    Texture analysis of magnetic resonance images of rat muscles during atrophy and regeneration

    OBJECTIVES: The goals of the current study were (i) to introduce texture analysis on magnetic resonance imaging (MRI-TA) as a noninvasive method of muscle investigation that can discriminate three muscle conditions in rats; these are normal, atrophy and regeneration; and (ii) to show consistency between MRI-TA results and histological results of muscle type 2 fibers\u27 cross-sectional area. METHOD: Twenty-three adult female Wistar rats were randomized into (i) control (C), (ii) immobilized (I) and (iii) recovering (R) groups. For the last two groups, the right hind limb calf muscles were immobilized against the abdomen for 14 days; then, the hind limb was remobilized only for the R group for 40 days. At the end of each experimental period, MRI was performed using 7-T magnet Bruker Avance DRX 300 (Bruker, Wissembourg); T1-weighted MRI acquisition parameters were applied to show predominantly muscle fibers. Rats were sacrificed, and the gastrocnemius muscle (GM) was excised immediately after imaging. (A) Histology: GM type 2 fibers (fast twitch) were selectively stained using the adenosine triphosphatase (ATPase) technique. The mean cross-sectional areas were compared between the three groups. (B) Image analysis: regions of interest (ROIs) were selected on GM MR images where statistical methods of texture analysis were applied followed by linear discriminant analysis (LDA) and classification. RESULTS: Histological analysis showed that the fibers\u27 mean cross-sectional areas on GM transversal sections represented a significant statistical difference between I and C rats (ANOVA, P<.001) as well as between R and I rats (ANOVA, P<.01), but not between C and R rats. Similarly, MRI-TA on GM transversal images detected different texture for each group with the highest discrimination values (Fisher F coefficient) between the C and I groups, as well as between I and R groups. The lowest discrimination values were found between C and R groups. LDA showed three texture classes schematically separated. CONCLUSION: Quantitative results of MRI-TA were statistically consistent with histology. MRI-TA can be considered as a potentially interesting, reproducible and nondestructive method for muscle examination during atrophy and regeneration

    Serine-threonine kinases and transcription factors active in signal transduction are detected at high levels of phosphorylation during mitosis in preimplantation embryos and trophoblast stem cells

    Serine-threonine kinases and transcription factors play important roles in the G1-S phase progression of the cell cycle. Assays that use quantitative ïŹ‚uorescence by immunocytochemical means, or that measure band strength during Western blot analysis, may have confused interpretations if the intention is to measure G1-S phase commitment of a small subpopulation of phosphorylated proteins, when a larger conversion of the same population of proteins can occur during late G2 and M phases. In mouse trophoblast stem cells (TSC), a human placental cell line (HTR), and/or mouse preimplantation embryos, 8/19 ser- ine-threonine and tyrosine kinases, 3/8 transcription factors, and 8/14 phospho substrate and miscellaneous proteins were phosphorylated at higher levels in M phase than in interphase. Most phosphoproteins appeared to associate with the spindle complex during M phase, but one (p38MAPK) associated with the spindle pole and ïŹve (Cdx2, MEK1, 2, p27, and RSK1) associated with the DNA. Phosphorylation was detected throughout apparent metaphase, anaphase and telophase for some proteins, or for only one of these segments for others. The phosphorylation was from 2.1- to 6.2-fold higher during M phase compared with interphase. These data suggest that, when planning and interpreting quantitative data and perturbation experiments, consideration must be given to the role of serine-threonine kinases and transcription factors during decision making in M phase as well as in G1-S phase

    Additive manufacturing of powder components based on subtractive sintering approach

    International audiencePurpose The purpose of this work is to introduce a novel approach of using additive manufacturing (AM) to produce dense complex ceramic and metallic parts. Powder 3D printing has been gaining popularity due to its ease of use and versatility. However, powder-based methods such as Selective Laser Melting (SLM) and Sintering (SLS), utilizes high power lasers which generate thermal shock conditions in metals and are not ideal for ceramics due to their high melting temperature. Indirect additive manufacturing methods have been explored to address the above issues but have proven to be wasteful and time-consuming. Design/methodology/approach In this work, a novel approach of producing high density net-shaped prototypes using subtractive sintering (SS) and solvent jetting is developed. AM combined with SS (AM-SS) is a process that includes five simple steps. AM-SS can produce repeatable and reliable results as has been shown in this work. Findings As a proof-of-concept, a zirconia dental crown with a high density of 97% is fabricated using this approach. Microstructure and properties of the fabricated components are analyzed. Originality/value A major advantage of this method is the ability to efficiently fabricate high density parts using either metal powder and more importantly, ceramic powder which is traditionally difficult to densify using AM. Additionally, any powder particle size (including nano) and shape can be used which is not the case for traditional powder-based 3D printing

Aussi l'optimisation des fonctions manducatrices par l'intermédiaire de fonctions occlusales "économes" reste un objectif de bon sens de tout traitement dentaire. Le bon sens se fondant sur les principes élémentaires de simplicité et d'ergonomie qui président à l'ensemble des "mécanismes" biologiques. Le respect du principe "centrage, calage, guidage" définit simplement mais précisément des rapports occlusaux induisant une stabilité mandibulaire facilitée (calage) et des fonctions globalement symétriques (centrage) et non verrouillées (guidage)

    Rehabilitation of children with ectodermal dysplasia. Part 1: An international delphi study

    Purpose: An international Delphi study was undertaken to determine by consensus an agreed approach to the management of children with dental manifestations of ectodermal dysplasia, including the use of dental implants. This was done using a questionnaire developed by an interdisciplinary team. Materials and Methods: The Delphi study questionnaire was built around 19 areas of clinical relevance and included 90 items. Topic areas included dental disability; initial diagnosis; global disability; oral health aspects of dental treatment (orthodontics, hypodontia, anodontia, implants); and case studies of selected treatment options. Eleven teams from six countries contributed to three iterations of the questionnaire. An algorithm was designed to standardize analysis of the questionnaire answers, all of which were blinded to ensure anonymity. The second and third rounds of the questionnaire excluded previously agreed-upon items but included the responses to the questions from the earlier rounds. The nonconsensus items inquired about the use of radiographs at initial diagnosis; sedation of an uncooperative child; use of a pretreatment questionnaire; the age range for specific treatments (eg, dentures, orthodontics, implants); specific uses of implants (eg, partial prostheses, overdentures, cantilevered prostheses); and case study 2. The residual nonconsensus questions were subsequently discussed at a 2-day meeting. Results: Among the 90 questions and partial questions, there was progressive consensus, with agreements in rounds 1, 2, and 3 of 61%, 21%, and 8%, respectively. At the conclusion of round 3, there was 90% agreement and it was considered that the nonconsensus items required in-depth face-to-face discussion at a consensus meeting, which is described in part 2 of the study. Conclusion: The Delphi study provided an opportunity to engage specialist teams in recognized centers to integrate their clinical knowledge and draw on published data to develop a consensus of evidencebased responses. © 2013 by Quintessence Publishing Co Inc
