    Mobile DNA and evolution in the 21st century

    Scientific history has had a profound effect on the theories of evolution. At the beginning of the 21st century, molecular cell biology has revealed a dense structure of information-processing networks that use the genome as an interactive read-write (RW) memory system rather than an organism blueprint. Genome sequencing has documented the importance of mobile DNA activities and major genome restructuring events at key junctures in evolution: exon shuffling, changes in cis-regulatory sites, horizontal transfer, cell fusions and whole genome doublings (WGDs). The natural genetic engineering functions that mediate genome restructuring are activated by multiple stimuli, in particular by events similar to those found in the DNA record: microbial infection and interspecific hybridization leading to the formation of allotetraploids. These molecular genetic discoveries, plus a consideration of how mobile DNA rearrangements increase the efficiency of generating functional genomic novelties, make it possible to formulate a 21st century view of interactive evolutionary processes. This view integrates contemporary knowledge of the molecular basis of genetic change, major genome events in evolution, and stimuli that activate DNA restructuring with classical cytogenetic understanding about the role of hybridization in species diversification

    The Guidelines’ policy behind counting most diversionary dispositions as if they were criminal convictions is that “defendants who receive the benefit of a rehabilitative sentence and continue to commit crimes should not be treated with further leniency.”15 However, it is not “further leniency” to treat a state diversionary disposition in a way that state statutes say they should not be treated, thereby ratcheting up a federal defendant’s sentence. “Further leniency” would be to give a federal defendant another diversionary disposition for the federal crime. It is not principled for the Guidelines to undo the original leniency of the state diversionary disposition by counting it the same way a conviction would be counted...The purpose of diversionary dispositions is to give an offender the chance to abide by the law during the diversionary period. If he behaves during that period, the record is supposed to be effectively “wiped clean.” If not, then the diversionary disposition turns into a conviction. If an offender commits another offense after he successfully completes the diversionary disposition, then it is a legitimate use of the first diversion not to give him another one. But it is not legitimate to use a successfully completed diversionary disposition as if it were a conviction to aggravate a subsequent sentence, in violation of express statutory language

    Butterflies in the family Lycaenidac are often the focus of conservation efforts. However, our understanding of lycaenid population dynamics has been limited to relatively few examples of long-term monitoring data that have been reported. Here, factors associated with population regulation are investigated using a complete record of a single population of the silvery blue, Glaucopsyche lygdamus Doubleday (Lepidoptera: Lycaenidae). Adults of G. lygdamus were first observed in an annual grassland near Davis, California, in 1982 and were last seen in 2003. Relationships between inter-annual variation in abundance and climatic variables were examined, accounting for density dependent effects. Significant effects of both negative density dependence and climatic variation were detected, particularly precipitation and temperature during winter months. Variation in precipitation, the strongest predictor of abundance, was associated directly and positively with butterfly abundance in the same year. Winter temperatures had a negative effect in the same year, but had a lagged, positive effect on abundance in the subsequent year. Mechanistic hypotheses are posed that include climatic effects mediated through both larval and adult plant resources

    Alemtuzumab pre-conditioning with tacrolimus monotherapy in pediatric renal transplantation

    We employed antibody pre-conditioning with alemtuzumab and posttransplant immunosuppression with low-dose tacrolimus monotherapy in 26 consecutive pediatric kidney transplant recipients between January 2004 and December 2005. Mean recipient age was 10.7 ± 5.8 years, 7.7% were undergoing retransplantation, and 3.8% were sensitized, with a PRA >20%. Mean donor age was 32.8 ± 9.2 years. Living donors were utilized in 65% of the transplants. Mean cold ischemia time was 27.6 ± 6.4 h. The mean number of HLA mismatches was 3.3 ± 1.3. Mean follow-up was 25 ± 8 months. One and 2 year patient survival was 100% and 96%. One and 2 year graft survival was 96% and 88%. Mean serum creatinine was 1.1 ± 0.6 mg/dL, and calculated creatinine clearance was 82.3 ± 29.4 mL/min/1.73 m 2. The incidence of pre-weaning acute rejection was 11.5%; the incidence of delayed graft function was 7.7%. Eighteen (69%) of the children were tapered to spaced tacrolimus monotherapy, 10.5 ± 2.2 months after transplantation. The incidence of CMV, PTLD and BK virus was 0%; the incidence of posttransplant diabetes was 7.7%. Although more follow-up is clearly needed, antibody pre-conditioning with alemtuzumab and tacrolimus monotherapy may be a safe and effective regimen in pediatric renal transplantation. © 2007 The Authors

    Early changes in brain structure correlate with language outcomes in children with neonatal encephalopathy.

    Global patterns of brain injury correlate with motor, cognitive, and language outcomes in survivors of neonatal encephalopathy (NE). However, it is still unclear whether local changes in brain structure predict specific deficits. We therefore examined whether differences in brain structure at 6 months of age are associated with neurodevelopmental outcomes in this population. We enrolled 32 children with NE, performed structural brain MR imaging at 6 months, and assessed neurodevelopmental outcomes at 30 months. All subjects underwent T1-weighted imaging at 3 T using a 3D IR-SPGR sequence. Images were normalized in intensity and nonlinearly registered to a template constructed specifically for this population, creating a deformation field map. We then used deformation based morphometry (DBM) to correlate variation in the local volume of gray and white matter with composite scores on the Bayley Scales of Infant and Toddler Development (Bayley-III) at 30 months. Our general linear model included gestational age, sex, birth weight, and treatment with hypothermia as covariates. Regional brain volume was significantly associated with language scores, particularly in perisylvian cortical regions including the left supramarginal gyrus, posterior superior and middle temporal gyri, and right insula, as well as inferior frontoparietal subcortical white matter. We did not find significant correlations between regional brain volume and motor or cognitive scale scores. We conclude that, in children with a history of NE, local changes in the volume of perisylvian gray and white matter at 6 months are correlated with language outcome at 30 months. Quantitative measures of brain volume on early MRI may help identify infants at risk for poor language outcomes

    Numerical Simulations of Thermal Detection of Disbonds in Lap Joints

    The detection of disbonds in riveted lap joints is of increasing interest to the aerospace community. For some cases a disbonded lapjoint gives rise to cracks at the rivets due to stress concentrations in the aluminum skin when the lapjoints are loaded. When a number of these cracks exist they have been known to suddenly link up and cause sudden failure of the part. Thermographic detection of these disbonds has advantages over other methods in that it is a noncontacting, quantitative method which can inspect large areas in a short period of time. By even application of heat to the outer surface of the lap joint, details of the inner structure become thermographically detectable. A disbond between layers of a laminated structure will prevent heat from penetrating from the surface layer to the subsurface layers and will result in an increase in surface temperature over the disbond.</p
