We bred a resistant strain to MPS by selection to decrease MPS pathogenic condition over 5 generations using Landrase (MPS strain), and a high immune performance strain by selection for peripheral phagocytosis, complement activity and antibody production against erysipelatous vaccine using Large Yorkshire (HI strain)

    The effect of dexamethasone on defective nephrin transport caused by ER stress: A potential mechanism for the therapeutic action of glucocorticoids in the acquired glomerular diseases

    The mechanism by which glucocorticoids govern antiproteinuric effect in nephrotic syndrome remains unknown. Present study examined the protective role of dexamethasone (DEX) in the intracellular trafficking of nephrin under endoplasmic reticulum (ER) stress. Human embryonic kidney-293 cell line expressing a full-length human nephrin was cultured in mediums containing 5.5 or 25 mM glucose with or without DEX. The result revealed that glucose starvation evoked a rapid ER stress leading to formation of underglycosylated nephrin that was remained in the ER as a complex with calreticulin/calnexin. DEX rescued this interfered trafficking through binding to its receptor and stimulating the mitochondrial transcripts and adenosine 5â€Č triphosphate (ATP) production, leading to synthesis of fully glycosylated nephrin. These results suggest that ER-stress in podocytes may cause alteration of nephrin N-glycosylation, which may be an underlying factor in the pathomechanism of the proteinuria in nephrotic syndrome. DEX may restore this imbalance by stimulating expression of mitochondrial genes, resulted in the production of ATP that is essential factor for proper folding machinery aided by the ER chaperones

    Experimental real-time optimization of a solid oxide fuel cell stack via constraint adaptation

    The experimental validation of a real-time optimization (RTO) strategy for the optimal operation of a solid oxide fuel cell (SOFC) stack is reported in this paper. Unlike many existing studies, the RTO approach presented here utilizes the constraint-adaptation methodology, which assumes that the optimal operating point lies on a set of active constraints and then seeks to satisfy those constraints in practice via the addition of a correction term to each constraint function. These correction terms, also referred to as “modifiers”, correspond to the difference between predicted and measured constraint values and are updated at each steady-state iteration, thereby allowing the RTO to iteratively meet the optimal operating conditions of an SOFC stack despite significant plant-model mismatch. The effects of the filter parameters used in the modifier update and of the RTO frequency on the general performance of the algorithm are also investigated

    MIBG detection of hepatic neuroblastoma: Correlation with CT, US and surgical findings

    Metaiodobenzylguanidine (MIBG) imaging is used in the diagnosis, staging and follow-up of virtually every case of neuroblastoma seen at our institution. Normal sites of MIBG uptake include the liver and therefore difficulties have been predicted and encountered in the diagnosis of hepatic neuroblastoma due to inability to separate abnormally increased tracer deposition from normal hepatic activity. We reviewed every MIBG (I 123 and I 131 ) study performed at our pediatric hospital over a 4 year period encompassing 88 patients, 67 of whom had biopsy proven neuroblastoma. Hepatic findings on MIBG studies were compared with concurrent abdominal CT and US studies in all 67 patients. The clinical records of all patients with abnormal MIBG scans or abnormal CT or US studies of the liver were also reviewed. Eight patients were found to have abnormal liver findings on one or more imaging studies (MIBG, CT, or US). There were 3 true positive MIBG studies, one of which was an early study in a patient who later went on to have one of the false positive studies. Two patients had false positive MIBG scans for liver neuroblastoma.     Production and reliability oriented SOFC cell and stack design

    The paper presents an innovative development methodology for a production and reliability oriented SOFC cell and stack design aiming at improving the stacks robustness, manufacturability, efficiency and cost. Multi-physics models allowed a probabilistic approach to consider statistical variations in production, material and operating parameters for the optimization phase. A methodology for 3D description of spatial distribution of material properties based on a random field models was developed and validated by experiments. Homogenized material models on multiple levels of the SOFC stack were established. The probabilistic models were related to the experimentally obtained properties of base materials to establish a statistical relationship between the material properties and the most relevant load effects. Software algorithms for meta models that allow the detection of relationships between input and output parameters and to perform a sensitivity analysis were developed and implemented. The capabilities of the methodology is illustrated on two practical cases
