    A Framework for Verifying Data-Centric Protocols

    International audienceData centric languages, such as recursive rule based languages, have been proposed to program distributed applications over networks. They simplify greatly the code, while still admitting efficient distributed execution. We show that they also provide a promising approach to the verification of distributed protocols, thanks to their data centric orientation, which allows us to explicitly handle global structures such as the topology of the network. We consider a framework using an original formalization in the Coq proof assistant of a distributed computation model based on message passing with either synchronous or asynchronous behavior. The declarative rules of the Netlog language for specifying distributed protocols and the virtual machines for evaluating these rules are encoded in Coq as well. We consider as a case study tree protocols, and show how this framework enables us to formally verify them in both the asynchronous and synchronous setting

    Vascular Remodeling in Health and Disease

    The term vascular remodeling is commonly used to define the structural changes in blood vessel geometry that occur in response to long-term physiologic alterations in blood flow or in response to vessel wall injury brought about by trauma or underlying cardiovascular diseases.1, 2, 3, 4 The process of remodeling, which begins as an adaptive response to long-term hemodynamic alterations such as elevated shear stress or increased intravascular pressure, may eventually become maladaptive, leading to impaired vascular function. The vascular endothelium, owing to its location lining the lumen of blood vessels, plays a pivotal role in regulation of all aspects of vascular function and homeostasis.5 Thus, not surprisingly, endothelial dysfunction has been recognized as the harbinger of all major cardiovascular diseases such as hypertension, atherosclerosis, and diabetes.6, 7, 8 The endothelium elaborates a variety of substances that influence vascular tone and protect the vessel wall against inflammatory cell adhesion, thrombus formation, and vascular cell proliferation.8, 9, 10 Among the primary biologic mediators emanating from the endothelium is nitric oxide (NO) and the arachidonic acid metabolite prostacyclin [prostaglandin I2 (PGI2)], which exert powerful vasodilatory, antiadhesive, and antiproliferative effects in the vessel wall

    Para-infectious brain injury in COVID-19 persists at follow-up despite attenuated cytokine and autoantibody responses

    To understand neurological complications of COVID-19 better both acutely and for recovery, we measured markers of brain injury, inflammatory mediators, and autoantibodies in 203 hospitalised participants; 111 with acute sera (1–11 days post-admission) and 92 convalescent sera (56 with COVID-19-associated neurological diagnoses). Here we show that compared to 60 uninfected controls, tTau, GFAP, NfL, and UCH-L1 are increased with COVID-19 infection at acute timepoints and NfL and GFAP are significantly higher in participants with neurological complications. Inflammatory mediators (IL-6, IL-12p40, HGF, M-CSF, CCL2, and IL-1RA) are associated with both altered consciousness and markers of brain injury. Autoantibodies are more common in COVID-19 than controls and some (including against MYL7, UCH-L1, and GRIN3B) are more frequent with altered consciousness. Additionally, convalescent participants with neurological complications show elevated GFAP and NfL, unrelated to attenuated systemic inflammatory mediators and to autoantibody responses. Overall, neurological complications of COVID-19 are associated with evidence of neuroglial injury in both acute and late disease and these correlate with dysregulated innate and adaptive immune responses acutely

    Topo IIIα and BLM Act within the Fanconi Anemia Pathway in Response to DNA-Crosslinking Agents

    The Bloom protein (BLM) and Topoisomerase IIIα are found in association with proteins of the Fanconi anemia (FA) pathway, a disorder manifesting increased cellular sensitivity to DNA crosslinking agents. In order to determine if the association reflects a functional interaction for the maintenance of genome stability, we have analyzed the effects of siRNA-mediated depletion of the proteins in human cells. Depletion of Topoisomerase IIIα or BLM leads to increased radial formation, as is seen in FA. BLM and Topoisomerase IIIα are epistatic to the FA pathway for suppression of radial formation in response to DNA interstrand crosslinks since depletion of either of them in FA cells does not increase radial formation. Depletion of Topoisomerase IIIα or BLM also causes an increase in sister chromatid exchanges, as is seen in Bloom syndrome cells. Human Fanconi anemia cells, however, do not demonstrate increased sister chromatid exchanges, separating this response from radial formation. Primary cell lines from mice defective in both Blm and Fancd2 have the same interstrand crosslink-induced genome instability as cells from mice deficient in the Fancd2 protein alone. These observations demonstrate that the association of BLM and Topoisomerase IIIα with Fanconi proteins is a functional one, delineating a BLM-Topoisomerase IIIα-Fanconi pathway that is critical for suppression of chromosome radial formation

    Grass reseeding technology as a means of rehabilitating degraded lands and improving livelihoods of agro-pastoral communities in semi-arid Kenya

    Land degradation is a major problem in the semi-arid environments of Sub-Saharan Africa. Fighting land degradation is essential to ensure the sustainable and long-term productivity of the inhabited semi-arid lands. Cenchrus ciliaris (African foxtail grass), Enteropogon macrostachyus (bush rye) and Eragrostis superba (Maasai love grass) are important perennial grasses in the Eastern African semi-arid lands. A study was conducted to establish the contribution of these indigenous grasses in improving soil hydrological properties, rehabilitation, food security and the livelihoods of agro-pastoral communities in semi-arid districts of Kenya. Soil hydrological properties were tested using a Kamphorst simulator at different stubble heights to represent three different grazing intensities (low, medium, high). Ground cover was estimated using the step-point method. A survey was also conducted across 50 agro-pastoral households to establish the multidimensional benefits of the grasses. Sediment production (as a function of run-off and infiltration capacity) was significantly different (P<0.05) at different stubble heights. Ground cover estimates of the grasses were also significantly different (P<0.05). Cenchrus ciliaris had the greatest influence in improving soil hydrological properties. Enteropogon macrostachyus and E. superba were ranked second and third, respectively. Enteropogon macrostachyus had the greatest ground cover. Cenchrus ciliaris and E. superba were ranked second and third, respectively. These results were attributed to the growth and morphological characteristics of the grasses. Generally, an increase in stubble height increased infiltration capacity and reduced run-off and sediment production. Results from the household survey showed that the grasses provide a source of income through the sale of hay, grass seeds and milk, which also achieve a balanced diet.
