    Toko Cucko Bandung adalah sebuah USAha yang bergerak dalam bidang penjualan sandal cucko yang beralamat di jalan leuwi Anyar VI , Kota Bandung – Jawa Barat. Toko Cucko ini menyediakan berupa penjualan sandal dan dapat menerima pemesanan sandal sesuai keinginan konsumen, dari model sandal ,warna sandal dan ukuran. Permasalahan saat ini pada Toko Cucko Bandung adalah dalam penjualan, mempromosikan dan pemesanan masih dengan manual ,hal ini kurang efektif karena tidak ada informasi yang detail mengenai sandal pada toko cucko untuk komsumen. Proses pemesanan yang sedang berjalan pada toko cucko bandung masih menggunakan via bbm, whatshap atau datang lansung ke tempat, hat tersebut masih kurang efesien. Pencatatan dan pengecekan data – data transaksi, data pelanggan, data barang ,masih dilakukan dengan manual dalam arsip dokumen tertulis, kehilangan data masih besar kemungkinan akan rentan terjadi.Metode penelitian yang digunakan adalah dengan wawancara, dokumentasi dan observasi. Adapun metode pendekatan sistem yang digunakan adalah Struktur dengan menggunakan Flowmap sebagai alat bantu untuk merancang sistem yang akan dibuat dengan metode pengembangan sistem ini adalah prototype. Pembangunan Sistem Informasi Penjualan sandal ini menggunakan PHP sebagai Bahasa pemrograman dan XAMPP sebagai database. Dengan adanya Sistem Informasi berbasis Website diharapkan dapat mempermudah proses pelayanan penjulan, pemensanan dan pengolahan data pada Perusahaan ini

    The Effects of Stocking Density and the Giving Tofu with Different Doses on the Population Growth Silk Worms ( Tubifex SP)

    This research was conducted on March - May 2016 in the Jln . Kamboja No. 38 , Simpang Baru , Pekanbaru Panam . This research aimed to determine the stocking density and feeding tofu dose is right for biomass growth , population and the length of silk worms ( Tubifex sp ) . The method used in this study is an experiment using a completely randomized design factorial 2 factors 3 levels of treatment with three replications . The first factor stocking density ( 35 g , 45 g and 55 g ) and doses of the feed tofu ( 35 g , 45 g , 55 g ) . The results showed that the stocking density and feeding tofu significant effect on biomass ( p < 0.05 ) . Values of biomass , length growth , and the population is highest in the treatment of stocking density 55 g / 0.098 m2 and dose feed 55 g of biomass 152g / 0.098 m2 , length growth of 1.2 cm , 19 765 ind / 0.098 m2

    Kabupaten Sukamara is a currently growing Kabupaten, especially Kecamatan Balai Riam and Kecamatan Permata Kecubung, but the location is difficult to reach and can be said isolated. One of the aspect that can improve and develop Kabupaten Sukamara is transportation, because the transportation facilities and infrastructure that exist currently is very low in Kabupaten Sukamara, in this case, especially the road which connects the villages because the lack of road development as a rural access. The methodology used for analyze the performance of the road is by calculate the number of trip generation based on the population of study area based on method developed by the Ministry of Public Works for planning transportation of urban road which belong to the small town category. The next step is to analyze the amount of trip that generates Origin Destination Matrice (OD) by using Double Constrain Gravity method (DCGR), assign OD matrice to the road network by using EMME 3.4.1 and calculate the road capacity,then calculate and analyze volume capacity ratio (VCR) based on the assignment result and the road capacity calculation. By determining the 20 year plan, the prediction of trip generation of Kabupaten Sukamara is 235.023 people/day. Based on OD matrice, can be known that the closer the distance trip, the greater the trip that occur and vice versa. From the calculation, the capacity of the collector road is 1.197 smp/hour, local road is 856 smp/hour and small road is 838 smp/hour. From the assignment result, the largest trip volume is 314 smp/hour and the smallest is 1 smp/hour, so the largest VCR value is 0,279 which means VCR &lt; 0,8 for every road that connects the villages. Therefore, the performance condition of the entire road that connects the villages can be concluded stable/safe

    Human GM3 Synthase Attenuates Taxol-Triggered Apoptosis Associated with Downregulation of Caspase-3 in Ovarian Cancer Cells

    BACKGROUND: Taxol (paclitaxel) inhibits proliferation and induces apoptosis in a variety of cancer cells, but it also upregulates cytoprotective proteins and/or pathways that compromise its therapeutic efficacy. MATERIALS AND METHOD: The roles of GM3 synthase (α2,3-sialyltransferase, ST3Gal V) in attenuating Taxol-induced apoptosis and triggering drug resistance were determined by cloning and overexpressing this enzyme in the SKOV3 human ovarian cancer cell line, treating SKOV3 and the transfectants (SKOV3/GS) with Taxol and determining apoptosis, cell survival, clonogenic ability, and caspase-3 activation. RESULTS: In this report, we demonstrated that Taxol treatment resulted in apoptosis which was associated with caspase-3 activation. Taxol treatment upregulated the expression of human GM3 synthase, an enzyme that transfers a sialic acid to lactosylceramide. Moreover, we cloned the full-length GM3 synthase gene and showed for the first time that forced expression of GM3 synthase attenuated Taxol-induced apoptosis and increased resistance to Taxol in SKOV3 cells. CONCLUSIONS: GM3 synthase overexpression inhibited Taxol-triggered caspase-3 activation, revealing that upregulation of GM3 synthase prevents apoptosis and hence reduces the efficacy of Taxol therapy

    c-FLIP, a Novel Biomarker for Cancer Prognosis, Immunosuppression, Alzheimer’s Disease, Chronic Obstructive Pulmonary Disease (COPD), and a Rationale Therapeutic Target

    Dysregulation of c-FLIP (cellular FADD-like IL-1β-converting enzyme inhibitory protein) has been shown in several diseases including cancer, Alzheimer’s disease, and chronic obstructive pulmonary disease (COPD). c-FLIP is a critical anti-cell death protein often overexpressed in tumors and hematological malignancies and its increased expression is often associated with a poor prognosis. c-FLIP frequently exists as long (c-FLIPL) and short (c-FLIPS) isoforms, regulates its anti-cell death functions through binding to FADD (FAS associated death domain protein), an adaptor protein known to activate caspases-8 and -10 and links c-FLIP to several cell death regulating complexes including the death-inducing signaling complex (DISC) formed by various death receptors. c-FLIP also plays a critical role in necroptosis and autophagy. Furthermore, c-FLIP is able to activate several pathways involved in cytoprotection, proliferation, and survival of cancer cells through various critical signaling proteins. Additionally, c-FLIP can inhibit cell death induced by several chemotherapeutics, anti-cancer small molecule inhibitors, and ionizing radiation. Moreover, c-FLIP plays major roles in aiding the survival of immunosuppressive tumor-promoting immune cells and functions in inflammation, Alzheimer’s disease (AD), and chronic obstructive pulmonary disease (COPD). Therefore, c-FLIP can serve as a versatile biomarker for cancer prognosis, a diagnostic marker for several diseases, and an effective therapeutic target.     4-(4-Chloro-2-methylphenoxy)-N-hydroxybutanamide (CMH) targets mRNA of the c-FLIP variants and induces apoptosis in MCF-7 human breast cancer cells

    Cellular FLICE (FADD-like IL-1β-converting enzyme)-inhibitory protein (c-FLIP) is a major resistance factor for the tumor necrosis factor-related apoptosis-inducing ligand TRAIL and in drug resistance in human malignancies. c-FLIP is an antagonist of caspases-8 and -10, which inhibits apoptosis and is expressed as long (c-FLIPL) and short (c-FLIPS) splice forms. c-FLIP is often overexpressed in various human cancers, including breast cancer. Several studies have shown that silencing c-FLIP by specific siRNAs sensitizes cancer cells to TRAIL and anticancer agents. However, systemic use of siRNA as a therapeutic agent is not practical at present. In order to reduce or inhibit c-FLIP expression, small molecules are needed to allow targeting c-FLIP without inhibiting caspases-8 and -10. We used a small molecule inhibitor of c-FLIP, 4-(4-chloro-2-methylphenoxy)-N-hydroxybutanamide (CMH), and show that CMH, but not its inactive analog, downregulated c-FLIPL and c-FLIPS mRNA and protein levels, caused poly(ADP-ribose) polymerase (PARP) degradation, reduced cell survival, and induced apoptosis in MCF-7 breast cancer cells. These results revealed that c-FLIP is a critical apoptosis regulator that can serve as a target for small molecule inhibitors that downregulate its expression and serve as effective targeted therapeutics against breast cancer cells

    Ipsilateral common iliac artery plus femoral artery clamping for inducing sciatic nerve ischemia/reperfusion injury in rats: a reliable and simple method

    The aim of this study was to develop a practical model of sciatic ischemia reperfusion (I/R) injury producing serious neurologic deficits and being technically feasible compared with the current time consuming or ineffective models. Thirty rats were divided into 6 groups (n = 5). Animal were anesthetized by using ketamine (50 mg/kg) and xylazine (4 mg/kg). Experimental groups included a sham-operated group and five I/R groups with different reperfusion time intervals (0 h, 3 h, 1 d, 4 d, 7 d). In I/R groups, the right common iliac artery and the right femoral artery were clamped for 3 hrs. Sham-operated animals underwent only laparotomy without induction of ischemia. Just before euthanasia, behavioral scores (based on gait, grasp, paw position, and pinch sensitivity) were obtained and then sciatic nerves were removed for light-microscopy studies (for ischemic fiber degeneration (IFD) and edema). Behavioral score deteriorated among the ischemic groups compared with the control group (p < 0.01), with maximal behavioral deficit occurring at 4 days of reperfusion. Axonal swelling and IFD were found to happen only after 4 and 7 days, respectively. Our observations led to an easy-to-use but strong enough method for inducing and studying I/R injury in peripheral nerves
