    Quantum Stability of (2+1)-Spacetimes with Non-Trivial Topology

    Quantum fields are investigated in the (2+1)-open-universes with non-trivial topologies by the method of images. The universes are locally de Sitter spacetime and anti-de Sitter spacetime. In the present article we study spacetimes whose spatial topologies are a torus with a cusp and a sphere with three cusps as a step toward the more general case. A quantum energy momentum tensor is obtained by the point stripping method. Though the cusps are no singularities, the latter cusps cause the divergence of the quantum field. This suggests that only the latter cusps are quantum mechanically unstable. Of course at the singularity of the background spacetime the quantum field diverges. Also the possibility of the divergence of topological effect by a negative spatial curvature is discussed. Since the volume of the negatively curved space is larger than that of the flat space, one see so many images of a single source by the non-trivial topology. It is confirmed that this divergence does not appear in our models of topologies.     Regulatory policy and pharmaceutical innovation in the United Kingdom after Brexit: initial insights

    Brexit was presented as an opportunity to promote innovation by breaking free from the European Union regulatory framework. Since the beginning of 2021 the Medicines and Healthcare products Regulatory Agency (MHRA) has operated as the independent regulatory agency for the United Kingdom. The MHRA's regulatory activity in 2021 was analyzed and compared to that of other international regulatory bodies. The MHRA remained reliant on EU regulatory decision-making for novel medicines and there were significant regulatory delays for a small number of novel medicines in the UK, the reasons being so far unclear. In addition, the MHRA introduced innovation initiatives, which show early promise for quicker authorization of innovative medicines for cancer and other areas of unmet need. Longer-term observation and analysis is needed to show the full impact of post-Brexit pharmaceutical regulatory policy

    A form of autosomal recessive spastic ataxia (ARSACS) has been described in the Charlevoix and Saguenay regions of Quebec. So far a frameshift and a nonsense mutation have been identified in the SACS gene. The authors report a new mutation (1859insC), leading to a frameshift with a premature termination of the gene product sacsin, in two sisters from consanguineous parents. The phenotype is similar to previously described patients with ARSACS

    Engineering REST-Specific Synthetic PUF Proteins to Control Neuronal Gene Expression: A Combined Experimental and Computational Study

    Regulation of gene transcription is an essential mechanism for differentiation and adaptation of organisms. A key actor in this regulation process is the repressor element 1 (RE1)-silencing transcription factor (REST), a transcriptional repressor that controls more than 2000 putative target genes, most of which are neuron-specific. With the purpose of modulating REST expression, we exploited synthetic, ad hoc designed, RNA binding proteins (RBPs) able to specifically target and dock to REST mRNA. Among the various families of RBPs, we focused on the Pumilio and FBF (PUF) proteins, present in all eukaryotic organisms and controlling a variety of cellular functions. Here, a combined experimental and computational approach was used to design and test 8- and 16-repeat PUF proteins specific for REST mRNA. We explored the conformational properties and atomic features of the PUF-RNA recognition code by Molecular Dynamics simulations. Biochemical assays revealed that the 8- and 16-repeat PUF-based variants specifically bind the endogenous REST mRNA without affecting its translational regulation. The data also indicate a key role of stacking residues in determining the binding specificity. The newly characterized REST-specific PUF-based constructs act as excellent RNA-binding modules and represent a versatile and functional platform to specifically target REST mRNA and modulate its endogenous expression

    Differences in the oxidative balance of dispersing and non-dispersing individuals: an experimental approach in a passerine bird.

    Dispersal is often associated with a suite of phenotypic traits that might reduce dispersal costs, but can be energetically costly themselves outside dispersal. Hence, dispersing and philopatric individuals might differ throughout their life cycle in their management of energy production. Because higher energy expenditure can lead to the production of highly reactive oxidative molecules that are deleterious to the organism if left uncontrolled, dispersing and philopatric individuals might differ in their management of oxidative balance. Here, we experimentally increased flight costs during reproduction via a wing load manipulation in female collared flycatchers (Ficedula albicollis) breeding in a patchy population. We measured the effects of the manipulation on plasmatic markers of oxidative balance and reproductive success in dispersing and philopatric females. The impact of the wing load manipulation on the oxidative balance differed according to dispersal status. The concentration of reactive oxygen metabolites (ROMs), a marker of pro-oxidant status, was higher in philopatric than dispersing females in the manipulated group only. Differences between dispersing and philopatric individuals also depended on habitat quality, as measured by local breeding density. In low quality habitats, ROMs as well as nestling body mass were higher in philopatric females compared to dispersing ones. Independently of the manipulation or of habitat quality, plasma antioxidant capacity differed according to dispersal status: philopatric females showed higher antioxidant capacity than dispersing ones. Nestlings raised by philopatric females also had a higher fledging success. Our results suggest that dispersing individuals maintain a stable oxidative balance when facing challenging environmental conditions, at the cost of lower reproductive success. Conversely, philopatric individuals increase their effort, and thus oxidative costs, in challenging conditions thereby maintaining their reproductive success. Our study sheds light on energetics and oxidative balance as possible processes underlying phenotypic differences between dispersing and philopatric individuals

    Lançado em 2018 e construído a partir do uso de softwares livres, o GeoInfo é atualmente a Infraestrutura de Dados Espaciais institucional da Embrapa. O GeoInfo é responsável tanto pela catalogação e pelo armazenamento quanto pela disponibilização/publicação desses dados, a partir dos padrões internacionais de compartilhamento de dados estabelecidos pela Open Geospatial Consortium (OGC) e dos princípios Free Open Source (FOSS). Ele está disponível para diferentes públicos (interno e externo) e integra-se com a Infraestrutura Nacional de Dados Espaciais (Inde). O GeoInfo agrega dados de 26 Unidades Descentralizadas da Embrapa que são produtoras e usuárias de dados geográficos. Após cinco anos de implantação e cerca de 12.000 conjuntos de dados catalogados, o GeoInfo passa atualmente por um processo de avaliação e diagnóstico, que visa nortear o planejamento de ações futuras sobre sua evolução e manutenção. Este trabalho tem o objetivo de apresentar esse diagnóstico e suas implicações na proposição de uma nova arquitetura para o GeoInfo. Trata-se de análises feitas nos metadados dos dados catalogados na plataforma, com a finalidade de compreender seu perfil de utilização.CIIC 2023. Nº 23509

    Caspase 3 and 8 deficiency in human neuroblastoma

    An altered apoptotic response represents a pivotal feature of cancer and is involved in cancerogenesis and resistance to chemotherapy. So far, however, only a few studies have been devoted to survey caspase content in malignant cell lines and primary tumor specimens. In this report, we investigated the expression of two pivotal caspases, 3 and 8, in 63 neuroblastoma specimens by three complementary techniques (i.e., reverse transcriptase polymerase chain reaction, immunoblotting, and immunohistochemistry). We confirmed the frequent absence of caspase 8 expression. Moreover and most important, we demonstrated, for the first time to our knowledge, that a significant percentage of neuroblastomas lack caspase 3 mRNA and protein. Both caspase alterations do not show any correlation with tumor stage and MYCN status. Immunohistochemistry showed a large number of caspase-negative cell islets also present in positive samples. Our findings suggest that the absence of caspases might play an important role in neuroblastoma development and resistance to apoptosisbased treatments
