    Ergodicity and spectral cascades in point vortex flows on the sphere

The computations were supported by the CUNY HPCC under NSF Grants No. CNS-0855217 and No. CNS-0958379.We present results for the equilibrium statistics and dynamic evolution of moderately large [n = O (102 - 103)] numbers of interacting point vortices on the sphere under the constraint of zero mean angular momentum. For systems with equal numbers of positive and negative identical circulations, the density of rescaled energies, p(E), converges rapidly with n to a function with a single maximum with maximum entropy. Ensemble-averaged wave-number spectra of the nonsingular velocity field induced by the vortices exhibit the expected k-1 behavior at small scales for all energies. Spectra at the largest scales vary continuously with the inverse temperature of the system. For positive temperatures, spectra peak at finite intermediate wave numbers; for negative temperatures, spectra decrease everywhere. Comparisons of time and ensemble averages, over a large range of energies, strongly support ergodicity in the dynamics even for highly atypical initial vortex configurations. Crucially, rapid relaxation of spectra toward the microcanonical average implies that the direction of any spectral cascade process depends only on the relative difference between the initial spectrum and the ensemble mean spectrum at that energy, not on the energy, or temperature, of the system.Publisher PDFPeer reviewe

    PRODUCTION OF ETHANOL BY FED-BATCH FERMENTATION

    The production of ethanol, from glucose in batch and fed batch culture, was investigated. In the fed batch culture, the glucose feeding was added into the culture at 16th hour of fermentation. The effects of different glucose concentration feeding rates on ethanol fermentation were investigated for fed batch culture. The 2gL-1hr-1 glucose concentration feeding rate was found to give higher ethanol yield (2.47 g ethanol g glucose-1), with respect to substrate consumed as compared to 8 gL-1hr-1 (0.23 g ethanol g glucose-1) and 4 gL-1hr-1 (0.20 g ethanol g glucose-1). The ethanol yield with respect to substrate consumed obtained in batch culture was 0.81 g ethanol g glucose-1. The fed batch culture at 2 gL-1hr-1 glucose concentration feeding rate was proven to be a better fermentation system than the batch culture. The specific growth rate, specific glucose consumption rate and specific ethanol production rate for the fed batch fermentation, at 2 gL-1hr-1 glucose concentration feeding rate, were 0.065 hr-1, 1.20 hr-1 and 0.0009 hr-1, respectively

    Consistency in Multi-Viewpoint Architectural Design of Enterprise Information Systems

    Different stakeholders in the design of an enterprise information system have their own view on that design. To help produce a coherent design this paper presents a framework that aids in specifying relations between such views. To help produce a consistent design the framework also aids in specifying consistency rules that apply to the view relations and in checking the consistency according to those rules. The framework focuses on the higher levels of abstraction in a design, we refer to design at those levels of abstraction as architectural design. The highest level of abstraction that we consider is that of business process design and the lowest level is that of software component design. The contribution of our framework is that it provides a collection of basic concepts that is common to viewpoints in the area of enterprise information systems. These basic concepts aid in relating viewpoints by providing: (i) a common terminology that helps stakeholders to understand each others concepts; and (ii) a basis for defining re-usable consistency rules. In particular we define re-usable rules to check consistency between behavioural views that overlap or are a refinement of each other. We also present an architecture for a tool suite that supports our framework. We show that our framework can be applied, by performing a case study in which we specify the relations and consistency rules between the RM-ODP enterprise, computational and information viewpoints

    Bioaugmentation of UASB reactors with immobilized Sulfurospirillum barnesii for simultaneous selenate and nitrate removal

    Whole-cell immobilization of selenate-respiring Sulfurospirillum barnesii in polyacrylamide gels was investigated to allow the treatment of selenate contaminated (790¿µg Se¿×¿L-1) synthetic wastewater with a high molar excess of nitrate (1,500 times) and sulfate (200 times). Gel-immobilized S. barnesii cells were used to inoculate a mesophilic (30°C) bioreactor fed with lactate as electron donor at an organic loading rate of 5 g chemical oxygen demand (COD)¿×¿L-1 day-1. Selenate was reduced efficiently (>97%) in the nitrate and sulfate fed bioreactor, and a minimal effluent concentration of 39¿µg Se¿×¿L-1 was obtained. Scanning electron microscopy with energy dispersive X-ray (SEM–EDX) analysis revealed spherical bioprecipitates of =2¿µm diameter mostly on the gel surface, consisting of selenium with a minor contribution of sulfur. To validate the bioaugmentation success under microbial competition, gel cubes with immobilized S. barnesii cells were added to an Upflow Anaerobic Sludge Bed (UASB) reactor, resulting in earlier selenate (24 hydraulic retention times (HRTs)) and sulfate (44 HRTs) removal and higher nitrate/nitrite removal efficiencies compared to a non-bioaugmented control reactor. S. barnesii was efficiently immobilized inside the UASB bioreactors as the selenate-reducing activity was maintained during long-term operation (58 days), and molecular analysis showed that S. barnesii was present in both the sludge bed and the effluent. This demonstrates that gel immobilization of specialized bacterial strains can supersede wash-out and out-competition of newly introduced strains in continuous bioaugmented systems. Eventually, proliferation of a selenium-respiring specialist occurred in the non-bioaugmented control reactor, resulting in simultaneous nitrate and selenate removal during a later phase of operatio

    Traditionally the position of reliability analysis in the design and production process of electronic circuits is a position of reliability verification. A completed design is checked on reliability aspects and either rejected or accepted for production. This paper describes a method to model physical failure mechanisms within components in such a way that they can be used for reliability optimization, not after, but during the early phase of the design process. Furthermore a prototype of a CAD software tool is described, which can highlight components likely to fail and automatically adjust circuit parameters to improve product reliability

    Glyoxalase II (GloII) is a ubiquitous thioester hydrolase catalyzing the last step of the glutathione-dependent conversion of 2-oxoaldehydes to 2-hydroxycarboxylic acids. Here, we present a detailed structure-function analysis of cGloII from the malaria parasite Plasmodium falciparum. The activity of the enzyme was salt-sensitive and pH-log k(cat) and pH-log k(cat)/K-m profiles revealed acid-base catalysis. An acidic pK(a)(app) value of approximately 6 probably reflects hydroxide formation at the metal center. The glutathione-binding site was analyzed by site-directed mutagenesis. Substitution of residue Arg(154) caused a 2.5-fold increase of K-m(app), whereas replacements of Arg(257) or Lys(260) were far more detrimental. Although the glutathione-binding site and the catalytic center are separated, six of six single mutations at the substrate-binding site decreased the k(cat)(app) value. Furthermore, product inhibition studies support a Theorell-Chance Bi Bi mechanism with glutathione as the second product. We conclude that the substrate is predominantly bound via ionic interactions with the conserved residues Arg(257) and Lys(260), and that correct substrate binding is a pH-and salt-dependent rate-limiting step for catalysis. The presented mechanistic model is presumably also valid for GloII from many other organisms. Our study could be valuable for drug development strategies and enhances the understanding of the chemistry of binuclear metallohydrolases
