    FLASH: ultra-fast protocol to identify RNA-protein interactions in cells

    Determination of the in vivo binding sites of RNA-binding proteins (RBPs) is paramount to understanding their function and how they affect different aspects of gene regulation. With hundreds of RNA-binding proteins identified in human cells, a flexible, high-resolution, high-throughput, highly multiplexible and radioactivity-free method to determine their binding sites has not been described to date. Here we report FLASH (Fast Ligation of RNA after some sort of Affinity Purification for High-throughput Sequencing), which uses a special adapter design and an optimized protocol to determine protein-RNA interactions in living cells. The entire FLASH protocol, starting from cells on plates to a sequencing library, takes 1.5 days. We demonstrate the flexibility, speed and versatility of FLASH by using it to determine RNA targets of both tagged and endogenously expressed proteins under diverse conditions in vivo

    Dichromacy: Color Vision Impairment and Consanguinity in Heterogenous Population of Pakistan

    Background and Objectives: Dichromacy, an X-linked recessive disorder is identified worldwide, more in males than females. In European Caucasians, its incidence is 8% in males and 0.5% in females. In India, it is 8.73% in males and 1.69% in females, and in Iran, it is 8.18% in males and 0.43% in females. Population based epidemiological data about dichromacy in different ethnic groups in Pakistan is not available. The aim of this study was to find out the population prevalence of inherited red-green dichromacy in a heterogenous population of the district of Chiniot, Punjab, Pakistan, and to determine the impact of consanguinity and ethnicity. Methods: In this cross-sectional study, boys and girls of the higher secondary schools were examined in the three tehsils of district Chiniot. Pseudoisochromatic Ishihara Test has been employed for detection of dichromacy in the study population. The sample size was calculated statistically as 260, which was expanded to 705 and divided by population density of the three tehsils. Results: Screening of 359 males and 346 females revealed 19 (5.29%) dichromat males and only 2 (0.58%) females. The study population belonged to 23 castes / isonym groups. The consanguinity found in the district of Chiniot is 84.82% and in the dichromat families, it is 85.71%, of which 52.37% are first cousin.     COX-2 inhibitors: a potential target for drug therapy in the management of colorectal cancer

    Colorectal cancer is currently the third most common cancer in Malaysia. Elevated expression of COX-2, an induced cyclooxygenase isoenzyme, has been seen in colonic adenomas and colorectal carcinoma. There is evidence that inhibition of this COX-2 can decrease the risk of colorectal cancer. Selective COX-2 inhibitors may have a role in reducing the risk of colorectal cancer in high-risk individuals

    Frequency of Clinical Symptoms of Gastroesophageal Reflux Disease in Asthmatic Patients

    Background: Gastroesophageal reflex is known as an acid reflex, is long term condition where stomach contents back into the oesophagus resulting in either symptoms or complications. GERD disease is caused by weakness or failure of the lower oesophageal sphincter. Symptoms include the acidic taste behind the mouth, heart burn, chest pain, difficult breathing and vomiting. Complication includes esophagitis, oesophageal strictures and barrettes oesophagus. Objective: The aim of this research was to introduce the symptoms of GERD disease in asthmatic patients and how these symptoms worsen the symptoms of asthma disease and what clinical pictures present with the asthmatic disease. Methodology: A designed performa was used to collect the data and after filling the performa, results were drawn and conclusion through the facts and the information given by patients. Results: In the present study among all 164 asthmatic patients, 70 (42.7%) patients showed dyspepsia, 58 (35.4%) were with chest burning, 23 (14%) were asking about chest pain, with acidic mouth taste were 39 (23.8%), 22 (13.4%) were feeling sore throat and 44 (26.8%) showed regurgitation reflex. Among these 164 patients 16 (9.8%) were smokers and 148 (90.2 %) were non-smokers. 47 (28.7%) were males and 117 (71.3%) were females. Conclusion: It is concluded that gastroesophageal reflux disease in asthmatic patients present symptoms of acidic mouth taste, chest burning, chest pain, dyspepsia, regurgitation reflex and sore throat

    First-principles data for solid-solution strengthening of magnesium: From geometry and chemistry to properties

    Solid-solution strengthening results from solutes impeding the glide of dislocations. Existing theories of strength rely on solute-dislocation interactions, but do not consider dislocation core structures, which need an accurate treatment of chemical bonding. Here, we focus on strengthening of Mg, the lightest of all structural metals and a promising replacement for heavier steel and aluminum alloys. Elasticity theory, which is commonly used to predict the requisite solute-dislocation interaction energetics, is replaced with quantum-mechanical first-principles calculations to construct a predictive mesoscale model for solute strengthening of Mg. Results for 29 different solutes are displayed in a "strengthening design map" as a function of solute misfits that quantify volumetric strain and slip effects. Our strengthening model is validated with available experimental data for several solutes, including Al and Zn, the two most common solutes in Mg. These new results highlight the ability of quantum-mechanical first-principles calculations to predict complex material properties such as strength.Comment: 9 pages, 7 figures, 2 table

    A study of residual stresses in low alloy steel theta ring castings.

    Residual stresses generated during the casting and heat treatment of a low alloy steel, BW2 have been studied using ta theta ring so that temperature differentials could be varied using different tie bar sizes. Residual stresses have been measured using centre hole drilling and tie bar sectioning techniques. Centre hole drilling was shown to be sensitive to surface preparation methods. Stresses induced by drilling were accounted for in measured stresses and drilling stresses were found to be greater in cast than annealed samples. Cast theta rings have been shown to have compressive residual stresses, becoming less compressive or tensile as the tie bar width was reduced. Tie bar sectioning producedexpansion which increased with increasing tie bar width, although there was no direct correlation between tie bar stress and width. Results from both techniques have been explained using factors contributing to residual stress formation. The S-shaped runner contained residual stresses and its removal altered residual stress levels.Heat treatments have been found to produce different amounts of stress relief according to the geometry and thermal cycle imposed. In a uniform section theta.ring normalising and tempering relieved stresses in the tie bar but not in the outer ring, and the tie bar contained no stresses after sectioning. For the non-uniform section theta ring annealing made stresses more compressive whilst normalising and tempering generated stresses due to differential cooling, and the tie bar contained compressive stresses after sectioning. Maximum Von Mises equivalent in the cast theta ring was shown to be 25% of the yield strength and reduced further after heat treatment.A computational model based on finite difference has been used to simulate solidification of a tie bar across the width. Temperature gradients computed along this bar axis were shown to be small and are believed not to significantly contribute to residual stresses
