Effects of Creatine Supplementation on Glucose Uptake in Rats Submitted to Exercise Training
Recently, studies have suggested that creatine supplementation can modulate glucose homeostasis by increasing glucose uptake in peripheral tissues. The aim of this study was to investigate the effects of creatine supplementation on glucose tolerance, muscle and hepatic glycogen content in rats submitted or not to physical activity for four and eight weeks. Wistar rats were divided in two groups: four and eight weeks of intervention. Afterwards, each group was subdivided in four subgroups, according to supplement intake and exercise: Sedentary Control; Trained Control; Supplemented Sedentary; and Supplemented Trained. The animals had free access to water and chow and the supplemented groups had two % of their diet as creatine monohydrated. The exercise groups swam for 40 minutes a day, four days a week, with two to five % of their body weight attached to their chests. After four and eight weeks, oral glucose tolerance tests were performed and both hepatic and muscle glycogen were determined. No significant differences were observed between groups on glucose tolerance and glycogen content in muscle and hepatic tissue. This study shows that creatine supplementation does not influence neither glucose tolerance nor glycogen concentrations in rats submitted or not to physical activity for four and eight weeks.14543143
Additive effects of beta-alanine and sodium bicarbonate on high-intensity upper-body intermittent performance
We examined the isolated and combined effects of beta-alanine (BA) and sodium bicarbonate (SB) on high-intensity intermittent upper-body performance in judo and jiu-jitsu competitors. 37 athletes were assigned to one of four groups: (1) placebo (PL)+PL; (2) BA+PL; (3) PL+SB or (4) BA+SB. BA or dextrose (placebo) = (6.4 g day-1) was ingested for 4 weeks and 500 mg kg-1 BM of SB or calcium carbonate (placebo) was ingested for 7 days during the 4th week. Before and after 4 weeks of supplementation, the athletes completed four 30-s upper-body Wingate tests, separated by 3 min. Blood lactate was determined at rest, immediately after and 5 min after the 4th exercise bout, with perceived exertion reported immediately after the 4th bout. BA and SB alone increased the total work done in +7 and 8 %, respectively. The co-ingestion resulted in an additive effect (+14 %, p < 0.05 vs. BA and SB alone). BA alone significantly improved mean power in the 2nd and 3rd bouts and tended to improve the 4th bout. SB alone significantly improved mean power in the 4th bout and tended to improve in the 2nd and 3rd bouts. BA+SB enhanced mean power in all four bouts. PL+PL did not elicit any alteration on mean and peak power. Post-exercise blood lactate increased with all treatments except with PL+PL. Only BA+ SB resulted in lower ratings of perceived exertion (p = 0.05). Chronic BA and SB supplementation alone equally enhanced high-intensity intermittent upper-body performance in well-trained athletes. Combined BA and SB promoted a clear additive ergogenic effect
Biochemical Effects of Carbohydrate Supplementation in a Simulated Competition of Short Terrestrial Duathlon
The purpose of the present study was to investigate the biochemical effects of carbohydrate supplementation in a simulated competition of short terrestrial duathlon. Ten duathletes participated in a simulated competition of short terrestrial duathlon 30 minutes after the ingestion of a 6% (30 g/500 ml) maltodextrin solution (MALT) or a placebo (PLA). This solution was also ingested every 15 minutes during the competition (12 g/200 ml); and immediately after the competition (18 g/300 ml). Samples of blood were collected at 3 time points: 1) at rest 1 hour before the beginning of the competition; 2) during the competition (approximately 1 hour and 45 minutes after the 1st collection); 3) immediately after the competition. Blood was analyzed for blood glucose, lactate, insulin and cortisol. Significant differences were observed in relation to blood glucose levels between MALT and PLA in the post-competition phase. There was also a significant difference in the lactate levels observed between MALT and PLA during the competition phase. Similarly, a significant difference in the cortisol concentrations during and after the competition phases (MALT and PLA) were observed. We conclude that maltodextrin supplementation appears to be beneficial during short terrestrial duathlon competition as evidenced by biochemical markers
Are Genome-Wide Association Study Identified Single-Nucleotide Polymorphisms Associated With Sprint Athletic Status? A Replication Study With 3 Different Cohorts
Purpose: To replicate previous genome-wide association study identified sprint-related polymorphisms in 3 different cohorts of top-level sprinters and to further validate the obtained results in functional studies. Methods: A total of 240 Japanese, 290 Russians, and 593 Brazilians were evaluated in a case-control approach. Of these, 267 were top-level sprint/power athletes. In addition, the relationship between selected polymorphisms and muscle fiber composition was evaluated in 203 Japanese and 287 Finnish individuals. Results: The G allele of the rs3213537 polymorphism was overrepresented in Japanese (odds ratio [OR]: 2.07, P = .024) and Russian (OR: 1.93, P = .027) sprinters compared with endurance athletes and was associated with an increased proportion of fast-twitch muscle fibers in Japanese (P = .02) and Finnish (P = .041) individuals. A meta-analysis of the data from 4 athlete cohorts confirmed that the presence of the G/G genotype rather than the G/A+A/A genotypes increased the OR of being a sprinter compared with controls (OR: 1.49, P = .01), endurance athletes (OR: 1.79, P = .001), or controls + endurance athletes (OR: 1.58, P = .002). Furthermore, male sprinters with the G/G genotype were found to have significantly faster personal times in the 100-m dash than those with G/A+A/A genotypes (10.50 [0.26] vs 10.76 [0.31], P = .014). Conclusion: The rs3213537 polymorphism found in the CPNE5 gene was identified as a highly replicable variant associated with sprinting ability and the increased proportion of fast-twitch muscle fibers, in which the homozygous genotype for the major allele (ie, the G/G genotype) is preferable for performance
Distinct effects of leucine or a mixture of the branched-chain amino acids (leucine, isoleucine, and valine) supplementation on resistance to fatigue, and muscle and liver-glycogen degradation, in trained rats
Fundação de Amparo à Pesquisa do Estado de São Paulo (FAPESP)Objective: The aim of this study was to evaluate the effects of the mixture of branched-chain amino acids (BCAAs) supplementation compared with leucine administered orally on muscle biochemical parameters of trained rats submitted to an exercise-induced protocol of glycogen depletion. Methods: After 6 wk of swimming exercise, 8 wk-old (250 g, adult) male Wistar rats were randomly divided into three experimental groups (n = 8 per group): the mixture of BCAAs (BCAAs), leucine (LEU), and placebo (PLA). All groups were submitted to swimming exercise for 6 wk and supplemented with either the mixture of BCAAs, leucine, or placebo during the last week of training. At week 7 of the protocol, the rats were submitted to an intermittent, progressive swimming test until exhaustion and sacrificed. Muscle gastrocnemius and liver were depicted to determine total glycogen, tricarboxylic acid cycle (TCA) intermediates, and enzymatic activities. Statistical evaluation was performed by one-way analysis of variance with Tukey post hoc test. Results: Both muscle and liver glycogen degradation ratio were significantly higher in the mixture of BCAAs group compared to the PLA group (P 0.05) Muscle citrate content was significantly higher in the mixture of BCAAs group compared with the PLA group (P < 0.001). Muscle malate content was significantly elevated in the mixture of BCAAs group compared with both the PLA (P < 0.001) and LEU groups (P < 0.001). BCAT activity was significantly reduced in the mixture of BCAAs supplementation group compared with the LEU group (P < 0.001). Conclusion: Leucine supplementation improved performance compared with the mixture of BCAAs supplementation, sparing muscle glycogen stores despite the augmentation of some TCA intermediate concentrations on the left side of the TCA cycle.
