    The transformation of the cultural memory through a prism of the Cyberspace

    The paper considers the analysis of the cultural memory, which at the beginning of the XXI century underwent essential changes. The beginning of the memory research as a special phenomenon belongs to the Russian scientist, Nikolaj Miljukov [1] who has proved the emergence of social memory with the emergence of the intellectuals. In the article β€œIntellectuals and Historical Tradition”, Miljukov emphasizes the importance of social memory for the forward positive development of society. Since Halbwachs social memory gets the status of the independent research project. At the end of the XIX century the main theoretical concepts of social memory are formed: cultural and historical, semiotics, informational, psychological. The beginning of the third millennium is marked by the basic change of research trajectories of social memory. Specific research projects are put in the forefront. Such articles can be used as an example. The main research objective consists in the identification of the sociocultural bases of the transformation of social memory research. In modern investigations of the cultural memory, this phenomenon finds the new paradigmatic characteristics, which are fully submitted in the modern Internet space

    Modern requirements to preparation of professional and pedagogical personnel

    The relevance of the researched problem is caused by the need of providing the educational organizations of the system of professional education with specialists in projecting and implementation of educational programs of training of workers and specialists of a middle link, and formation of a personnel resource for innovative productions. The purpose of the presented article consists in consideration of the discrepancies arising while introduction of the professional standard "Teacher of Vocational Training, Vocational Education and Further Vocational Education" and changes revelation of a pedagogical personnel which are necessary for improvement of preparation quality. The leading approach to the research of this problem is the system-based approach which allows us to reveal the discrepancies arising by the comparison of the professional standard and educational programs that are developed taking into account requirements of the educational standard for the direction of preparation Vocational Training (branch-wise). According to the analysis results, the option of adjustment of educational programs for the direction of preparation Vocational Training (branch-wise) is offered; it promotes improvement of preparation quality of pedagogical personnel for the system of professional education and further vocational education, and also the corporate educational organizations and innovative productions.     Probing High Frequency Noise with Macroscopic Resonant Tunneling

    We have developed a method for extracting the high-frequency noise spectral density of an rf-SQUID flux qubit from macroscopic resonant tunneling (MRT) rate measurements. The extracted noise spectral density is consistent with that of an ohmic environment up to frequencies ~ 4 GHz. We have also derived an expression for the MRT lineshape expected for a noise spectral density consisting of such a broadband ohmic component and an additional strongly peaked low-frequency component. This hybrid model provides an excellent fit to experimental data across a range of tunneling amplitudes and temperatures

    Dynamics of Josephson junctions and single-flux-quantum networks with superconductor-insulator-normal metal junction shunts

    Within the framework of the microscopic model of tunneling, we modelled the behavior of the Josephson junction shunted by the Superconductor-Insulator-Normal metal (SIN) tunnel junction. We found that the electromagnetic impedance of the SIN junction yields both the frequency-dependent damping and dynamic reactance which leads to an increase in the effective capacitance of the circuit. We calculated the dc I-V curves and transient characteristics of these circuits and explained their quantitative differences to the curves obtained within the resistively shunted junction model. The correct operation of the basic single-flux-quanta circuits with such SIN-shunted junctions, i.e. the Josephson transmission line and the toggle flip-flop, have also been modelled.Comment: 8 pages incl. 7 figure

    Single flux quantum circuits with damping based on dissipative transmission lines

    We propose and demonstrate the functioning of a special Rapid Single Flux Quantum (RSFQ) circuit with frequency-dependent damping. This damping is achieved by shunting individual Josephson junctions by pieces of open-ended RC transmission lines. Our circuit includes a toggle flip-flop cell, Josephson transmission lines transferring single flux quantum pulses to and from this cell, as well as DC/SFQ and SFQ/DC converters. Due to the desired frequency-dispersion in the RC line shunts which ensures sufficiently low noise at low frequencies, such circuits are well-suited for integrating with the flux/phase Josephson qubit and enable its efficient control.Comment: 6 pages incl. 6 figure

    Functional Analysis of DNMT3A DNA Methyltransferase Mutations Reported in Patients with Acute Myeloid Leukemia

    In mammals, DNA methylation is necessary for the maintenance of genomic stability, gene expression regulation, and other processes. During malignant diseases progression, changes in both DNA methylation patterns and DNA methyltransferase (MTase) genes are observed. Human de novo MTase DNMT3A is most frequently mutated in acute myeloid leukemia (AML) with a striking prevalence of R882H mutation, which has been extensively studied. Here, we investigate the functional role of the missense mutations (S714C, R635W, R736H, R771L, P777R, and F752V) found in the catalytic domain of DNMT3A in AML patients. These were accordingly mutated in the murine Dnmt3a catalytic domain (S124C, R45W, R146H, R181L, P187R, and F162V) and in addition, one-site CpG-containing DNA substrates were used as a model system. The 3-15-fold decrease (S124C and P187R) or complete loss (F162V, R45W, and R146H) of Dnmt3a-CD methylation activity was observed. Remarkably, Pro 187 and Arg 146 are not located at or near the Dnmt3a functional motives. Regulatory protein Dnmt3L did not enhance the methylation activity of R45W, R146H, P187R, and F162V mutants. The key steps of the Dnmt3a-mediated methylation mechanism, including DNA binding and transient covalent intermediate formation, were examined. There was a complete loss of DNA-binding affinity for R45W located in the AdoMet binding region and for R146H. Dnmt3a mutants studied in vitro suggest functional impairment of DNMT3A during pathogenesis

It reflects the investment activity in the state, which reflects the direction, goals, forms of state authority in the field of science and technology. The financial growth of the state is based on a combination of extensive and intensive factors. Under extensive factors mean weight gain used in the production of resources under intensive factors - an increase in resources and performance properties of their use. In modern times, the ability to finance growth through involvement in the turnover of new resources are extremely limited. As a consequence become crucial intense moments. A decisive impact on the macro-economic characteristics have innovation. They affect the social strength, on the environment, on the intensity of international technical cooperation, the degree of public safety and the competitiveness of the national economy in the world economy. Currently, the innovation process determines the chances of becoming a state. Innovative activity is characterized by complexity and high risk. To initiate innovation is insufficient availability of internal incentives and capabilities. Here, a decisive driving force can be external stimuli and in particular state aid.     Objective: to perform cost-effectiveness analysis of the treatment for adult patients with psoriatic arthritis (PsA) with a Russian interleukin- 17А inhibitor netakimab in comparison with other biologic disease-modifying antirheumatic drugs (bDMARDs) and targeted synthetic disease-modifying antirheumatic drugs (tsDMARDs) and to evaluate the influence of the inclusion of netakimab in the therapy for PsA on the budget of the Russian healthcare system.Material and methods. The evaluation of cost-effectiveness was performed from the position of the Russian healthcare system for patients with moderate and severe PsA. The evaluation was performed based on the results of the network meta-analysis of the randomized clinical studies. The criterion of clinical effectiveness included the changes in the condition of the joints by the criteria of the American College of Rheumatology (ACR20, ACR50, and ACR70) and changes in skin symptoms by the index of the prevalence and severity of psoriasis (PASI 75 and PASI 90) with a recalculation into quality-adjusted life-year (QALY). The time horizon of the cost-effectiveness analysis was 2 years. The calculation was based on the registered prices and VAT. If there was an original drug and a biosimilar registered on the pharmaceutical market, the calculation was based on the median of the registered prices. The budget impact analysis of the influence of netakimab inclusion in the therapy for patients with PsA was performed considering the structure of the prescription of bDMARDs and tsDMARDs that was determined in the pharmaco-epidemiological study conducted in the Russian Federation in 2020. The analysis was performed for patients that received medication by the scheme of reimbursement. The time horizon of the study was 3 years old.Results. In the base-case analysis, the cost-effectiveness ratio for netakimab was 1.210 mln rub/QALY (by 66.2–88.5% lower than in cases when comparison drugs were used). The budget impact analysis showed that the inclusion of netakimab in the therapy for PsA could reduce the costs by 376.60 mln rub (21.1%). Considering budget saving, the number of additional patients that can be treated will increase by 26.7% within 3 years.Conclusion. Netakimab is characterized by higher cost-effectiveness in comparison with other bDMARDs (adalimumab, golimumab, guselkumab, ixekizumab, infliximab, secukinumab, ustekinumab, certolizumab pegol, etanercept) and tsDMARDs (apremilast, tofacitinib) prescribed in the Russian Federation for patients with PsA.
