    Homeostatic response under carcinogen withdrawal, heme oxygenase 1 expression and cell cycle association

    BACKGROUND: Chronic injury deregulates cellular homeostasis and induces a number of alterations leading to disruption of cellular processes such as cell cycle checkpoints and apoptosis, driving to carcinogenesis. The stress protein heme oxygenase-1 (HO-1) catalyzes heme degradation producing biliverdin, iron and CO. Induction of HO-1 has been suggested to be essential for a controlled cell growth. The aim of this work was to analyze the in vivo homeostatic response (HR) triggered by the withdrawal of a potent carcinogen, p-dimethylaminoazobenzene (DAB), after preneoplastic lesions were observed. We analyzed HO-1 cellular localization and the expression of HO-1, Bcl-2 and cell cycle related proteins under these conditions comparing them to hepatocellular carcinoma (HC). METHODS: The intoxication protocol was designed based on previous studies demonstrating that preneoplastic lesions were evident after 89 days of chemical carcinogen administration. Male CF1 mice (n = 18) were used. HR group received DAB (0.5 % w/w) in the diet for 78 days followed by 11 days of carcinogen deprivation. The HC group received the carcinogen and control animals the standard diet during 89 days. The expression of cell cycle related proteins, of Bcl-2 and of HO-1 were analyzed by western blot. The cellular localization and expression of HO-1 were detected by immnunohistochemistry. RESULTS: Increased expression of cyclin E/CDK2 was observed in HR, thus implicating cyclin E/CDK2 in the liver regenerative process. p21(cip1/waf1 )and Bcl-2 induction in HC was restituted to basal levels in HR. A similar response profile was found for HO-1 expression levels, showing a lower oxidative status in the carcinogen-deprived liver. The immunohistochemical studies revealed the presence of macrophages surrounding foci of necrosis and nodular lesions in HR indicative of an inflammatory response. Furthermore, regenerative cells displayed changes in type, size and intensity of HO-1 immunostaining. CONCLUSION: These results demonstrate that the regenerative capacity of the liver is still observed in the pre-neoplastic tissue after carcinogen withdrawal suggesting that reversible mechanism/s to compensate necrosis and to restitute homeostasis are involved

    Advanced application of transient stability constrained-optimal power flow to a transmission system including an HVDC-LCC link

    In this study, a transient stability constrained-optimal power flow (TSC-OPF) model is proposed and applied to a real case in the currently interconnected Balearic Islands-Iberian Peninsula system. The TSC-OPF retains the dynamics of all generators in the islands with fourth degree transient synchronous generator models and includes a representation of a high-voltage direct current (HVDC) link on the inverter side. The proposed large-scale non-linear optimisation problem, programmed in GAMS and solved using the CONOPT tool, is used to assess the economic impact of the HVDC on the Balearic Islands generation cost under different circumstances, using TSC-OPF and traditional OPF on the dispatch.     Mission analysis for two potential asteroids threat scenarios: Optimal impact strategies and technology evaluation

    The Space Mission Planning Advisory Group SMPAG's mission is to prepare for an international response to a Near Earth Object impact threat through the exchange of information, development of options for collaborative research and mission opportunities, and to conduct Near Earth Object (NEO) impact threat mitigation planning activities. This paper presents the preliminary work performed by the Italian Space Agency Delegation for defining few reference missions for different NEO-threat scenarios and carrying out Phase 0 studies. In this paper two scenarios are identified to study the possible response in case of a real NEO-threat. A direct and resonant impact scenario for an asteroid deflection mission are identified resembling to the asteroid 2010RF12 but with an increased asteroid mass. Then the mission analysis and spacecraft design for the direct impact case is performed and the results discussed

    Effect of base–acid properties of the mixtures of water with methanol on the solution enthalpy of selected cyclic ethers in this mixture at 298.15 K

    The enthalpies of solution of cyclic ethers: 1,4- dioxane, 12-crown-4 and 18-crown-6 in the mixture of water and methanol have been measured within the whole mole fraction range at T = 298.15 K. Based on the obtained data, the effect of base–acid properties of water– methanol mixtures on the solution enthalpy of cyclic ethers in these mixtures has been analyzed. The solution enthalpy of cyclic ethers depends on acid properties of water– methanol mixtures in the range of high and medium water contents in the mixture.     Design, construction, and test of the Gas Pixel Detectors for the IXPE mission

    Due to be launched in late 2021, the Imaging X-Ray Polarimetry Explorer (IXPE) is a NASA Small Explorer mission designed to perform polarization measurements in the 2-8 keV band, complemented with imaging, spectroscopy and timing capabilities. At the heart of the focal plane is a set of three polarization-sensitive Gas Pixel Detectors (GPD), each based on a custom ASIC acting as a charge-collecting anode. In this paper we shall review the design, manufacturing, and test of the IXPE focal-plane detectors, with particular emphasis on the connection between the science drivers, the performance metrics and the operational aspects. We shall present a thorough characterization of the GPDs in terms of effective noise, trigger efficiency, dead time, uniformity of response, and spectral and polarimetric performance. In addition, we shall discuss in detail a number of instrumental effects that are relevant for high-level science analysis -- particularly as far as the response to unpolarized radiation and the stability in time are concerned.Comment: To be published in Astroparticle Physic
