    Trial by Fire: The Marshall-Bell Kiln Site in Fredericksburg, Virginia

    In 2012 and 2013 community members and local professional archaeologists led by Dovetail Cultural Resource Group worked together to salvage a stoneware waster dump in Fredericksburg, Virginia. More than 17,000 artifacts were recovered, representing two successive potters, Hugh R. Marshall and Francis H. Bell. This article brings together the archaeological and documentary evidence to discuss this short-lived pottery operation of the early 1830s. Considered are the physical attributes of the vessel forms and decorations, and the broader aspects of how this pottery operated within the local community and regional markets

    A Novel Monoallelic Nonsense Mutation in the NFKB2 Gene Does Not Cause a Clinical Manifestation

    NF-κB signaling, acting through NFKB1 dependent canonical and NFKB2 dependent non-canonical pathways plays a critical role in inflammatory and immune responses. Recent studies have associated mutations in these two genes with a common variable immunodeficiency (CVID). While evaluating a female patient seeking a diagnosis explaining her recurrent infections, we found a novel heterozygous c.1831C > T (p.Arg611∗) nonsense mutation in the NFKB2 gene which introduces a Stop codon in the ankyrin repeat domain of p100. Whole exome sequencing (WES) analysis, followed by Sanger sequencing, identified this previously unknown mutation in two other family members. Penetrance of the c.1831C > T variant was assessed by flow-cytometry and protein expression in peripheral blood mononuclear cells (PBMC); whereas, activation of the NF-κB2 signaling pathway was examined through immunoblotting and real-time PCR. Heterozygous c.1831C > T variant led to the expansion of lymphocyte B subpopulations with concomitant reduction of plasmablasts, low IgG levels, and accumulation of p52 in PBMC. On the other hand, tested subjects had normal levels of IgM, IgA, IgE and no impairment in lymphocytes proliferation. Although evaluated patients did not fulfill all clinical features of CVID, their health should be monitored in the future for possible late manifestation of the disease. In conclusion, we showed that NFKB2 haplodeficiency caused by c.1831C > T nonsense mutation is asymptomatic, possibly due to the compensatory mechanisms and allele redundancy

    Targeted Genomic Sequencing of TSC1 and TSC2 Reveals Causal Variants in Individuals for Whom Previous Genetic Testing for Tuberous Sclerosis Complex Was Normal

    Tuberous sclerosis complex (TSC) is caused by inactivating variants in TSC1 and TSC2. Somatic mosaicism, as well as the size and complexity of the TSC1 and TSC2 loci, makes variant identification challenging. Indeed, in some individuals with a clinical diagnosis of TSC, diagnostic testing fails to identify an inactivating variant. To improve TSC1 and TSC2 variant detection, we screened the TSC1 and TSC2 genomic regions using targeted HaloPlex custom capture and next-generation sequencing (NGS) in genomic DNA isolated from peripheral blood of individuals with definite, possible or suspected TSC in whom no disease-associated variant had been identified by previous diagnostic genetic testing. We obtained &gt;95% target region coverage at a read depth of 20 and &gt;50% coverage at a read depth of 300 and identified inactivating TSC1 or TSC2 variants in 83/155 individuals (54%); 65/113 (58%) with clinically definite TSC and 18/42 (43%) with possible or suspected TSC. These included 19 individuals with deep intronic variants and 54 likely cases of mosaicism (variant allele frequency 1-28%; median 7%). In 13 cases (8%), we identified a variant of uncertain significance (VUS). Targeted genomic NGS of TSC1 and TSC2 increases the yield of inactivating variants found in individuals with suspected TSC.</p

    This paper examines the cultural landscape of Peel town, a transient camp occupied for a short time in 1829–1830 during the Swan River settlement’s initial colonisation phase. The camp comprised indentured free colonists from Britain under the leadership of Thomas Peel. The research described shows that the camp’s layout altered over time as disease, low morale and the addition of further colonists from Britain into the camp area eroded the predetermined ideas of class segregation and the control of resources, seriously affecting the leadership group’s ability to maintain authority

    NASTRAN thermal analyzer in a unified finite-element treatment of thermo-structural analyses

    The NASTRAN thermal analyzer (NTA) which performs large-scale unified thermo-structural analyses with the NASTRAN (NASA structural analysis) computer program is described. The mathematical similitude between these two distinct disciplines of thermal and structure is examined. It serves as the theoretical basis upon which the implementation of the thermal capability in NASTRAN was accomplished. The program structure, the functional flow, the solution algorithms, the organization of an input data deck and the solution capabilities of NTA are summarized. Emphasis is placed on the interface of the unified approach in thermo-structural analyses where stresses, deflections, vibrations and bucklings induced by the effect of temperature change are of concern. Attentions are also directed to the preprocessor and post processors. As a specially designed preprocessor, the VIEW program is capable of generating exchange factors which can be output, at user's option, in formats compatible with that required by NTA. Two post processors that serve specific objectives are included. They are the thermal variance analysis and the graphical displaying capability of temperatures in color or black and white

    O objetivo deste trabalho foi averiguar o comportamento de cultivares de girassol, de ensaio de final de segundo ano, quando avaliados em diversas áreas do Nordeste brasileiro, para fins de recomendação. Os ensaios foram realizados nos estados de Sergipe, Alagoas e Rio Grande do Norte. Utilizou-se o delineamento experimental em blocos ao acaso, com quatro repetições de dezoito tratamentos. Constataram-se, na análise de variância conjunta, diferenças significativas para os efeitos de genótipos, ambientes e interação genótipos x ambientes, evidenciando diferenças entre os genótipos e os ambientes e comportamento não consistente destes genótipos nos ambientes avaliados. Os genótipos com rendimentos médios de grãos acima da média geral mostraram melhor adaptação, destacando-se, entre eles, os EXP 1452 CL, BRS G6, M 734, BRS G26, V20041 e NEON, os quais se constituem em ótimas alternativas para exploração comercial na região
