    Re-evaluation of individual diameter : height allometric models to improve biomass estimation of tropical trees

    The first author was supported by the European Union under a IEF Marie-Curie Action.Accurate estimation of tree biomass is necessary to provide realistic values of the carbon stored in the terrestrial biosphere. A recognized source of errors in tree above-ground biomass (AGB) estimation is introduced when individual tree height values (H) are not directly measured but estimated from diameter at breast height (DBH) using allometric equations. In this paper we evaluate the performance of 12 alternative DBH : H equations and compare their effects on AGB estimation for three tropical forests that occur in contrasting climatic and altitudinal zones. We found that fitting a 3-parameter Weibull function using data collected locally generated the lowest errors and bias in H estimation, and that equations fitted to these data were more accurate than equations with parameters derived from the literature. For computing AGB, the introduced error values differed notably among DBH : H allometric equations, and in most cases showed a clear bias that resulted in either over- or under-estimation of AGB. Fitting the three-parameter Weibull function minimized errors in AGB estimates in our study and we recommend its widespread adoption for carbon stock estimation. We conclude that many previous studies are likely to present biased estimates of AGB due to the method of H estimation.Publisher PDFPeer reviewe

    AN ASSESSMENT OF LAND USE SURROUNDING HYDROPOWER RESERVOIRS USING REMOTE SENSING IN SARAWAK, MALAYSIA

    The existence of a complex river system in Sarawak has contributed to the abundance of its water resources. The State is drained by 22 major river basins and has a combined installed capacity of hydropower potentials of approximately 20,000 MW of sustainable energy. However, little has been documented on land use around these hydropower dams. The current study is aimed at understanding the general land usecomposition surrounding the existing and planned hydropower reservoirs in Sarawak. The land uses surrounding these dams are expected to influence the livelihood, water discharge and water quality of the dams. A total of ten sites were selected and land use surrounding these study sites identified using Remote Sensing and Geographic Information System tools. Temporal assessment on land-use changes was only conducted for Batang Ai. The study found that a total of six sites have higher forest cover (> 50%) compared to built-up and agricultural lands and showed that 85% variation in built-up land is explained by reservoir area. Agricultural land has increased at the mean rate of 2.25% within 500 metres distance from Batang Ai reservoir shoreline. The findings of this study will hopefully contribute to the knowledge of hydropower reservoir planning and management

    Agent-based simulation in a normative environment using the EPMP domain.

    Simulation is a way of doing thought experiments besides the deduction and induction methods. Agent-based Simulation (ABS) falls under the domain of artificial intelligence when agent is used to perform certain tasks such as behaviour prediction, optimization of functions or time-constrained work-flow management. This work discussed the use of ABS on a computational normative framework based on a set of empirical characteristics that influence agents’ performance in time-constrained environment. The ABS simulates a domain called the Examination Preparation and Moderation Process (EPMP) which entails document submission processes with deadlines. The simulation is conducted in six different environments and the results of the agent performance in each environment are presented and discussed. The results indicate that the simulation conducted in the EPMP is suitable and effective for evaluating normative agent-based systems

    Con el objetivo de seguir documentando proyectos de mejora para Ciudad Obregón y el municipio de Cajeme, Sonora, México y en el mediano plazo candidatearla como Ciudad Creativa ante la iniciativa de la ONU-UNESCO y que la misma sea valorada favorablemente, principalmente en el área de Gastronomía, pero reconocida también por otros atributos como sus sitios turísticos, su servicio y atención, su cultura, arte y tradiciones; se implementó el Tercer Foro “Obregón hacia una Ciudad Creativa”. Se aplicó un método basado en grupos focales con audiencias invitadas de acuerdo con las temáticas o líneas de acción que señalan los 17 Objetivos de Desarrollo Sustentable de la ONU. Se obtuvo información de proyectos de mejora comunitaria que en alianza gubernamental, universitaria y de Ongs´s se aplican en la ciudad, así como diversas propuestas que ya se empiezan a planificar para ser aplicadas a través de las instituciones comprometidas. Se prevé seguir documentando los avances alcanzados y el número de beneficiarios de la ciudad; así como incidir en la imagen que se tiene de ella, para concretar un mayor compromiso ciudadano y finalmente el reconocimiento de la ciudad como Creativa

    Preclinical and clinical evaluation of German-sourced ONC201 for the treatment of H3K27M-mutant diffuse intrinsic pontine glioma

    Background Diffuse intrinsic pontine glioma (DIPG) is a fatal childhood brainstem tumor for which radiation is the only treatment. Case studies report a clinical response to ONC201 for patients with H3K27M-mutant gliomas. Oncoceutics (ONC201) is only available in the United States and Japan; however, in Germany, DIPG patients can be prescribed and dispensed a locally produced compound-ONC201 German-sourced ONC201 (GsONC201). Pediatric oncologists face the dilemma of supporting the administration of GsONC201 as conjecture surrounds its authenticity. Therefore, we compared GsONC201 to original ONC201 manufactured by Oncoceutics Inc. Methods Authenticity of GsONC201 was determined by high-resolution mass spectrometry and nuclear magnetic resonance spectroscopy. Biological activity was shown via assessment of on-target effects, in vitro growth, proliferation, and apoptosis analysis. Patient-derived xenograft mouse models were used to assess plasma and brain tissue pharmacokinetics, pharmacodynamics, and overall survival (OS). The clinical experience of 28 H3K27M+ mutant DIPG patients who received GsONC201 (2017-2020) was analyzed. Results GsONC201 harbored the authentic structure, however, was formulated as a free base rather than the dihydrochloride salt used in clinical trials. GsONC201 in vitro and in vivo efficacy and drug bioavailability studies showed no difference compared to Oncoceutics ONC201. Patients treated with GsONC201 (n = 28) showed a median OS of 18 months (P = .0007). GsONC201 patients who underwent reirradiation showed a median OS of 22 months compared to 12 months for GsONC201 patients who did not (P = .012). Conclusions This study confirms the biological activity of GsONC201 and documents the OS of patients who received the drug; however, GsONC201 was never used as a monotherapy

    ONC201 in combination with paxalisib for the treatment of H3K27-altered diffuse midline glioma

    Diffuse midline gliomas (DMG), including diffuse intrinsic pontine gliomas (DIPGs), are the most lethal of childhood cancers. Palliative radiotherapy is the only established treatment, with median patient survival of 9-11 months. ONC201 is a DRD2 antagonist and ClpP agonist that has shown preclinical and emerging clinical efficacy in DMG. However, further work is needed to identify the mechanisms of response of DIPGs to ONC201 treatment and to determine whether recurring genomic features influence response. Using a systems-biological approach, we showed that ONC201 elicits potent agonism of the mitochondrial protease ClpP to drive proteolysis of electron transport chain and tricarboxylic acid cycle proteins. DIPGs harboring PIK3CA-mutations showed increased sensitivity to ONC201, while those harboring TP53-mutations were more resistant. Metabolic adaptation and reduced sensitivity to ONC201 was promoted by redox-activated PI3K/Akt signaling, which could be counteracted using the brain penetrant PI3K/Akt inhibitor, paxalisib. Together, these discoveries coupled with the powerful anti-DIPG/DMG pharmacokinetic and pharmacodynamic properties of ONC201 and paxalisib have provided the rationale for the ongoing DIPG/DMG phase II combination clinical trial NCT05009992

    The unfinished agenda of communicable diseases among children and adolescents before the COVID-19 pandemic, 1990-2019: a systematic analysis of the Global Burden of Disease Study 2019

    BACKGROUND: Communicable disease control has long been a focus of global health policy. There have been substantial reductions in the burden and mortality of communicable diseases among children younger than 5 years, but we know less about this burden in older children and adolescents, and it is unclear whether current programmes and policies remain aligned with targets for intervention. This knowledge is especially important for policy and programmes in the context of the COVID-19 pandemic. We aimed to use the Global Burden of Disease (GBD) Study 2019 to systematically characterise the burden of communicable diseases across childhood and adolescence. METHODS: In this systematic analysis of the GBD study from 1990 to 2019, all communicable diseases and their manifestations as modelled within GBD 2019 were included, categorised as 16 subgroups of common diseases or presentations. Data were reported for absolute count, prevalence, and incidence across measures of cause-specific mortality (deaths and years of life lost), disability (years lived with disability [YLDs]), and disease burden (disability-adjusted life-years [DALYs]) for children and adolescents aged 0-24 years. Data were reported across the Socio-demographic Index (SDI) and across time (1990-2019), and for 204 countries and territories. For HIV, we reported the mortality-to-incidence ratio (MIR) as a measure of health system performance. FINDINGS: In 2019, there were 3·0 million deaths and 30·0 million years of healthy life lost to disability (as measured by YLDs), corresponding to 288·4 million DALYs from communicable diseases among children and adolescents globally (57·3% of total communicable disease burden across all ages). Over time, there has been a shift in communicable disease burden from young children to older children and adolescents (largely driven by the considerable reductions in children younger than 5 years and slower progress elsewhere), although children younger than 5 years still accounted for most of the communicable disease burden in 2019. Disease burden and mortality were predominantly in low-SDI settings, with high and high-middle SDI settings also having an appreciable burden of communicable disease morbidity (4·0 million YLDs in 2019 alone). Three cause groups (enteric infections, lower-respiratory-tract infections, and malaria) accounted for 59·8% of the global communicable disease burden in children and adolescents, with tuberculosis and HIV both emerging as important causes during adolescence. HIV was the only cause for which disease burden increased over time, particularly in children and adolescents older than 5 years, and especially in females. Excess MIRs for HIV were observed for males aged 15-19 years in low-SDI settings. INTERPRETATION: Our analysis supports continued policy focus on enteric infections and lower-respiratory-tract infections, with orientation to children younger than 5 years in settings of low socioeconomic development. However, efforts should also be targeted to other conditions, particularly HIV, given its increased burden in older children and adolescents. Older children and adolescents also experience a large burden of communicable disease, further highlighting the need for efforts to extend beyond the first 5 years of life. Our analysis also identified substantial morbidity caused by communicable diseases affecting child and adolescent health across the world.     The state of health in the European Union (EU-27) in 2019: a systematic analysis for the Global Burden of Disease study 2019

    Background: The European Union (EU) faces many health-related challenges. Burden of diseases information and the resulting trends over time are essential for health planning. This paper reports estimates of disease burden in the EU and individual 27 EU countries in 2019, and compares them with those in 2010.Methods: We used the Global Burden of Disease 2019 study estimates and 95% uncertainty intervals for the whole EU and each country to evaluate age-standardised death, years of life lost (YLLs), years lived with disability (YLDs) and disability-adjusted life years (DALYs) rates for Level 2 causes, as well as life expectancy and healthy life expectancy (HALE).Results:In 2019, the age-standardised death and DALY rates in the EU were 465.8 deaths and 20,251.0 DALYs per 100,000 inhabitants, respectively. Between 2010 and 2019, there were significant decreases in age-standardised death and YLL rates across EU countries. However, YLD rates remained mainly unchanged. The largest decreases in age-standardised DALY rates were observed for "HIV/AIDS and sexually transmitted diseases" and "transport injuries" (each -19%). "Diabetes and kidney diseases" showed a significant increase for age-standardised DALY rates across the EU (3.5%). In addition, "mental disorders" showed an increasing age-standardised YLL rate (14.5%).Conclusions: There was a clear trend towards improvement in the overall health status of the EU but with differences between countries. EU health policymakers need to address the burden of diseases, paying specific attention to causes such as mental disorders. There are many opportunities for mutual learning among otherwise similar countries with different patterns of disease

    Case Reports1. A Late Presentation of Loeys-Dietz Syndrome: Beware of TGFβ Receptor Mutations in Benign Joint Hypermobility

    Background: Thoracic aortic aneurysms (TAA) and dissections are not uncommon causes of sudden death in young adults. Loeys-Dietz syndrome (LDS) is a rare, recently described, autosomal dominant, connective tissue disease characterized by aggressive arterial aneurysms, resulting from mutations in the transforming growth factor beta (TGFβ) receptor genes TGFBR1 and TGFBR2. Mean age at death is 26.1 years, most often due to aortic dissection. We report an unusually late presentation of LDS, diagnosed following elective surgery in a female with a long history of joint hypermobility. Methods: A 51-year-old Caucasian lady complained of chest pain and headache following a dural leak from spinal anaesthesia for an elective ankle arthroscopy. CT scan and echocardiography demonstrated a dilated aortic root and significant aortic regurgitation. MRA demonstrated aortic tortuosity, an infrarenal aortic aneurysm and aneurysms in the left renal and right internal mammary arteries. She underwent aortic root repair and aortic valve replacement. She had a background of long-standing joint pains secondary to hypermobility, easy bruising, unusual fracture susceptibility and mild bronchiectasis. She had one healthy child age 32, after which she suffered a uterine prolapse. Examination revealed mild Marfanoid features. Uvula, skin and ophthalmological examination was normal. Results: Fibrillin-1 testing for Marfan syndrome (MFS) was negative. Detection of a c.1270G > C (p.Gly424Arg) TGFBR2 mutation confirmed the diagnosis of LDS. Losartan was started for vascular protection. Conclusions: LDS is a severe inherited vasculopathy that usually presents in childhood. It is characterized by aortic root dilatation and ascending aneurysms. There is a higher risk of aortic dissection compared with MFS. Clinical features overlap with MFS and Ehlers Danlos syndrome Type IV, but differentiating dysmorphogenic features include ocular hypertelorism, bifid uvula and cleft palate. Echocardiography and MRA or CT scanning from head to pelvis is recommended to establish the extent of vascular involvement. Management involves early surgical intervention, including early valve-sparing aortic root replacement, genetic counselling and close monitoring in pregnancy. Despite being caused by loss of function mutations in either TGFβ receptor, paradoxical activation of TGFβ signalling is seen, suggesting that TGFβ antagonism may confer disease modifying effects similar to those observed in MFS. TGFβ antagonism can be achieved with angiotensin antagonists, such as Losartan, which is able to delay aortic aneurysm development in preclinical models and in patients with MFS. Our case emphasizes the importance of timely recognition of vasculopathy syndromes in patients with hypermobility and the need for early surgical intervention.
