    Purification and characterization of the first recombinant bird pancreatic lipase expressed in Pichia pastoris: The turkey

    <p>Abstract</p> <p>Background</p> <p>The turkey pancreatic lipase (TPL) was purified from delipidated pancreases. Some biochemical properties and kinetic studies were determined using emulsified system and monomolecular film techniques. Those studies have shown that despite the accumulation of free fatty acids at the olive oil/water interface, TPL continues to hydrolyse efficiently the olive oil and the TC<sub>4 </sub>in the absence of colipase and bile salts, contrary to most classical digestive lipases which denaturate rapidly under the same conditions. The aim of the present study was to express TPL in the methylotrophic yeast <it>Pichia pastoris </it>in order to get a large amount of this enzyme exhibiting interesting biochemical properties, to purify and characterize the recombinant enzyme.</p> <p>Results</p> <p>The recombinant TPL was secreted into the culture medium and the expression level reached about 15 mg/l after 4 days of culture. Using Q-PCR, the number of expression cassette integrated on <it>Pichia </it>genomic DNA was estimated to 5. The purified rTPL, with molecular mass of 50 kDa, has a specific activity of 5300 U/mg on emulsified olive oil and 9500 U/mg on tributyrin. The optimal temperature and pH of rTPL were 37°C and pH 8.5. The stability, reaction kinetics and effects of calcium ions and bile salts were also determined.</p> <p>Conclusions</p> <p>Our results show that the expressed TPL have the same properties as the native TPL previously purified. This result allows us the use of the recombinant enzyme to investigate the TPL structure-function relationships.</p

    Primary resistance to clarithromycin, metronidazole and amoxicillin of Helicobacter pylori isolated from Tunisian patients with peptic ulcers and gastritis: a prospective multicentre study

    <p>Abstract</p> <p>Background</p> <p>The frequency of primary resistance to antibiotics in H. pylori isolates is increasing worldwide. In Tunisia, there are limited data regarding the pattern of H. pylori antibiotic primary resistance.</p> <p>Aim</p> <p>To evaluate the primary resistance of H. pylori to clarithromycin, metronidazole and amoxicillin and to detect the mutations involved in clarithromycin resistance.</p> <p>Materials and methods</p> <p>273 strains isolated from adults and children were enrolled. The primary resistance to clarithromycin, metronidazole and amoxicillin was evaluated by means of E-test minimal inhibitory concentration (MIC). The real-time PCR using Scorpion primers was performed in all cases to assess clarithromycin primary resistance and point mutations involved.</p> <p>Results</p> <p>No resistance to amoxicillin was detected. For adults, resistance to clarithromycin and metronidazole was found respectively in 14.6% and 56.8%, and respectively in 18.8% and 25% in children. Overall, the rates of global primary resistance to clarithromycin and metronidazole in Tunisia were respectively determined in 15.4% and 51.3%.</p> <p>By the use of Scorpion PCR, the A2143G was the most frequent point mutation observed (88.1%), followed by the A2142G (11.9%); the A2142C was not found and 18 of 42 patients (42.8%) were infected by both the resistant and the susceptible genotype.</p> <p>The association of clarithromycin resistance with gender was not statistically significant, but metronidazole resistant strains were isolated more frequently in females (67.8%) than in males (32.2%) and the difference was significant. As for gastroduodenal diseases, the difference between strains isolated from patients with peptic ulceration and those with non peptic ulceration was not statistically significant. When about the distribution of resistant strains to clarithromycin and metronidazole between the three Tunisian cities (Tunis, Menzel Bourguiba and Mahdia), the difference was not statistically significant.</p> <p>Conclusion</p> <p>Local data regarding the primary resistance of H. pylori to clarithromycin, metronidazole and amoxicillin and the main genetic mutation involved in clarithromycin resistance in vivo (A2143G) are necessary to prove a clear need for a periodic evaluation of antibiotic consumption and new therapeutic strategies in Tunisia in order to avoid the emergence of resistant strains.</p

    Neglected Tropical Diseases of the Middle East and North Africa: Review of Their Prevalence, Distribution, and Opportunities for Control

    The neglected tropical diseases (NTDs) are highly endemic but patchily distributed among the 20 countries and almost 400 million people of the Middle East and North Africa (MENA) region, and disproportionately affect an estimated 65 million people living on less than US$2 per day. Egypt has the largest number of people living in poverty of any MENA nation, while Yemen has the highest prevalence of people living in poverty. These two nations stand out for having suffered the highest rates of many NTDs, including the soil-transmitted nematode infections, filarial infections, schistosomiasis, fascioliasis, leprosy, and trachoma, although they should be recognized for recent measures aimed at NTD control. Leishmaniasis, especially cutaneous leishmaniasis, is endemic in Syria, Iran, Iraq, Libya, Morocco, and elsewhere in the region. Both zoonotic (Leishmania major) and anthroponotic (Leishmania tropica) forms are endemic in MENA in rural arid regions and urban regions, respectively. Other endemic zoonotic NTDs include cystic echinococcosis, fascioliasis, and brucellosis. Dengue is endemic in Saudi Arabia, where Rift Valley fever and Alkhurma hemorrhagic fever have also emerged. Great strides have been made towards elimination of several endemic NTDs, including lymphatic filariasis in Egypt and Yemen; schistosomiasis in Iran, Morocco, and Oman; and trachoma in Morocco, Algeria, Iran, Libya, Oman, Saudi Arabia, Tunisia, and the United Arab Emirates. A particularly noteworthy achievement is the long battle waged against schistosomiasis in Egypt, where prevalence has been brought down by regular praziquantel treatment. Conflict and human and animal migrations are key social determinants in preventing the control or elimination of NTDs in the MENA, while local political will, strengthened international and intersectoral cooperative efforts for surveillance, mass drug administration, and vaccination are essential for elimination

    Impedimetric Sensor System for Edible Oil Quality Assessment

    The repeated usage of frying oil is hazardous due to the degradation caused by chemical reactions, which happen while heating. The total polar compounds and the free fatty acids are the main two chemical parameters affected by frying. These parameters increase significantly with the use of oil for frying and are reported as reasons for causing serious illnesses like heart diseases. For this purpose, sensor systems for oil quality assessment are necessary. In fact, changes of the composition due to frying leads to variation of its dielectric parameters. This can be measured using a capacitive sensor and the measurement of its impedance change. The main challenge thereby is that the impedance changes are very small and stray capacitances have a big influence on the measurements. In this context, this work proposes a sensor system with high accuracy able to detect the small changes that occur in the resistance and capacitance under influence of stry capacitances. Theoretical and simulation studies are carried out for different cap acitive sensors as well as meas urement procedures of its cornp lex imp edance. The sensor should provide a high sensitivity to relative perrnittivity and the electrical conductiv ity, and at the same time a small size and a high reproducibility. Interdigital electrodes sensor with a suitable design fulfils all these requirements. A deep consideration of stray capacitances is needed to realize an accurate sensor system. For t hese reasons, the design of the measurement circuit is crucial within this work. We propose, a measurernent circuit based on a combinat ion of the method of capacitance to voltage conversion and the phase shift measurement method. By cornbining both rnethods together it is possible to rneasure accurate ly the complex irnpedance of edible oil. Experimental results show that measurement systern is capable to detect small changes of dielectric parameters, which are correlated to the chemical parameters.Die mehrfach wiederholte Verwendung von Frittieröl ist aufgrund der QualitĂ€tsver­ schlechterung, die wĂ€hrend des Erhitzens auftreten durch chemische Reaktionen verursacht wird, gefĂ€hrlich fĂŒr die Gesundheit.     Measurement System for Lossy Capacitive Sensors: Application to Edible Oils Quality Assessment

    This paper aimed to develop a portable, low-cost, and easy-to-use measurement system for oil quality degradation assessment. The main two chemical parameters affected by frying are the total polar compounds (TPC) and free fatty acids. The system should characterize the change of chemical parameters by measuring the changes in its dielectric parameters. The dielectric parameters, relative permittivity, and conductivity are measured by measuring the capacitance and resistance of a capacitive sensor dipped in oil. The main challenges are that the corresponding changes of the capacitance and resistance are very small and the presence of stray effects. For this reason, the measurement system should be able to detect changes in capacitance and resistance with high resolution and with good immunity to stray effects. The proposed measurement system is based on the conversion of impedance to voltage and time and combining, therefore, having two measurement methods in one circuit. In this way, it is possible to measure the dielectric and resistive parameters and not only the relative permittivity as was done in previous works. The results showed a strong correlation between the chemical and electrical parameters with a coefficient of determination in the range of 0.9

    Simple Sequence Repeat Identification and Endocarp Characterization of Olive Tree Accessions in a Tunisian Germplasm Collection

    8 påginas, 2 figuras, 3 tabvas.Most traditional olive-producing countries possess a diversified genetic patrimony in Olea europaea L. Since the emergence of modern olive growing system, the identification, classification, and conservation of autochthonous olive cultivars is a priority for these countries. In this work, a total of 84 accessions belonging to the "Boughrara"-Sfax olive germplasm collection located in Tunisia have been screened using a powerful set of eight simple sequence repeat markers (SSRs). The study revealed a high genetic variability among the collection and detected a total of 64 alleles. For better management of the mentioned germplasm bank, an improved classification of the entries, including new denominations, has been proposed. In addition, several cases of mislabeling, synonymy, and homonymy have been clarified. Genetic relationships among cultivars have been analyzed showing four major clusters. Finally, a correspondence factor analysis demonstrated that cultivars tend to cluster depending on their main use as oil or table olives. No clear clustering tendencies were observed when the geographical origin of cultivars was used as the criteria for the analysis.
