    Knowledge, attitudes, and practices among healthcare providers on cervical cancer, human papilloma virus and it’s vaccine at ESI PGIMSR, MGM Hospital Parel Mumbai, India

    Background: Cervical cancer is the second most common cause of cancer in females Worldwide and nearly 85% of the cervical cancer cases are diagnosed in developing countries. The health care providers can play an important role in promoting cervical cancer screening and knowledge about HPV vaccination. So this study was conducted to assess the knowledge, attitude and practices regarding cervical cancer screening and HPV vaccination and to analyze the factors influencing the knowledge about HPV vaccination, among nursing staff.Methods: A cross-sectional study was conducted among 143 nurses and 75nursing students (total 218) at ESIPGIMSR MGM Hospital, Parel Mumbai. Data was collected using Questionnaire, designed based on the study objectives.Results: 73 (33.49%) of the nurses did received education on cervical cancer and HPV in the past. Most of the nurses, 184 (84.4%) considered that they were at no risk of cervical cancer. 143 (65.60%) nurses knew that Pap smear can be done to screen patients. Out of 141 married nurses, only 39 (27.28%) regularly had a gynaecological examination and 105 (74.47%) had never done Pap smear. In addition, 119 (54.59%) participants did not know at what age Pap smear should commence. Only 73 (33.49%) nurses knew about the route of transmission of HPV and 57 (26.15%) knew about HPV vaccine. 215 (98.62%) nurses did not receive an HPV vaccine.Conclusions: The nurses and nursing students did not have prior adequate education on cervical cancer, HPV, and vaccine and the desired level of knowledge on risk factors and signs of cervical cancer and prevention of the disease

    Prevalence of thyroid disorders among patients attending the antenatal clinic at tertiary care centre, Parel, Mumbai, India

    Background: Pregnancy is associated with major changes in the physiology of the thyroid gland. Undiagnosed thyroid disorders can adversely affect fetal and maternal outcome. This study was done to study the prevalence of thyroid disorder among antenatal women.Methods: This was a prospective study done in the antenatal clinic in the Department of Obstetrics and Gynaecology, ESI PGIMSRMGM Hospital, Parel, Mumbai, India.Results: In present study, fifty out of three hundred and fifty pregnant women screened had thyroid disorder. The prevalence of thyroid disorder in this study was 14.2%. The prevalence of subclinical hypothyroidism was 7.7% (n= 27), overt hypothyroidism was 4% (n=14), subclinical hyperthyroidism was 1.7% (n=6) and overt hyperthyroidism was 0.8% (n=3).Conclusions: Thyroid disorders in pregnancy are associated with adverse fetomaternal outcome. Its prevalence is high in Indian women. Thus thyroid screening should be included in the routine antenatal investigations

    Design and Evaluation of Heterobivalent PAR1–PAR2 Ligands as Antagonists of Calcium Mobilization

    A novel class of bivalent ligands targeting putative protease-activated receptor (PAR) heteromers has been prepared based upon reported antagonists for the subtypes PAR1 and PAR2. Modified versions of the PAR1 antagonist RWJ-58259 containing alkyne adapters were connected via cycloaddition reactions to azide-capped polyethylene glycol (PEG) spacers attached to imidazopyridazine-based PAR2 antagonists. Initial studies of the PAR1–PAR2 antagonists indicated that they inhibited G alpha q-mediated calcium mobilization in endothelial and cancer cells driven by both PAR1 and PAR2 agonists. Compounds of this novel class hold promise for the prevention of restenosis, cancer cell metastasis, and other proliferative disorders

But while taking decision on investment, Investors consider different resources and information, mean while they forget to check their own knowledge on financial terms and wisdom on how to invest and where to invest. It is important to know the effect of financial knowledge on the choice of investment avenues in order to study the reliability of their investment decisions. The problem is to check whether the investment decisions are based upon simple investment tips and opinions of other investors or a rational analysis of risk and return associated with the various investment avenues. In this paper an attempt has been made to evaluate the effect of Financial Knowledge on investment decisions of 200 Investors from Gandhinagar District. The study is based upon primary data collected through a structured questionnaire administered to 100 rural and 100 urban investors, drawn through quota sampling according to their various occupation categories. It is hypothesized that investors take rational decisions based upon their financial knowledge

    The present study used text analytics software to examine the unstructured (or qualitative) data provided by job references during confidential, multi-rater reference checks. With respect to both work-related strengths and areas of improvement, job references more frequently provided words or short phrases relating to “soft skills” such as working with others and communication as opposed to “hard skills” such as computer programming or mathematics. While some commonalities across jobs were found to exist, the frequency of identified categories for both work-related strengths and areas of improvement did differ across jobs

    Monitoring Replication Protein A (RPA) Dynamics in Homologous Recombination Through Site-specific Incorporation of Non-canonical Amino Acids

    An essential coordinator of all DNA metabolic processes is Replication Protein A (RPA). RPA orchestrates these processes by binding to single-stranded DNA (ssDNA) and interacting with several other DNA binding proteins. Determining the real-time kinetics of single players such as RPA in the presence of multiple DNA processors to better understand the associated mechanistic events is technically challenging. To overcome this hurdle, we utilized non-canonical amino acids and bio-orthogonal chemistry to site-specifically incorporate a chemical fluorophore onto a single subunit of heterotrimeric RPA. Upon binding to ssDNA, this fluorescent RPA (RPAf) generates a quantifiable change in fluorescence, thus serving as a reporter of its dynamics on DNA in the presence of multiple other DNA binding proteins. Using RPAf, we describe the kinetics of facilitated self-exchange and exchange by Rad51 and mediator proteins during various stages in homologous recombination. RPAf is widely applicable to investigate its mechanism of action in processes such as DNA replication, repair and telomere maintenance

    Localizing ECoG electrodes on the cortical anatomy without post-implantation imaging

    AbstractIntroductionElectrocorticographic (ECoG) grids are placed subdurally on the cortex in people undergoing cortical resection to delineate eloquent cortex. ECoG signals have high spatial and temporal resolution and thus can be valuable for neuroscientific research. The value of these data is highest when they can be related to the cortical anatomy. Existing methods that establish this relationship rely either on post-implantation imaging using computed tomography (CT), magnetic resonance imaging (MRI) or X-Rays, or on intra-operative photographs. For research purposes, it is desirable to localize ECoG electrodes on the brain anatomy even when post-operative imaging is not available or when intra-operative photographs do not readily identify anatomical landmarks.MethodsWe developed a method to co-register ECoG electrodes to the underlying cortical anatomy using only a pre-operative MRI, a clinical neuronavigation device (such as BrainLab VectorVision), and fiducial markers. To validate our technique, we compared our results to data collected from six subjects who also had post-grid implantation imaging available. We compared the electrode coordinates obtained by our fiducial-based method to those obtained using existing methods, which are based on co-registering pre- and post-grid implantation images.ResultsOur fiducial-based method agreed with the MRI–CT method to within an average of 8.24mm (mean, median=7.10mm) across 6 subjects in 3 dimensions. It showed an average discrepancy of 2.7mm when compared to the results of the intra-operative photograph method in a 2D coordinate system. As this method does not require post-operative imaging such as CTs, our technique should prove useful for research in intra-operative single-stage surgery scenarios.To demonstrate the use of our method, we applied our method during real-time mapping of eloquent cortex during a single-stage surgery. The results demonstrated that our method can be applied intra-operatively in the absence of post-operative imaging to acquire ECoG signals that can be valuable for neuroscientific investigations

    Reliability of Bell’s Test Conducted with Virtual Reality

    Cancellation tasks are commonly used assessment tools to detect unilateral neglect. Bell’s Test, one of the most commonly administered cancellation tasks, requires individuals to quickly and accurately identify “bells” randomly placed in an array of symbols. The reliability of Bells Test conducted with a paper and pencil is well established. A newly developed, commercial software application, allows Bell’s Test to be administered in fully immersive virtual reality environment. PURPOSE: The purpose of this study was to measure the reliability of the Bell’s Test using a virtual reality (VR) system and to establish the level of agreement between the pen-paper and VR administration. METHODS: Fourteen apparently healthy individuals between the age of 24 – 73y volunteered (47.9±20.7y; 166.0±5.2cm; 77.5±16.3kg). Subjects were excluded if they had a history of concussion or had perceptual or visual deficits. Participants completed Bell’s Test a total of four times. The first time, it was completed with a pen-paper (PP). Then, participants completed an initial assessment in VR (VRbase); the same test was administered again in VR one-hour (PostVR1hr) and one-week (PostVR1wk) post baseline. Realization time, total time, and the number of errors committed in the right and left field of view were recorded. Cronbach’s alpha was computed on realization and total time in all VR conditions. Additionally, an ANOVA with repeated measures was used to determine differences in PP, VRbase, PostVR1HR, and PostVR1Wk. The Holms-Sidak method was used to identify pairwise differences. Alpha was set at pa priori. RESULTS: Reliability for realization time for the Bell’s test conducted in VR was acceptable (α=0.79). There was, however, a significant difference between trials (F=6.65; p=0.013). VRbase (29.25±8.11s) was significantly different than PostVR1HR (15.52±4.30; p=0.006). and VRpost1wk (21.24±5.89s; p=0.01). Additionally, PP realization time (15.31±4.25s) was significantly different than PostVR1HR (p=0.02) and PostVR1WK (p=0.03). Reliability for total time for the Bell’s test conducted in VR was good. (α=0.82). There was not a significant difference between the trials (F=4.34; p=0.06) for VRbase(24.1±6.7s), PostVR1HR (45.44±12.6) or VRpost1wk(p=0.06). However, there was a significant difference found in PPtotal time taken and VR1wk with the (p=0.03). The average number of left side omitted bells was 0.50±0.65; 0.07±0.26; 0.07±0.26; 0.14±0.36 for PP, VRbase, Post VR1HR, and PostVR1WK; respectively. The number of bells omitted on the right side was 0.50±0.85, 0.21±0.57, 0.14±0.36, for 0.14±0.36 for PP, VRbase, PostVR1HR, and PostVR1WK; respectively. CONCLUSION: These data indicate that the reliability of the Bell’s Test for the realization time is acceptable, and the total time reliability was good in the virtual reality system. Healthy individuals performing the Bell’s Test in VR show slight improvements one hour after baseline, but there was no difference after one week. More data are needed within different age groups to determine reliability in young and older individuals. Additionally, future studies are required to determine the reliability of Bell’s Test in VR for individuals suffering from neurological injuries or diseases

    Checkpoint Blockade Rescues the Repressive Effect of Histone Deacetylases Inhibitors on γδ T Cell Function

    Histone deacetylases (HDAC) are one of the key epigenetic modifiers that control chromatin accessibility and gene expression. Their role in tumorigenesis is well established and HDAC inhibitors have emerged as an effective treatment modality. HDAC inhibitors have been investigated for their specific antitumor activities and also clinically evaluated in treatment of various malignancies. In the present study, we have investigated the effect of HDAC inhibitors on the effector functions of human γδ T cells. HDAC inhibitors inhibit the antigen-specific proliferative response of γδ T cells and cell cycle progression. In antigen-activated γδ T cells, the expression of transcription factors (Eomes and Tbet) and effector molecules (perforin and granzyme B) were decreased upon treatment with HDAC inhibitors. Treatment with HDAC inhibitors attenuated the antitumor cytotoxic potential of γδ T cells, which correlated with the enhanced expression of immune checkpoints programmed death-1 (PD-1) and programmed death ligand-1 in γδ T cells. Interestingly, PD-1 blockade improves the antitumor effector functions of HDAC inhibitor-treated γδ T cells, which is reflected in the increased expression of Granzyme B and Lamp-1. This study provides a rationale for designing HDAC inhibitor and immune check point blockade as a combinatorial treatment modality for cancer
