    Age-related increase of kynurenic acid in human cerebrospinal fluid-IgG and beta(2)-microglobulin changes

    Kynurenic acid (KYNA) is an endogenous metabolite in the kynurenine pathway of tryptophan degradation and is an antagonist at the glycine site of the N-methyl-D-aspartate as well as at the alpha 7 nicotinic cholinergic receptors. In the brain tissue KYNA is synthesised from L-kynurenine by kynurenine aminotransferases (KAT) I and II. A host of immune mediators influence tryptophan degradation. In the present study, the levels of KYNA in cerebrospinal fluid (CSF) and serum in a group of human subjects aged between 25 and 74 years were determined by using a high performance liquid chromatography method. In CSF and serum KAT I and II activities were investigated by radioenzymatic assay, and the levels of β2-microglobulin, a marker for cellular immune activation, were determined by ELISA. The correlations between neurochemical and biological parameters were evaluated. Two subject groups with significantly different ages, i.e. 50 years, p < 0.001, showed statistically significantly different CSF KYNA levels, i.e. 2.84 ± 0.16 fmol/μl vs. 4.09 ± 0.14 fmol/μl, p < 0.001, respectively; but this difference was not seen in serum samples. Interestingly, KYNA is synthesised in CSF principally by KAT I and not KAT II, however no relationship was found between enzyme activity and ageing. A positive relationship between CSF KYNA levels and age of subjects indicates a 95% probability of elevated CSF KYNA with ageing (R = 0.6639, p = 0.0001). KYNA levels significantly correlated with IgG and β2-microglobulin levels (R = 0.5244, p = 0.0049; R = 0.4253, p = 0.043, respectively). No correlation was found between other biological parameters in CSF or serum. In summary, a positive relationship between the CSF KYNA level and ageing was found, and the data would suggest age-dependent increase of kynurenine metabolism in the CNS. An enhancement of CSF IgG and β2-microglobulin levels would suggest an activation of the immune system during ageing. Increased KYNA metabolism may be involved in the hypofunction of the glutamatergic and/or nicotinic cholinergic neurotransmission in the ageing CNS

    This document presents the proceedings of a one-day Workshop on Investing in Biodiversity Conservation held at the Inter-American Development Bank in Washington, D.C., on October 28, 1996. The first part of the workshop was dedicated to the presentation of key topics on biodiversity financing by five leaders in the field. The second part of the workshop was dedicated to a discussion and exchange of ideas on the role of the IDB in investing in biodiversity conservation. Three main recommendations emerged: 1) The Bank should prepare a report on on its experience in biodiversity projects and development programs with biodiversity components; 2) A task force should be formed to work on a bio-diversity policy or strategy; 3) IDB staff should be trained to understand the biodiversity concept and its implications in project preparation and implementation.Environmental Policy, Biodiversity, Natural Resources Management

    Becoming Director: An Internal Candidate\u27s View

    Articles reviewing the challenges of assuming a library directorship typically begin in the middle of the story. The new hire arrives from another campus to face a range of challenges. The accounts from there go on to extract a general map of the initial experiences along with helpful tools to navigate this intimidating terrain. That view, we suggest, obscures natural fault lines within the community of new directors. These divisions can fundamentally influence the initial experiences upon which the authors are offering their advice. On such variable concerns the route by which the successful applicant has been named the new director. Some are in-house candidates with specific experience in the library they will now lead, while others are outside candidates who face the added challenge to understand the ways that their understanding of other institutions will translate to the new context. The latter is so much the more common that it is often taken for granted. On the contrary, we suggest that this distinction impacts not only the early actions of the new director, but the hiring process as well. To bring greater balance to the literature of director hiring, we offer the personal experience of the lead author as a successful internal candidate, and put this personal account into the broader context of the hiring process for library leadership positions

    Development of mental rotation in 3- to 5-year-old children

    We assessed 3- to 5-year-olds’ mental rotation abilities using a new puzzle paradigm. It allows for assessment of mental rotation abilities in children younger than 5 years, using a task comparable to ones used with older children and adults. Children saw pairs of asymmetrical ghost figures, either as three-dimensional cut-outs or two- dimensional paper versions, in seven orientations. One of the ghosts fit into a hole if rotated right-side up – the other ghost was its mirror image and would not fit. Children were asked to turn the ghosts in their heads and choose the one that would fit into the hole. The number of children who chose the correct ghost above chance in the three- dimensional version of the task increased dramatically from 10% of 3-year-olds to 95% of 5-year-olds; average accuracy also increased significantly, from 54% to 83%. The two-dimensional paper version yielded similar results. These results indicate considerable development in mental rotation between 3 and 5 years

Epigenetic aberrations are common in adult AML, and many novel epigenetic compounds are in clinical development that may improve patient outcomes. Mutations in epigenetic regulators occur less frequently in AML in children compared to adults. However, investigating the potential benefits of epigenetic therapy in paediatric AML is an important issue that will be discussed in this review
