Knowledge-based load leveling and task allocation in human-machine systems
Conventional human-machine systems use task allocation policies which are based on the premise of a flexible human operator. This individual is most often required to compensate for and augment the capabilities of the machine. The development of artificial intelligence and improved technologies have allowed for a wider range of task allocation strategies. In response to these issues a Knowledge Based Adaptive Mechanism (KBAM) is proposed for assigning tasks to human and machine in real time, using a load leveling policy. This mechanism employs an online workload assessment and compensation system which is responsive to variations in load through an intelligent interface. This interface consists of a loading strategy reasoner which has access to information about the current status of the human-machine system as well as a database of admissible human/machine loading strategies. Evaluating academic and media nongovernmental organization partnerships for participatory data gathering
This article discusses participatory methods for data gathering in the context of a partnership between a Swiss-based media development organization, Fondation Hirondelle, and a research team at the University of Sheffield. In 2018–2019, the partnership conducted fieldwork which focused on the impact of radio on women listeners in Niger. The project used participatory methods of data gathering in the form of workshops and focus group discussions (FGDs). The article examines the advantages and limitations of combining the practical experience of international development organizations and the in-depth research capabilities of academia. To triangulate this collaboration and to navigate the limitations of FGDs, the use of workshops is discussed as an important method for providing feedback among the radio practitioners and experts in Niger. The article examines the usefulness of combining these methods and reshaping their application to promote participatory research with radio audiences and practitioners
Getting Acquainted with Kant
My question here concerns whether Kant claims that experience has
nonconceptual content, or whether, on his view, experience is
essentially conceptual. However there is a sense in which this debate
concerning the content of intuition is ill-conceived. Part of this has
to do with the terms in which the debate is set, and part to do with
confusion over the connection between Kant’s own views and contemporary
concerns in epistemology and the philosophy of mind. However, I think
much of the substance of the debate concerning Kant’s views on the
content of experience can be salvaged by reframing it in terms of a
debate about the dependence relations, if any, that exist between
different cognitive capacities. Below, in Section 2, I clarify the
notion of ‘content’ I take to be at stake in the interpretive debate.
Section 3 presents reasons for thinking that intuition cannot have
content in the relevant sense. I then argue, in Section 4, that the
debate be reframed in terms of dependence. We should distinguish between
Intellectualism, according to which all objective representation
(understood in a particular way) depends on acts of synthesis by the
intellect, and Sensibilism, according to which at least some forms of
objective representation are independent of any such acts (or the
capacity for such acts). Finally, in Section 5, I further elucidate the
cognitive role of intuition. I articulate a challenge which Kant
understands alethic modal considerations to present for achieving
cognition, and argue that a version of Sensibilism that construes
intuition as a form of acquaintance is better positioned to answer this
challenge than Intellectualism
Comedian Hosts and the Demotic Turn
Podcasting is a showcase for what cultural studies scholar Graeme Turner coined “the demotic turn” or the increasing visibility of the ordinary person in the today’s media landscape. Collins argues that the emergence of a particular breed of podcasts – comedian-hosted interviews with celebrities – function in an “off-label” manner as a form of self-help or vicarious therapy. The emergence and rapid growth of this genre can attributed to three main factors: a confessional culture, the triumph of experience over expertise, and the democratization allowed by the form’s technology. She explores the link between emotional intimacy and comedy, and analyzes podcasts like Marc Maron’s WTF that are, in expression, a rejection of the pedestal version of stardom
Identification of the Biotransformation Products of 2-Ethylhexyl 4-(N,N-Dimethylamino)benzoate
Nowadays, 2-ethylhexyl 4-(N,N-dimethylamino)benzoate (EDP) is one of the most widely used UV filters in sunscreen cosmetics and other cosmetic products. However, undesirable processes such as percutaneous absorption and biological activity have been attributed to this compound. The in vitro metabolism of EDP was elucidated in the present work. First of all, the phase I biotransformation was studied in rat liver microsomes and two metabolites, N,N-dimethyl-p-aminobenzoic acid (DMP) and N-monomethyl-p-aminobenzoic acid (MMP), were identified by GC-MS analysis. Secondly, the phase II metabolism was investigated by means of LC-MS. The investigated reactions were acetylation and glucuronidation working with rat liver cytosol and with both human and rat liver microsomes, respectively. Analogue studies with p-aminobenzoic acid (PABA) were carried out in order to compare the well established metabolic pathway of PABA with the unknown biotransformation of EDP. In addition, a method for the determination of EDP and its two phase I metabolites in human urine was developed. The methodology requires a solid-phase extraction prior to LC-MS analysis. The method is based on standard addition quantification and has been fully validated. The repeatability of the method, expressed as relative standard deviation, was in the range 3.4–7.4% and the limit of detection for all quantified analytes was in the low ng mL−1 range
Evidentialism and Moral Encroachment
Moral encroachment holds that the epistemic justification of a belief can be affected by moral factors. If the belief might wrong a person or group more evidence is required to justify the belief. Moral encroachment thereby opposes evidentialism, and kindred views, which holds that epistemic justification is determined solely by factors pertaining to evidence and truth. In this essay I explain how beliefs such as ‘that woman is probably an administrative assistant’—based on the evidence that most women employees at the firm are administrative assistants—motivate moral encroachment. I then describe weaknesses of moral encroachment. Finally I explain how we can countenance the moral properties of such beliefs without endorsing moral encroachment, and I argue that the moral status of such beliefs cannot be evaluated independently from the understanding in which they are embedded
Modularity in Protein Complex and Drug Interactions Reveals New Polypharmacological Properties
Recent studies have highlighted the importance of interconnectivity in a large range of molecular and human disease-related systems. Network medicine has emerged as a new paradigm to deal with complex diseases. Connections between protein complexes and key diseases have been suggested for decades. However, it was not until recently that protein complexes were identified and classified in sufficient amounts to carry out a large-scale analysis of the human protein complex system. We here present the first systematic and comprehensive set of relationships between protein complexes and associated drugs and analyzed their topological features. The network structure is characterized by a high modularity, both in the bipartite graph and in its projections, indicating that its topology is highly distinct from a random network and that it contains a rich and heterogeneous internal modular structure. To unravel the relationships between modules of protein complexes, drugs and diseases, we investigated in depth the origins of this modular structure in examples of particular diseases. This analysis unveils new associations between diseases and protein complexes and highlights the potential role of polypharmacological drugs, which target multiple cellular functions to combat complex diseases driven by gain-of-function mutations
Kant's conception of proper science
Kant is well known for his restrictive conception of proper science. In the present paper I will try to explain why Kant adopted this conception. I will identify three core conditions which Kant thinks a proper science must satisfy: systematicity, objective grounding, and apodictic certainty. These conditions conform to conditions codified in the Classical Model of Science. Kant's infamous claim that any proper natural science must be mathematical should be understood on the basis of these conditions. In order to substantiate this reading, I will show that only in this way it can be explained why Kant thought (1) that mathematics has a particular foundational function with respect to the natural sciences and (2) as such secures their scientific status. © 2009 The Author(s)
A synergistic antiproliferation effect of curcumin and docosahexaenoic acid in SK-BR-3 breast cancer cells: unique signaling not explained by the effects of either compound alone
<p>Abstract</p> <p>Background</p> <p>Breast cancer is a collection of diseases in which molecular phenotypes can act as both indicators and mediators of therapeutic strategy. Therefore, candidate therapeutics must be assessed in the context of multiple cell lines with known molecular phenotypes. Docosahexaenoic acid (DHA) and curcumin (CCM) are dietary compounds known to antagonize breast cancer cell proliferation. We report that these compounds in combination exert a variable antiproliferative effect across multiple breast cell lines, which is synergistic in SK-BR-3 cells and triggers cell signaling events not predicted by the activity of either compound alone.</p> <p>Methods</p> <p>Dose response curves for CCM and DHA were generated for five breast cell lines. Effects of the DHA+ CCM combination on cell proliferation were evaluated using varying concentrations, at a fixed ratio, of CCM and DHA based on their individual ED<sub>50</sub>. Detection of synergy was performed using nonlinear regression of a sigmoid dose response model and Combination Index approaches. Cell molecular network responses were investigated through whole genome microarray analysis of transcript level changes. Gene expression results were validated by RT-PCR, and western blot analysis was performed for potential signaling mediators. Cellular curcumin uptake, with and without DHA, was analyzed via flow cytometry and HPLC.</p> <p>Results</p> <p>CCM+DHA had an antiproliferative effect in SK-BR-3, MDA-MB-231, MDA-MB-361, MCF7 and MCF10AT cells. The effect was synergistic for SK-BR-3 (ER<sup>- </sup>PR<sup>- </sup>Her2<sup>+</sup>) relative to the two compounds individually. A whole genome microarray approach was used to investigate changes in gene expression for the synergistic effects of CCM+DHA in SK-BR-3 cells lines. CCM+DHA triggered transcript-level responses, in disease-relevant functional categories, that were largely non-overlapping with changes caused by CCM or DHA individually. Genes involved in cell cycle arrest, apoptosis, inhibition of metastasis, and cell adhesion were upregulated, whereas genes involved in cancer development and progression, metastasis, and cell cycle progression were downregulated. Cellular pools of PPARγ and phospho-p53 were increased by CCM+DHA relative to either compound alone. DHA enhanced cellular uptake of CCM in SK-BR-3 cells without significantly enhancing CCM uptake in other cell lines.</p> <p>Conclusions</p> <p>The combination of DHA and CCM is potentially a dietary supplemental treatment for some breast cancers, likely dependent upon molecular phenotype. DHA enhancement of cellular curcumin uptake is one potential mechanism for observed synergy in SK-BR-3 cells; however, transcriptomic data show that the antiproliferation synergy accompanies many signaling events unique to the combined presence of the two compounds.</p
