    Characterization and evaluation of antifungal activity in vitro of Aloe vera fractions against postharvest fungi

    Aloe vera leaves can be separated in a liquid fraction (exudate) and a mucilaginous pulp (gel). Recently, the antifungal activity of their components has been reported. The common technique for gel extraction from A. vera leaves are the traditional manual (in this case the liquid fraction is not separated) and a mechanical methods. Moreover, as a result of its processing the bagasse is obtained. The aim of this work was to recover and characterize the fractions of A. vera and to identify the fraction with highest antifungal activity against phytopathogen postharvest fungi (Botrytis cinerea and Penicillium spp.). A simple and inexpensive extraction method was used to obtain A. vera fractions from 50 kg leaves by means of a designed laboratory roll processor. The yields of extraction were as follows: 15.76% ± 4.0, 51.20% ± 5.20, and 33.02% ± 5.0 for gel, liquid fraction and bagasse, respectively. The three fractions were physico-chemically characterized (protein, monosaccharide composition, ashes, lipids) and resulted to be mainly composed by glucose and mannose in all the cases.     [Resumo] Apresentam-se os primeiros resultados de um levantamento cartográfico do pormenor das formas e depósitos glaciários e periglaciários da Serra do Geres-Xurés, levado a efeito por urna equipa de investigadores portugueses e espanhois. Os levantamentos de campo foram realizados nas escalas de 1:5.000 ou 1: 10.000 e tiveram urna importante componente sedimentológica, nomeadamente pelo estudo dos tzlls subglaciários, que se revelaram da maior importancia para o diagnóstico preciso de formas de erosao e de acumula~ao quase sempre pouco nítidas. Esse levantamento nao está ainda completo, mas é possível, desde já, deduzir várias conclus5es acerca da glacia~aoe da morfogénese periglaciária, que sao indicadas no final do texto principal[Résumé] Nous présentons les premiers résultats du levé cartographique détaillé mené par une équipe de chercheurs portugais et espagnols. Les levés de terrain ont été réalisés aux échelles du 1:5.000 ou du 1.10.000. Une attention particuliere a été accordée al'étude des tzlls sous-glaciaires, indispensables au diagnostic rigoureux de formes souvent trop peu claires. Ce levé pas encore terminé, mais il semble possible, des maintenat, de dégager quelques conclusions: 1) Présence, indiscutable, de manifestations glaciaires, tant du coté du Minho que du coté galicien. Cela confume, donc, idées de Schmidt-Thomé et de Coudé-Gaussen. 2) Les formes d' érosion et d' accumulation conservées révelent une glaciation essentiellement de cirque et de vallée. Dans une hypothése de glaciation restreinte, l'épaisseur maximale des langues glaciaires serait de l'ordre de 150 m. Mais ces manifestations représentent, tres probablement, un épisode de retrait des glaciers. L'étude des tills sous-glaciaires suggere, en effet, une glaciation plus ample, avec formation probable d'une calotte de plateau. 3) La répartition spatiale des manifestations glaciaires canographiées montre deux types de contraintes: a) structural, qui commande les directions de grandes vallées englacées; b) climatique, révélant une dyssimétrie NW-SE, avec accumulation préférentielle des neiges du coté oriental (probablement due aun effet d'abri vis-a-vis des vents dominants). 4) Absence d'une morphologie glaciaire typique, ce qui signifie que l'action des glaciers a été peu efficace; elle s'est limitée, pratiquement, a la destruetion et au transport des manteaux d'altérites pré-glaciaires. 5) Monotonie des manÍfestations periglaciaires; il s'agit, surtout, de versant réglés, avec tabliers d'éboulis, et de quelques coulées de solif1uxi6n. Mais c'est ala dynamique cryonivale que revient l'essentiel de l'a1lure des grands versants de la montagne. 6) Absence de pergélisol. Les éboulis et les blocs des grands versants ne sont pas dus ala macrogélifraction. Les actions cryonivales se sont certainement limitées au déchaussemet des blocs, déja préfigurés dans la roche par la fracturation tectonique et par la météoris~tion pré-glaciaire. 7) Absence d'une séparation nette entre l'étage périglaciaire et l'étage glaciaire, al'exception pres du versant galicien. Tout au plus peut-on parler d'un étage forestier (<<étage des arenes», COUDE-GAUSSEN, 1979, 1981), OU les actions cryonivales seraient tres limitees, et d'un étage «glacionival» (ETIICHER, 1988), pres de la limite des neiges persistante

    Simultaneous saccharification and fermentation of hydrothermal pretreated lignocellulosic biomass: evaluation of process performance under multiple stress conditions

    Industrial lignocellulosic bioethanol processes are exposed to different environmental stresses (such as inhibitor compounds, high temperature, and high solid loadings). In this study, a systematic approach was followed where the liquid and solid fractions were mixed to evaluate the influence of varied solid loadings, and different percentages of liquor were used as liquid fraction to determine inhibitor effect. Ethanol production by simultaneous saccharification and fermentation (SSF) of hydrothermally pretreated Eucalyptus globulus wood (EGW) was studied under combined diverse stress operating conditions (3038 °C, 6080 g of liquor from hydrothermal treatment or autohydrolysis (containing inhibitor compounds)/100 g of liquid and liquid to solid ratio between 4 and 6.4 g liquid in SSF/g unwashed pretreated EGW) using an industrial Saccharomyces cerevisiae strain supplemented with low-cost byproducts derived from agro-food industry. Evaluation of these variables revealed that the combination of temperature and higher solid loadings was the most significant variable affecting final ethanol concentration and cellulose to ethanol conversion, whereas solid and autohydrolysis liquor loadings had the most significant impact on ethanol productivity. After optimization, an ethanol concentration of 54 g/L (corresponding to 85 % of conversion and 0.51 g/Lh of productivity at 96 h) was obtained at 37 °C using 60 % of autohydrolysis liquor and 16 % solid loading (liquid to solid ratio of 6.4 g/g). The selection of a suitable strain along with nutritional supplementation enabled to produce noticeable ethanol titers in quite restrictive SSF operating conditions, which can reduce operating cost and boost the economic feasibility of lignocellulose-to-ethanol processes.The authors thank the financial support from the Strategic Project of UID/BIO/04469/2013 CEB Unit and A Romaní postdoctoral grant funded by Xunta of Galicia (Plan I2C, 2014)

    Origin, accumulation and fate of dissolved organic matter in an extreme hypersaline shallow lake

    Hypersaline endorheic aquatic systems (H-SEAS) are lakes/shallow playas in arid and semiarid regions that undergo extreme oscillations in salinity and severe drought episodes. Although their geochemical uniqueness and microbiome have been deeply studied, very little is known about the availability and quality of dissolved organic matter (DOM) in the water column. A H-SEAS from the Monegros Desert (Zaragoza, NE Spain) was studied during a hydrological wetting-drying-rewetting cycle. DOM analysis included: (i) a dissolved organic carbon (DOC) mass balance; (ii) spectroscopy (absorbance and fluorescence) and (iii) a molecular characterization with Fourier-transform ion cyclotron resonance mass spectrometry (FT-ICR-MS). The studied system stored a large amount of DOC and under the highest salinity conditions, salt-saturated waters (i.e., brines with salinity > 30%) accumulated a disproportionate quantity of DOC, indicating a significant in-situ net DOM production. Simultaneously, during the hydrological transition from wet to dry, the DOM pool showed strong alterations of it molecular composition. Spectroscopic methods indicated that aromatic and degraded DOM was rapidly replaced by fresher, relatively small, microbial-derived moieties with a large C/N ratio. FT-ICR-MS highlighted the accumulation of small, saturated and oxidized molecules (molecular O/C > 0.5), with a remarkable increase in the relative contribution of highly oxygenated (molecular O/C > 0.9) compounds and a decrease of aliphatic and carboxyl-rich alicyclic moleculesThese results indicated that H-SEAS are extremely active in accumulating and processing DOM, with the notable release of organic solutes probably originated from decaying microplankton under large osmotic stress at extremely high salinitie

    Effect of hemicellulose liquid phase on the enzymatic hydrolysis of autohydrolyzed Eucalyptus globulus wood

    In this work, Eucalyptus globulus wood was pretreated under non-isothermal autohydrolysis process at 210, 220, and 230 °C, obtaining a pretreated solid with high cellulose content and a hemicellulosic liquid phase (HLP) containing mainly xylose, acetic acid, furfural, xylooligosaccharides, and phenolic compounds. The maximum concentration of xylooligosaccharides (8.97 g/L) and phenolic compounds (2.66 g/L) was obtained at 210 and 230 °C, respectively. To evaluate the effect of HLP addition on the enzymatic hydrolysis using unwashed pretreated solid as substrate, different proportions of HLP were studied. Also, in order to use the whole slurry on enzymatic hydrolysis, the supplementation of xylanases was evaluated. Glucose concentration of 107.49 g/L (corresponding to 74.65 % of conversion) was obtained using pretreated solid at 220 °C liquid/solid ratio (LSR) of 4 g/g and enzyme solid ratio (ESR) of 25 FPU/gwithout the addition of HLP. Thus, it was shown that the unwashed pretreated solids are susceptible to enzymatic hydrolysis contributing to reduce operational cost (water consumption). Additionally, the influence of the inhibitory compounds in the HLP was shown to affect the enzymatic hydrolysis. Results indicated that 82.52 g/L of glucose (59.37 % of conversion) was obtained, using 100 % of HLP at LSR of 4 g/g and ESR of 16 FPU/g at 210 °C of pretreated solid. However, a positive effect was shown on the enzymatic hydrolysis when the xylanases were added using 100 % of HLP, increasing to 35 and 27 % in the glucose production with respect to whole slurry without addition of xylanases.The authors A.     Recombinant family 3 carbohydrate-binding module as a new additive for enhanced enzymatic saccharification of whole slurry from autohydrolyzed eucalyptus globulus wood

    By-products resulting from lignocellulosics pretreatment affect the digestibility of resulting whole slurries, but this can be minimized by additives supplementation. In this work, a family 3 carbohydrate-binding module (CBM3), recombinantly produced from Escherichia coli, was used as additive in the enzymatic hydrolysis of the whole slurry from autohydrolyzed Eucalyptus globulus wood (EGW). At the higher dosage used (30 mg/gsolids), CBM3 led to an increase in glucose yield from 75 to 89%. A similar result was obtained for bovine serum albumin (BSA) (11% increase), which has a well-documented additive effect. CBM3 had no effect on the non-productive binding of enzymes, since it could not bind to EGW lignin, while it rapidly bound to cellulose, as shown by fluorescence microscopy.     Determinants on an efficient cellulase recycling process for the production of bioethanol from recycled paper sludge under high solid loadings

    Background: In spite of the continuous efforts and investments in the last decades, lignocellulosic ethanol is still not economically competitive with fossil fuels. Optimization is still required in different parts of the process. Namely, the cost effective usage of enzymes has been pursued by different strategies, one of them being recycling. Results: Cellulase recycling was analyzed on Recycled Paper Sludge (RPS) conversion into bioethanol under intensified conditions. Different cocktails were studied regarding thermostability, hydrolysis efficiency, distribution in the multiphasic system and recovery from solid. Celluclast showed inferior stability at higher temperatures (45-55 ºC), nevertheless its performance at moderate temperatures (40ºC) was slightly superior to other cocktails (ACCELLERASE®1500 and Cellic®CTec2). Celluclast distribution in the solid-liquid medium was also more favorable, enabling to recover 88 % of final activity at the end of the process. A Central Composite Design studied the influence of solids concentration and enzyme dosage on RPS conversion by Celluclast. Solids concentration showed a significant positive effect on glucose production, no major limitations being found from utilizing high amounts of solids under the studied conditions. Increasing enzyme loading from 20 to 30 FPU/ gcellulose had no significant effect on sugars production, suggesting that 22 % solids and 20 FPU/gcellulose are the best operational conditions towards an intensified process. Applying these, a system of multiple rounds of hydrolysis with enzyme recycling was implemented, allowing to maintain steady levels of enzyme activity with only 50 % of enzyme on each recycling stage. Additionally, interesting levels of solid conversion (70-81 %) were also achieved, leading to considerable improvements on glucose and ethanol production comparatively with the reports available so far (3.4 and 3.8 fold, respectively). Conclusions: Enzyme recycling viability depends on enzyme distribution between the solid and liquid phases at the end of hydrolysis, as well as enzymes thermostability. Both are critical features to be observed for a judicious choice of enzyme cocktail. This work demonstrates that enzyme recycling in intensified biomass degradation can be achieved through simple means.     Fractionation of eucalyptus globulus wood by glycerol-water pretreatment: optimization and modeling

    A glycerol-organosolv process can be a good alternative for Eucalyptus wood fractionation into its main compounds, improving the enzymatic saccharification of the cellulose. A study of process variables - glycerol−water percent content, temperature, and process time - was carried out using a Box-Behnken experimental design. The cellulose obtained from pretreated solids was recovered almost quantitatively, leading to a solid with a high percentage of cellulose (77 g/100 g of pretreated solid), low lignin content (9 g/100 g of pretreated solid), and 18% of residual hemicellulose in the solid at 200 °C, 56% of glycerol−water and 69 min. The enzymatic saccharification was enhanced achieving 98% cellulose-to-glucose conversion (under conditions: liquid to solid ratio 20 g/g and enzyme loading 20 FPU/g of solid). This study contributes to the improvement of biomass fractionation by exploring an eco-friendly treatment which allows for almost complete wood fractionation into constituents and high levels of glucose recovery available for subsequent yeast fermentation to bioethanol.The authors A.     Paleogenomic Evidence for Multi-generational Mixing between Neolithic Farmers and Mesolithic Hunter-Gatherers in the Lower Danube Basin

    The transition from hunting and gathering to farming involved profound cultural and technological changes. In Western and Central Europe, these changes occurred rapidly and synchronously after the arrival of early farmers of Anatolian origin [1-3], who largely replaced the local Mesolithic hunter-gatherers [1, 4-6]. Further east, in the Baltic region, the transition was gradual, with little or no genetic input from incoming farmers [7]. Here we use ancient DNA to investigate the relationship between hunter-gatherers and farmers in the Lower Danube basin, a geographically intermediate area that is characterized by a rapid Neolithic transition but also by the presence of archaeological evidence that points to cultural exchange, and thus possible admixture, between hunter-gatherers and farmers. We recovered four human paleogenomes (1.1× to 4.1× coverage) from Romania spanning a time transect between 8.8 thousand years ago (kya) and 5.4 kya and supplemented them with two Mesolithic genomes (1.7× and 5.3×) from Spain to provide further context on the genetic background of Mesolithic Europe. Our results show major Western hunter-gatherer (WHG) ancestry in a Romanian Eneolithic sample with a minor, but sizeable, contribution from Anatolian farmers, suggesting multiple admixture events between hunter-gatherers and farmers. Dietary stable-isotope analysis of this sample suggests a mixed terrestrial/aquatic diet. Our results provide support for complex interactions among hunter-gatherers and farmers in the Danube basin, demonstrating that in some regions, demic and cultural diffusion were not mutually exclusive, but merely the ends of a continuum for the process of Neolithization.This research was supported by a European Research Council (ERC) Starting Grant (ERC-2010-StG 263441) to R.P.
