    Microscopic Observation of Plastic Deformation of Polycrystalline Aluminum by Laser Scanning Microscope

    Free surface of polycrystalline metal becomes roughened after plastic deformation. The surface roughening is closely related to the inhomogeneity of polycrystalline metals, that is, to the inhomogeneous plastic deformation of respective grains. In the present study, inhomogeneous deformation on the free surface of polycrystalline aluminum specimen during uniaxial tension is studied. The inhomogeneous deformation of grains in the central area of the free surface of specimen is observed by the laser scanning microscope, while the inhomogeneous deformation perpendicular to the surface is studied by the laser scanning microscope as well as the stylus measuring instrument. It is shown that the surface roughness and the strain of respective grains increase with the applied strain. Discussions are made on the change in the surface roughness, the strain in each grain and the slip-line angles with the applied strain

    Real-Time PCR-Based Analysis of the Human Bile MicroRNAome Identifies miR-9 as a Potential Diagnostic Biomarker for Biliary Tract Cancer

    Biliary tract cancer (BTC) is often difficult to diagnose definitively, even through histological examination. MicroRNAs (miRNAs) regulate a variety of physiological processes. In recent years, it has been suggested that profiles for circulating miRNAs, as well as those for tissue miRNAs, have the potential to be used as diagnostic biomarkers for cancer. The aim of this study was to confirm the existence of miRNAs in human bile and to assess their potential as clinical biomarkers for BTC. We sampled bile from patients who underwent biliary drainage for biliary diseases such as BTC and choledocholithiasis. PCR-based miRNA detection and miRNA cloning were performed to identify bile miRNAs. Using high-throughput real-time PCR-based miRNA microarrays, the expression profiles of 667 miRNAs were compared in patients with malignant disease (n = 9) and age-matched patients with the benign disease choledocholithiasis (n = 9). We subsequently characterized bile miRNAs in terms of stability and localization. Through cloning and using PCR methods, we confirmed that miRNAs exist in bile. Differential analysis of bile miRNAs demonstrated that 10 of the 667 miRNAs were significantly more highly expressed in the malignant group than in the benign group at P<0.0005. Setting the specificity threshold to 100% showed that some miRNAs (miR-9, miR-302c*, miR-199a-3p and miR-222*) had a sensitivity level of 88.9%, and receiver-operating characteristic analysis demonstrated that miR-9 and miR-145* could be useful diagnostic markers for BTC. Moreover, we verified the long-term stability of miRNAs in bile, a characteristic that makes them suitable for diagnostic use in clinical settings. We also confirmed that bile miRNAs are localized to the malignant/benign biliary epithelia. These findings suggest that bile miRNAs could be informative biomarkers for hepatobiliary disease and that some miRNAs, particularly miR-9, may be helpful in the diagnosis and clinical management of BTC

    Investigation on Ignition Coil Specification for Dilution Combustion System

    To improve thermal efficiency of internal combustion engine, dilution combustion system, such as lean burn and Exhaust Gas Recirculation (EGR) system, have been developed with spark ignition coils generating large discharge current and energy. Several researches have clarified that large discharge current increases discharge channel stretch and decreases possibility of discharge channel blow-off and misfire. However, these investigations don\u27t mention effect of discharge current profile on combustion speed and discharge channel behaviour enough. Purpose of this research is to investigate relation among dilution rate, combustion speed, discharge channel behaviour and discharge current. To achieve this purpose, five coils having different current profiles were evaluated by combustion test and in-cylinder optical measurement test with research single cylinder engine. The combustion test results showed a correlation between dilution limit and initial combustion period. And optical measurement test results showed a correlation of initial combustion period with discharge channel stretch. Moreover, saturation of discharge channel stretch were observed from a certain discharge current value on up. Based on these results, adequate coil for dilution system was selected. Finally, the coil was equipped on a vehicle and performance test was conducted. The vehicle with LP-EGR system was stably driven with 18% of EGR rate, and 2.3% of fuel reduction rate were verified in the WLTC mode. The selected high energy ignition coil was contributing reduction of misfire during EGR operation

    Drosophila Blimp-1 Is a Transient Transcriptional Repressor That Controls Timing of the Ecdysone-Induced Developmental Pathway▿

    Regulatory mechanisms controlling the timing of developmental events are crucial for proper development to occur. ftz-f1 is expressed in a temporally regulated manner following pulses of ecdysteroid and this precise expression is necessary for the development of Drosophila melanogaster. To understand how insect hormone ecdysteroids regulate the timing of FTZ-F1 expression, we purified a DNA binding regulator of ftz-f1. Mass spectroscopy analysis revealed this protein to be a fly homolog of mammalian B lymphocyte-induced maturation protein 1 (Blimp-1). Drosophila Blimp-1 (dBlimp-1) is induced directly by 20-hydroxyecdysone, and its product exists during high-ecdysteroid periods and turns over rapidly. Forced expression of dBlimp-1 and RNA interference analysis indicate that dBlimp-1 acts as a repressor and controls the timing of FTZ-F1 expression. Furthermore, its prolonged expression results in delay of pupation timing. These results suggest that the transient transcriptional repressor dBlimp-1 is important for determining developmental timing in the ecdysone-induced pathway
