, 134.5 ± 40.3 and 212.7±89.6, respectively, and they were increased significantly (p≤0.05). The mean serum concentrations of C3 and C4 were consistently decreased in children and there were no significant differences (p≤0.05) between the two groups. The mean Hb of β- thalassemic children was 8.32±0.91 g/dl  and this lower from the level of control, RBC count is also low 3.7±0.7 but the levels of  WBCs  was higher in β- thalassemic children 10.4±1.67, several types of bacterial specious were isolated included  Staphylococcus aureus 30% (12 patients) , Escherichia coli 25%(10 patients) , Streptococcus pneumoniae 17.5% (7 patients), Salmonella enteritidis 10% (4 patients), Serratia  marcescens  7.5% (3 patients), Pseudomonas aeruginosa 5%(2 patients), Klebsiella pneumoniae 2.5%(1 patients) and Haemophilus influenza 2.5%(1patients).  Conclusion: The alteration in serum immunoglobulins and complements levels in thalassemia major children probably can be due to marked heterogeneity of the patients,

    Barriers to the delivery of diabetes care in the Middle East and South Africa: a survey of 1,082 practising physicians in five countries

    SUMMARY Aims: Developing countries face a high and growing burden of type 2 diabetes. We surveyed physicians in a diverse range of countries in the Middle East and Africa (Egypt, Kingdom of Saudi Arabia, United Arab Emirates, South Africa and Lebanon) with regard to their perceptions of barriers to type 2 diabetes care identified as potentially important in the literature and by the authors. Methods: One thousand and eighty-two physicians completed a questionnaire developed by the authors. Results: Most physicians enroled in the study employed guideline-driven care; 80–100% of physicians prescribed metformin (with lifestyle intervention, where there are no contraindications) for newly diagnosed type 2 diabetes, with lifestyle intervention alone used where metformin was not prescribed. Sulfonylureas were prescribed widely, consistent with the poor economic status of many patients. About one quarter of physicians were not undertaking any form of continuing medical education, and relatively low proportions of practices had their own diabetes educators, dieticians or diabetic foot specialists. Physicians identified the deficiencies of their patients (unhealthy lifestyles, lack of education and poor diet) as the most important barriers to optimal diabetes care. Low-treatment compliance was not ranked highly. Access to physicians did not appear to be a problem, as most patients were seen multiple times per year. Conclusions: Physicians in the Middle East and South Africa identified limitations relating to their patients as the main barrier to delivering care for diabetes, without giving high priority to issues relating to processes of care delivery. Further study would be needed to ascertain whether these findings reflect an unduly physician-centred view of their practice. More effective provision of services relating to the prevention of complications and improved lifestyles may be needed. What's known It is known that the success of care for diabetes depends critically on the delivery of optimised care for diabetic patients. Many barriers to the delivery of such care have been identified. Relatively little is known regarding how these barriers influence the delivery of diabetes care in the Middle East and South Africa. What's new Physicians generally followed management guidelines in type 2 diabetes care. Perceived barriers to optimal diabetes care mainly focussed on attributes of patients, rather than process issues in care or aspects of the physicians' practice

    Music for Thought: Examining Saudi Identities Expressed Through Music on Social Media

    The aim of this chapter is to investigate how social media help create and diffuse popular music in Saudi Arabia and how such music questions the officially accepted identities and social practices. Social media are widely used as means of promotion among artists around the world; yet Saudi Arabia deserves special attention. This is because traditional media act much more strictly as gatekeepers in the kingdom; hence, social media offer greater possibility for circulation of popular entertainment, especially when it touches upon the questions related to the core values of the Saudi society. Such is the case of recordings by Hesham Fageeh “No woman, No drive” (2013) or the recently trending video clip “Hwages” that, both, received peak visibility online and sparked debates about women empowerment. Similarly, some videos trending online prompted a dance craze, for instance, “Barbs” showed a choreography that was subsequently copied by fans who uploaded their performances online. Dancing is a prohibition among the conservative segments of society; yet, the vastly young population of Saudi Arabia seeks entertainment.Social media in Saudi Arabia have become primordial means of communication in the kingdom with highest rates of social media participation in the Middle East and seventh rank in the world (2016). Consequently, this chapter employs a mixed-method approach in order to uncover how social media breaks the monopoly on entertainment by promoting different musical genres and encouraging social debate

    Generation and characterisation of Friedreich ataxia YG8R mouse fibroblast and neural stem cell models

    This article has been made available through the Brunel Open Access Publishing Fund.Background: Friedreich ataxia (FRDA) is an autosomal recessive neurodegenerative disease caused by GAA repeat expansion in the first intron of the FXN gene, which encodes frataxin, an essential mitochondrial protein. To further characterise the molecular abnormalities associated with FRDA pathogenesis and to hasten drug screening, the development and use of animal and cellular models is considered essential. Studies of lower organisms have already contributed to understanding FRDA disease pathology, but mammalian cells are more related to FRDA patient cells in physiological terms. Methodology/Principal Findings: We have generated fibroblast cells and neural stem cells (NSCs) from control Y47R mice (9 GAA repeats) and GAA repeat expansion YG8R mice (190+120 GAA repeats). We then differentiated the NSCs in to neurons, oligodendrocytes and astrocytes as confirmed by immunocytochemical analysis of cell specific markers. The three YG8R mouse cell types (fibroblasts, NSCs and differentiated NSCs) exhibit GAA repeat stability, together with reduced expression of frataxin and reduced aconitase activity compared to control Y47R cells. Furthermore, YG8R cells also show increased sensitivity to oxidative stress and downregulation of Pgc-1α and antioxidant gene expression levels, especially Sod2. We also analysed various DNA mismatch repair (MMR) gene expression levels and found that YG8R cells displayed significant reduction in expression of several MMR genes, which may contribute to the GAA repeat stability. Conclusions/Significance: We describe the first fibroblast and NSC models from YG8R FRDA mice and we confirm that the NSCs can be differentiated into neurons and glia. These novel FRDA mouse cell models, which exhibit a FRDA-like cellular and molecular phenotype, will be valuable resources to further study FRDA molecular pathogenesis. They will also provide very useful tools for preclinical testing of frataxin-increasing compounds for FRDA drug therapy, for gene therapy, and as a source of cells for cell therapy testing in FRDA mice. © 2014 Sandi et al

    Quality-of-life among women with breast cancer: application of the international classification of functioning, disability and health model

    BackgroundThis study aimed to identify the factors that influence Breast Cancer (BC) women’s quality of life (QoL) based on the International Classification of Functioning, Disability and Health (ICF) framework.MethodA cross-sectional study was conducted among 188 women with BC. The dependent variable, QoL, was measured using the Quality of Life Index (QLI-c). The independent variables were measured using the following Arabic-validated questionnaires: Pittsburgh Sleep Quality Index (PSQI), Female Sexual Function Index (FSFI), Modified Fatigue Impact Scale (MFIS), Hospital Anxiety and Depression Scale (HADS), and the International Physical Activity Questionnaire (IPAQ).ResultsThere was a significant positive correlation between monthly income (r = 0.17, p = 0.016) and QoL, and significant negative correlation between the stage of disease (r = −0.221, p = 0.002) and duration of first diagnosis (r = −0.280, p = 0.004) with QoL. Poor sleep quality, sexual dysfunction, fatigue, depression, and anxiety had significant negative correlations with QoL (p < 0.01). Multiple regression analysis revealed that among the various factors that might affect QoL, sexual dysfunction, poor sleep quality, depression, and anxiety were significant predictors of QoL (p ≤ 0.05).ConclusionThe ICF provided an excellent framework to explore the factors influencing QoL among women with BC. This study has given evidence for the relationship of demographic, clinical, and body functional factors with QoL among women with BC. Interestingly, sexual dysfunction, poor sleep quality, depression, and anxiety factors are predictors of QoL. Awareness of these factors that predict QoL will guide healthcare professionals to improve the health and QoL of BC women

    Tissue Proteome of 2-Hydroxyacyl-CoA Lyase Deficient Mice Reveals Peroxisome Proliferation and Activation of ω-Oxidation

    Peroxisomal fatty acid α-oxidation is an essential pathway for the degradation of β-carbon methylated fatty acids such as phytanic acid. One enzyme in this pathway is 2-hydroxyacyl CoA lyase (HACL1), which is responsible for the cleavage of 2-hydroxyphytanoyl-CoA into pristanal and formyl-CoA. Hacl1 deficient mice do not present with a severe phenotype, unlike mice deficient in other α-oxidation enzymes such as phytanoyl-CoA hydroxylase deficiency (Refsum disease) in which neuropathy and ataxia are present. Tissues from wild-type and Hacl1−/− mice fed a high phytol diet were obtained for proteomic and lipidomic analysis. There was no phenotype observed in these mice. Liver, brain, and kidney tissues underwent trypsin digestion for untargeted proteomic liquid chromatography-mass spectrometry analysis, while liver tissues also underwent fatty acid hydrolysis, extraction, and derivatisation for fatty acid gas chromatography-mass spectrometry analysis. The liver fatty acid profile demonstrated an accumulation of phytanic and 2-hydroxyphytanic acid in the Hacl1−/− liver and significant decrease in heptadecanoic acid. The liver proteome showed a significant decrease in the abundance of Hacl1 and a significant increase in the abundance of proteins involved in PPAR signalling, peroxisome proliferation, and omega oxidation, particularly Cyp4a10 and Cyp4a14. In addition, the pathway associated with arachidonic acid metabolism was affected; Cyp2c55 was upregulated and Cyp4f14 and Cyp2b9 were downregulated. The kidney proteome revealed fewer significantly upregulated peroxisomal proteins and the brain proteome was not significantly different in Hacl1−/− mice. This study demonstrates the powerful insight brought by proteomic and metabolomic profiling of Hacl1−/− mice in better understanding disease mechanism in fatty acid α-oxidation disorders

    Larvicidal activities of 2-Aryl-2,3-Dihydroquinazolin -4-ones against malaria vector Anopheles arabiensis, In Silico ADMET prediction and molecular target investigation

    Malaria, affecting all continents, remains one of the life-threatening diseases introduced by parasites that are transmitted to humans through the bites of infected Anopheles mosquitoes. Although insecticides are currently used to reduce malaria transmission, their safety concern for living systems, as well as the environment, is a growing problem. Therefore, the discovery of novel, less toxic, and environmentally safe molecules to effectively combat the control of these vectors is in high demand. In order to identify new potential larvicidal agents, a series of 2-aryl-1,2-dihydroquinazolin-4-one derivatives were synthesized and evaluated for their larvicidal activity against Anopheles arabiensis. The in silico absorption, distribution, metabolism, excretion, and toxicity (ADMET) properties of the compounds were also investigated and most of the derivatives possessed a favorable ADMET profile. Computational modeling studies of the title compounds demonstrated a favorable binding interaction against the acetylcholinesterase enzyme molecular target. Thus, 2-aryl-1,2-dihydroquinazolin-4-ones were identified as a novel class of Anopheles arabiensis insecticides which can be used as lead molecules for the further development of more potent and safer larvicidal agents for treating malaria.Fil: Venugopala, K.
