    Optical microscopes are widely used in science classes from elementary to high schools. The introduction of how to handle a microscope is described in a textbook of science at every grade level. It is important for us in microscopic observations to handle a microscope properly as well as to make a correct preparation of glass slides. In the 6th grade of elementary school, students observe planktons using a microscope with a prepared slide made by him/herself. We have noticed that it is hard to control the amount of water applied with a pipette on a hole slide. If the amount of water is too much, a cover glass was floating on the water, and then it went off the center and turned around. Here, we developed “3D hole slides” using a 3D printer for controlling the amount of water on the slide and making microscopic observations easy. We printed out 5 types of 3D hole slides and examined whether too much water can be drained. A hole slide with a circular moat whose edge on the center side has an inclination of 45 degrees is the best among them to control water samples. Commercial hole glass slides and the developed 3D hole slides were compared by two groups of undergraduates in biological classes at our university, and the effectiveness of the slide preparation was evaluated. The ratio of students who prepared a proper amount of water on the slide was improved from 36% to 63%. If a proper amount of water can be prepared, students can save time without absorbing the rest of the water and can spend more time observing an object. As the 3D hole slide is considered useful for undergraduate students, we will evaluate it for elementary school students, and in the future, we hope it to be popular in science classes.textapplication/pdfdepartmental bulletin pape
